ABSTRACT: BACKGROUND
A growing body of evidence indicates that epigenetic silencing of genes involved in antigen processing and immune recognition results in immune escape and resistance to immunotherapy. Pre-clinical experiments have shown that use of histone deacetylases inhibitors such as vorinostat can restore tumor immune recognition and synergize with anti-PD 1/PD-L1 antibodies. Moreover, vorinostat has radiosensitizing properties. This report describes an ongoing phase I trial of vorinostat in combination with an antibody against PD1 (pembrolizumab), radiotherapy and temozolomide (TMZ) in patients (pts) with newly diagnosed glioblastoma (GBM). METHOD: This study employs a standard 3 + 3 dose escalation design exploring 2 sequential dose escalation cohorts of vorinostat. Elig
