Use of qPCR-Based Cercariometry to Assess Swimmer's Itch in Recreational Lakes.

ABSTRACT: Swimmer's itch (cercarial dermatitis) is a nuisance encountered by bathers and recreational water users worldwide. The condition is caused by the penetration of larval digenean trematodes (cercariae) of the family Schistosomatidae, into the skin, following their release into freshwater from pulmonate snails that serve as the intermediate hosts for these parasites. This study utilizes qPCR-based cercariometry to monitor and quantify cercariae from water samples collected at 5 lakes in northern Michigan. The resolution provided by qPCR facilitated assessment of the environmental and biological drivers of swimmer's itch-causing cercariae concentrations, allowing us to demonstrate that cercarial abundance is greatest at the top of the water column, in locations with prevailing on- and alongshore winds.

Project description:Swimmer's itch (SI) is a dermatitis in humans caused by cercariae of avian and mammalian schistosomes which emerge from infected snails on a daily basis. Mitigation methods for SI have long been sought with little success. Copper sulfate application to the water to kill the snail hosts is the historically employed method, but is localized, temporary, and harmful to many aquatic species. Here, we test an alternative method to control Trichobilharzia stagnicolae, a species well-known to cause SI in northern Michigan and elsewhere in North America. Summer relocation of broods of the only known vertebrate host, common merganser (Mergus merganser), greatly reduced snail infection prevalence the following year on two large, geographically separated lakes in northern Michigan. Subsequent years of host relocation achieved and maintained snail infection prevalence at ~0.05%, more than an order of magnitude lower than pre-intervention. A Before-After-Control-Intervention (BACI) study design using multiple-year snail infection data from two intervention lakes and three control lakes demonstrates that dramatic lake-wide reduction of an avian schistosome can be achieved and is not due to natural fluctuations in the parasite populations. The relevance of reducing snail infection prevalence is demonstrated by a large seven-year data set of SI incidence in swimmers at a high-use beach, which showed a substantial reduction in SI cases in two successive years after relocation began. In addition, data from another Michigan lake where vertebrate-host based intervention occurred in the 1980's are analyzed statistically and show a remarkably similar pattern of reduction in snail infection prevalence. Together, these results demonstrate a highly effective SI mitigation strategy that avoids the use of environmentally suspect chemicals and removes incentive for lethal host removal. Biologically, the results strongly suggest that T. stagnicolae is reliant on the yearly hatch of ducklings to maintain populations at high levels on a lake and that the role of migratory hosts in the spring and fall is much less significant.

Project description:BackgroundCercarial dermatitis, colloquially "swimmer's itch", is a rash contracted in natural bodies of water, when people are exposed to skin-penetrating, larval flatworm parasites of the family Schistosomatidae, that emerge from aquatic snails. Swimmer's itch is a globally-distributed, allergic condition, of which we know very little regarding local dynamics of transmission. This study aims to gather relevant information on swimmer's itch in Canada, from multiple perspectives, including the human experience, parasite and host presence and distributions, and insight from historical perspectives.MethodsHerein we utilize a mixed-methods approach towards examining the environmental health issue of swimmer's itch in Canadian lakes from a nation-wide viewpoint, with an example from Alberta. We examine the human perspective of having contracted swimmer's itch through a self-reporting surveillance system implemented over a 5-year period. We also conducted a 3-year species survey of parasites and intermediate snail hosts within lakes in central Alberta and compiled this data with snail and vertebrate (definitive) host survey data from across Alberta to examine potential for future spread. We compare the results from our surveys to a historical review of the literature to examine the extent of swimmer's itch across Canada and identify where future efforts should be focused.ResultsOver 3800 cases of swimmer's itch were captured across Canada by the self-reporting surveillance system. Swimmer's itch cases were reported from every province except Prince Edward Island. Species surveys in Alberta revealed 7 new parasite and host records, with potential for swimmer's itch to occur throughout most of the province based on host distributions. A review and comparison to the literature has highlighted several knowledge gaps surrounding schistosome species, host species and their distributions and contributions towards swimmer's itch.ConclusionsSwimmer's itch is a greater environmental health hazard across Canada than previous literature would have alluded. This study provides proof-of-concept for the utility of a self-reporting surveillance system for swimmer's itch in Canada. Recommendations are made towards implementing a systems-thinking approach that incorporates citizen-scientists for future research, management, and policy surrounding swimmer's itch.

Project description:Long-distance dispersal pathways, which frequently relate to human activities, facilitate the spread of alien species. One pathway of concern in North America is the possible spread of forest pests in firewood carried by visitors to campgrounds or recreational facilities. We present a network model depicting the movement of campers and, by extension, potentially infested firewood. We constructed the model from US National Recreation Reservation Service data documenting more than seven million visitor reservations (including visitors from Canada) at campgrounds nationwide. This bi-directional model can be used to identify likely origin and destination locations for a camper-transported pest. To support broad-scale decision making, we used the model to generate summary maps for 48 US states and seven Canadian provinces that depict the most likely origins of campers traveling from outside the target state or province. The maps generally showed one of two basic spatial patterns of out-of-state (or out-of-province) origin risk. In the eastern United States, the riskiest out-of-state origin locations were usually found in a localized region restricted to portions of adjacent states. In the western United States, the riskiest out-of-state origin locations were typically associated with major urban areas located far from the state of interest. A few states and the Canadian provinces showed characteristics of both patterns. These model outputs can guide deployment of resources for surveillance, firewood inspections, or other activities. Significantly, the contrasting map patterns indicate that no single response strategy is appropriate for all states and provinces. If most out-of-state campers are traveling from distant areas, it may be effective to deploy resources at key points along major roads (e.g., interstate highways), since these locations could effectively represent bottlenecks of camper movement. If most campers are from nearby areas, they may have many feasible travel routes, so a more widely distributed deployment may be necessary.
