Cytotoxic Labdane Diterpenes from Hedychium ellipticum Buch.-Ham. ex Sm.

ABSTRACT: In order to reveal the constituents and their biological activities, we carried out a phytochemical study on Hedychium ellipticum Buch.-Ham. ex Sm. (Zingiberaceae). Ten labdane diterpenoids (1-10) were isolated from the rhizomes of H. ellipticum for the first time. Their structures were identified on the basis of spectroscopic analyses including two-dimensional NMR and comparison with literature data. All of these compounds were evaluated for their antimycobacterial activity against Mycobacterium tuberculosis and cytotoxicity against KB, MCF7, NCI-H187 and Vero cells. The result showed that compounds 1 and 7 exhibited moderate activity against Mycobacterium tuberculosis and compounds 4, 6 and 7 displayed remarkable cytotoxic activity. This is the first report on the presence of all compounds in H. ellipticum and the first time that their structure activity relationship has been discussed.

Project description:Over the years, labdane diterpenes, norlabdane diterpenes, and bis-labdanic diterpenes with cytotoxic activities have been identified across various families in the plant kingdom including the Zingiberaceae. The present review discusses the distribution of these labdane-type diterpenes within the Zingiberaceae; their extraction, isolation, and characterization from the respective Zingiberaceae species; the structural similarities and differences within each group and between the different groups of the labdane-type diterpenes; and their cytotoxic activities against breast, cervical, liver, colorectal, pancreatic, lung and prostate cancer cell lines. The review will also provide insight into how the cytotoxic activities of the labdane-type diterpenes are influenced by their structural features.

New Cytotoxic Seco-Type Triterpene and Labdane-Type Diterpenes from Nuxia oppositifolia.

Project description:Chromatographic purification of the n-hexane and dichloromethane extracts of Nuxia oppositifolia aerial parts, growing in Saudi Arabia, resulted in the isolation and characterization of three new labdane-type diterpene acids, 2β-acetoxy-labda-7-en-15-oic acid (1), 2β-acetoxy-7-oxolabda-8-en-15-oic acid (2), 2β-acetoxy-6-oxolabda-7-en-15-oic acid (3), and one new seco-triterpene, 3,4-seco olean-12-en-3,30 dioic acid (4), together with 10 known lupane, oleanane and ursane-type triterpenes, as well as the common phytosterols, β-sitosterol and stigmasterol (5-16). Their structures have been assigned on the basis of different spectroscopic techniques including 1D and 2D NMR. Moreover, 13 of the isolated compounds were tested on the human cancer cell lines HeLa (cervical), A549 (lung) and MDA (breast), and most of the compounds showed potent cytotoxic activities in vitro.

Dimeric labdane diterpenes: synthesis and antiproliferative effects.

Project description:Several diterpenes with the labdane skeleton show biological activity, including antiproliferative effects. Most of the research work on bioactive labdanes has been carried out on naturally occurring diterpenes and semisynthetic derivatives, but much less is known on the effects of diterpene dimers. The aim of the present work was to synthesize dimeric diterpenes from the labdane imbricatolic acid using esters, ethers and the triazole ring as linkers. Some 18 new derivatives were prepared and the compounds were evaluated for antiproliferative activity on human normal fibroblasts (MRC-5) and the following human tumor cell lines: AGS, SK-MES-1, J82 and HL-60. The diethers 8-10, differing in the number of CH₂ units in the linker, presented better antiproliferative activity with a maximum effect for the derivative 9. The best antiproliferative effect against HL-60 cells was found for compounds 3 and 17, with IC₅₀ values of 22.3 and 23.2 μM, lower than that found for the reference compound etoposide (2.23 μM). The compounds 9, 17 and 11 were the most active derivatives towards AGS cells with IC₅₀ values of 17.8, 23.4 and 26.1 μM. A free carboxylic acid function seems relevant for the effect as several of the compounds showed less antiproliferative effect after methylation.

Labdane Diterpenes from the Fruits of Sinopodophyllum emodi.

Project description:Two new labdane diterpenes, sinoditerpene A (1) and B (2), were isolated from the fruits of Sinopodophyllum emodi, along with two known analogues 3 and 4. Their structures were established on the basis of extensive spectroscopic analysis. The isolation of compounds 1-4 represents the first report of diterpenes from the genus Sinopodophyllum. The cytotoxic activities of all isolated compounds were evaluated in comparison with 5-fluorouracil against the MCF-7 and HepG2 cell lines, towards which 3 showed more potent cytotoxicity.

Project description:Labdanes, leonurenones A-C, two known labdanes, luteolin 7-O-β-glucoside and luteolin were isolated and characterized from a commercial source of Leonotis leonurus. Genetic methods allowed for identification of the plant material. The leonurenones contain an uncommon α,β-unsaturated enone moiety in ring B, and leonurenones A and B were evaluated in a competitive inhibition assay at the GABA A neuroreceptor site.

Characterization and phylogenetic analysis of the chloroplast genome of Duhaldea cappa (Buch.-Ham. ex D.Don) Pruski & Anderb. (Asteraceae).

Project description:Duhaldea cappa, a valuable medicinal plant of genus Duhaldea in the tribe Inuleae, is predominantly found in China, Bhutan, India, Malaysia, Nepal, Pakistan, Thailand, and Vietnam. However, the genomic studies of Duhaldea cappa are limited. In this study, we successfully sequenced and assembled the complete chloroplast genome of Duhaldea cappa. The chloroplast genome is 150,819 bp in length with a 37.73% GC content. The chloroplast genome has a quadripartite structure, consisting of a large single-copy region of 82,731 bp, a small single-copy region of 18,168 bp, and a pair of inverted repeat sequences of 24,960 bp. The genome contains 133 genes. Among these genes, there are 88 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. The phylogeny reconstructed from data of the complete chloroplast genome indicated that Duhaldea cappa is closely related to Pluchea indica in the tribe Inuleae. Analyzing and reporting the chloroplast genome of Duhaldea cappa will establish a solid theoretical and data foundation for the efficient development, conservation, and utilization of this plant species.

The complete chloroplast genome characteristics of Polygala crotalarioides Buch.-Ham. ex DC. (Polygalaceae) from Yunnan, China.

Project description:Polygala crotalarioides Buch.-Ham. ex DC. (Polygalaceae) is a perennial herbaceous plant widely distributed in southwest China. Its chloroplast genome sequence was obtained using Illumina NovaSeq sequencing technology, and then was assembled, annotated, and characterized. The total length of chloroplast genome is 164,269 bp and it contained a large (LCS, 82,697 bp) and a small (SSC, 8083 bp) single copy region, separated by a pair of inverted repeats (IRA/IRB, 36,744 bp). The overall GC content of genome was 36.8% and corresponding values of the LSC, SSC, and IR regions are 34.9%, 29.5%, and 39.7%, respectively. There are 135 genes that were predicted including 89 protein-coding, 38 tRNA, and 8 rRNA genes. The maximum-likelihood (ML) phylogenetic tree reconstructed by IQ-TREE indicated that P. crotalarioides had a strong genetic relationship with P. tenuifolia, P. sibirica, and P. japonica. In summary, the complete chloroplast genome of P. crotalarioides is reported for the first time, which is of great significance for future research on the evolutionary phylogeny and protection of Polygala.

Horizontal gene transfer from a flowering plant to the insular pine Pinus canariensis (Chr. Sm. Ex DC in Buch).

Project description:Horizontal gene transfer (HGT) is viewed as very common in the plant mitochondrial (mt) genome, but, to date, only one case of HGT has been found in gymnosperms. Here we report a new case of HGT, in which a mt nad5-1 fragment was transferred from an angiosperm to Pinus canariensis. Quantitative assay and sequence analyses showed that the foreign nad5-1 is located in the mt genome of P. canariensis and is nonfunctional. An extensive survey in the genus Pinus revealed that the angiosperm-derived nad5-1 is restricted to P. canariensis and present across the species' range. Molecular dating based on chloroplast DNA suggested that the HGT event occurred in the late Miocene after P. canariensis split from its closest relatives, and that the foreign copy became fixed in P. canariensis owing to drift during its colonization of the Canary Islands. The mechanism of this HGT is unclear but it was probably achieved through either direct cell-cell contact or external vectors. Our discovery provides evidence for an important role of HGT in plant mt genome evolution.

Project description:Senecio scandens Buch.-Ham. ex D. Don, an important antibacterial source of Chinese traditional medicine, has a widespread distribution in a few ecological habitats of China. We generated a full-length complementary DNA (cDNA) library from a sample of elite individuals with superior antibacterial properties, with satisfactory parameters such as library storage (4.30 × 106 CFU), efficiency of titre (1.30 × 106 CFU/mL), transformation efficiency (96.35%), full-length ratio (64.00%) and redundancy ratio (3.28%). The BLASTN search revealed the facile formation of counterparts between the experimental sample and Arabidopsis thaliana in view of high-homology cDNA sequence (90.79%) with e-values <1e - 50. Sequence similarities to known proteins indicate that the entire sequences of the full-length cDNA clones consist of the major of functional genes identified by a large set of microarray data from the present experimental material. For other Compositae species, a large set of full-length cDNA clones reported in the present article will serve as a useful resource to facilitate further research on the transferability of expressed sequence tag-derived simple sequence repeats (EST-SSR) development, comparative genomics and novel transcript profiles.
