A Six-Crossing Doubly Interlocked [2]Catenane with Twisted Rings, and a Molecular Granny Knot.

ABSTRACT: A molecular 623 link (a six crossing, doubly interlocked, [2]catenane with twisted rings) and a 3₁ #3₁ granny knot (a composite knot made up of two trefoil tangles of the same handedness) were constructed by ring-closing olefin metathesis of an iron(II)-coordinated 2×2 interwoven grid. The connections were directed by pendant phenyl groups to be between proximal ligand ends on the same faces of the grid. The 623 link was separated from the topoisomeric granny knot by recycling size-exclusion chromatography. The identity of each topoisomer was determined by tandem mass spectrometry and the structure of the 623 link confirmed by X-ray crystallography, which revealed two 82-membered macrocycles, each in figure-of-eight conformations, linked through both pairs of loops.

Transmembrane Ion Channels Formed by a Star of David [2]Catenane and a Molecular Pentafoil Knot.

Project description:A (FeII)6-coordinated triply interlocked ("Star of David") [2]catenane (612 link) and a (FeII)5-coordinated pentafoil (51) knot are found to selectively transport anions across phospholipid bilayers. Allostery, topology, and building block stoichiometry all play important roles in the efficacy of the ionophoric activity. Multiple FeII cation coordination by the interlocked molecules is crucial: the demetalated catenane exhibits no anion binding in solution nor any transmembrane ion transport properties. However, the topologically trivial, Lehn-type cyclic hexameric FeII helicates-which have similar anion binding affinities to the metalated Star of David catenane in solution-also display no ion transport properties. The unanticipated difference in behavior between the open- and closed-loop structures may arise from conformational restrictions in the linking groups that likely enhances the rigidity of the channel-forming topologically complex molecules. The (FeII)6-coordinated Star of David catenane, derived from a hexameric cyclic helicate, is 2 orders of magnitude more potent in terms of ion transport than the (FeII)5-coordinated pentafoil knot, derived from a cyclic pentamer of the same building block. The reduced efficacy is reminiscent of multisubunit protein ion channels assembled with incorrect monomer stoichiometries.

A unique non-catenane interlocked self-assembled supramolecular architecture and its photophysical properties.

Project description:A novel, interlocked, self-assembled (M(2)L(2))(2) molecular architecture was constructed from an arene-Ru acceptor and a 1,4-di(pyridin-4-yl)buta-1,3-diyne donor. Two M(2)L(2) units, with cavities of ~7.21 Å, spontaneously interlock, with one unit encapsulating a twin in a non-catenane fashion. The dimeric host-guest complex thus formed is unique among two-dimensional self-assemblies and is stabilized by π-π interactions between the M(2)L(2) units.

An S10-Symmetric 5-Fold Interlocked [2]Catenane.

Project description:The reaction of sym-pentakis(4-aminothiophenyl)corannulene with 2-formyl-6-methylpyridine and CuI or 2-formyl-1,10-phenanthroline and MII (M = Co, Zn) yields an S10-symmetric 5-fold interlocked [2]catenane of two interpenetrating [CuI5L2]5+ cages or D5-symmetric [MII5L2]10+ cages, respectively. The new structures were characterized by X-ray crystallography, NMR spectroscopy, and mass spectrometry. Density functional theory computations point to dispersive energies on par with traditional covalent bond energies. Subcomponent exchange reactions favored formation of the [CoII5L2]10+ cage over the [CuI10L4]10+ catenane. The single cage and catenane each cocrystallized with a corannulene guest to form a bowl-in-bowl substructure.

Enhanced intersystem crossing in core-twisted aromatics.

Project description:We describe the design, bottom-up synthesis and X-ray single crystal structure of systematically twisted aromatics 1c and 2d for efficient intersystem crossing. Steric congestion at the cove region creates a nonplanar geometry that induces a significant yield of triplet excited states in the electron-poor core-twisted aromatics 1c and 2d. A systematic increase in the number of twisted regions in 1c and 2d results in a concomitant enhancement in the rate and yield of intersystem crossing, monitored using femtosecond and nanosecond transient absorption spectroscopy. Time-resolved absorption spectroscopic measurements display enhanced triplet quantum yields (Φ T = 10 ± 1% for 1c and Φ T = 30 ± 2% for 2d) in the twisted aromatics when compared to a negligible Φ T (<1%) in the planar analog 3c. Twist-induced spin-orbit coupling via activated out-of-plane C-H/C[double bond, length as m-dash]C vibrations can facilitate the formation of triplet excited states in twisted aromatics 1c and 2d, in contrast to the negligible intersystem crossing in the planar analog 3c. The ease of synthesis, high solubility, access to triplet excited states and strong electron affinity make such imide functionalized core-twisted aromatics desirable materials for organic electronics such as solar cells.

A Doubly Bridged Bis(phenylethynyl)benzene: Different from a Twisted Tolan.

The target displays photophysical properties, distinctly different from that of its congeners, the singly bridged tolans. Quantum-chemical calculations suggest a lack of planarization of the bridged bis(phenylethynyl)benzene in the first excited state.

Stereoselective Synthesis of Molecular Square and Granny Knots.

Project description:We report on the stereoselective synthesis of both molecular granny and square knots through the use of lanthanide-complexed overhand knots of specific handedness as three-crossing "entanglement synthons". The composite knots are assembled by combining two entanglement synthons (of the same chirality for a granny knot; of opposite handedness for a square knot) in three synthetic steps: first, a CuAAC reaction joins together one end of each overhand knot. Ring-closing olefin metathesis (RCM) then affords the closed-loop knot, locking the topology. This allows the lanthanide ions necessary for stabilizing the entangled conformation of the synthons to subsequently be removed. The composite knots were characterized by 1H and 13C NMR spectroscopy and mass spectrometry and the chirality of the knot stereoisomers compared by circular dichroism. The synthetic strategy of combining building blocks of defined stereochemistry (here overhand knots of Λ- or Δ-handed entanglement) is reminiscent of the chiron approach of using minimalist chiral synthons in the stereoselective synthesis of molecules with multiple asymmetric centers.

A multiplexed circulating tumor DNA detection platform engineered from 3D-coded interlocked DNA rings.

Project description:Circulating tumor DNA (ctDNA) is a critical biomarker not only important for the early detection of tumors but also invaluable for personalized treatments. Currently ctDNA detection relies on sequencing. Here, a platform termed three-dimensional-coded interlocked DNA rings (3D-coded ID rings) was created for multiplexed ctDNA identification. The ID rings provide a ctDNA recognition ring that is physically interlocked with a reporter ring. The specific binding of ctDNA to the recognition ring initiates target-responsive cutting via a restriction endonuclease; the cutting then triggers rolling circle amplification on the reporter ring. The signals are further integrated with internal 3D codes for multiplexed readouts. ctDNAs from non-invasive clinical specimens including plasma, feces, and urine were detected and validated at a sensitivity much higher than those obtained through sequencing. This 3D-coded ID ring platform can detect any multiple DNA fragments simultaneously without sequencing. We envision that our platform will facilitate the implementation of future personalized/precision medicine.

Coupling across a DNA helical turn yields a hybrid DNA/organic catenane doubly tailed with functional termini.

Project description:We describe the synthesis of a hybrid DNA/organic macrocycle that is prepared by formation of an amide linkage across one full turn of DNA. Formation of a catenane proved that the linkage crossed a turn rather than running along the phosphodiester backbone contour. The product, a doubly tailed catenane, contains 5'- and 3'-termini that can be functionalized further or used to incorporate the catenane structure into other DNA assemblies.

The 1α-hydroxy-A-rings of norditerpenoid alkaloids are twisted-boat conformers.

Project description:The skeletal conformations of naturally occurring norditerpenoid alkaloids fix their substituent functional groups in space, thereby directing their bioactivities. Solution conformations of the A-rings of 4 selected norditerpenoid alkaloid free bases: mesaconitine, karacoline (karakoline), condelphine, and neoline (bullatine B), were analysed by NMR spectroscopy and single-crystal X-ray crystallography. They adopt twisted-chair, twisted-boat, twisted-boat, twisted-boat conformations, respectively. That the A-ring is stabilised in a boat conformer by an intramolecular H-bond from 1α-OH to the N-ethyl tertiary amine is also confirmed in the condelphine single crystal data. The conformations are a result of through-space repulsion between 12-He' and atoms attached to C1 (in the equatorial positions). This causes the A-rings with 1α-OR always to be twisted whether in a chair or a boat conformation. The impact of these studies is in providing a detailed understanding of the shape of the A-ring of these important biologically active natural product alkaloids.

Mechanically Interlocked Molecular Rotors on Pb(100).

Project description:The mechanical coupling between molecules represents a promising route for the development of molecular machines. Constructing molecular gears requires easily rotatable and mutually interlocked pinions. Using scanning tunneling microscopy (STM), it is demonstrated that aluminum phthalocyanine (AlPc) molecules on Pb(100) exhibit these properties. Unlike other phthalocyanines on this substrate, isolated AlPc molecules fluctuate between two azimuthal orientations. Density functional theory (DFT) calculations confirm two stable orientations of single molecules and indicate a relatively low rotation barrier. In STM-constructed dimers and trimers, fluctuations diminish, and various molecular orientations are stabilized. Induced collective rotation of all molecules in the trimers is observed, demonstrating their mechanical interlocking. Potential functions describing angle and distance dependencies of intermolecular and molecule-substrate interactions are derived from DFT calculations of dimers; 52 experimentally determined trimer geometries are reproduced using these potentials. This intuitive approach may prove to be useful in modeling larger structures beyond the scope of quantum mechanical descriptions.
