Photothermal-Triggered Shape Memory Polymer Prepared by Cross-Linking Porphyrin-Loaded Micellar Particles.

ABSTRACT: In this work, we fabricated porphyrin-loaded shape memory polymer (SMP) film by cross-linking micellar particles prepared by co-assembly of porphyrin compounds and amphiphilic macromolecules formulated by copolymerization of 2-butoxy ethanol (BCS), methyl methacrylate (MMA), butyl acrylate (BA) acrylic acid (AA), and diacetone acrylamide (DAAM). The experimental results revealed that this film was able to respond to the red light in terms of photothermal effect enabled by the porphyrin filler. The photothermal-triggered shape memory behaviors of the film were further examined in detail. It was noteworthy that this material was expected to have potential applications in the biomedical field due to the excellent biocompatibility of the porphyrin filler and the red-light source, which was optimal and safe enough for biomedical treatment.

Shape induced inhibition of phagocytosis of polymer particles.

Project description:PurposeTo determine if particle shape can be engineered to inhibit phagocytosis of drug delivery particles by macrophages, which can be a significant barrier to successful therapeutic delivery.MethodsNon-spherical polystyrene particles were fabricated by stretching spherical particles embedded in a polymer film. A rat alveolar macrophage cell line was used as model macrophages. Phagocytosis of particles was assessed using time-lapse video microscopy and fluorescence microscopy.ResultsWe fabricated worm-like particles with very high aspect ratios (>20). This shape exhibits negligible phagocytosis compared to conventional spherical particles of equal volume. Reduced phagocytosis is a result of decreasing high curvature regions of the particle to two single points, the ends of the worm-like particles. Internalization is possible only at these points, while attachment anywhere along the length of the particles inhibits internalization due to the low curvature.ConclusionsShape-induced inhibition of phagocytosis of drug delivery particles is possible by minimizing the size-normalized curvature of particles. We have created a high aspect ratio shape that exhibits negligible uptake by macrophages.

A facile synthesis of dynamic, shape-changing polymer particles.

Project description:We herein report a new facile strategy to ellipsoidal block copolymer nanoparticles that exhibit a pH-triggered anistropic swelling profile. In a first step, elongated particles with an axially stacked lamellae structure are selectively prepared by utilizing functional surfactants to control the phase separation of symmetric polystyrene-b-poly(2-vinylpyridine) (PS-b-P2VP) in dispersed droplets. In a second step, the dynamic shape change is realized by cross-linking the P2VP domains, thereby connecting glassy PS discs with pH-sensitive hydrogel actuators.

Multifunctional shape and size specific magneto-polymer composite particles.

Project description:Interest in uniform multifunctional magnetic particles is driven by potential applications in biomedical and materials science. Here we demonstrate the fabrication of highly tailored nanoscale and microscale magneto-polymer composite particles using a template based approach. Regiospecific surface functionalization of the particles was performed by chemical grafting and evaporative Pt deposition. Manipulation of the particles by an applied magnetic field was demonstrated in water and hydrogen peroxide.

Photothermal cancer therapy using graphitic carbon-coated magnetic particles prepared by one-pot synthesis.

Project description:We describe here a simple synthetic strategy for the fabrication of carbon-coated Fe3O4 (Fe3O4@C) particles using a single-component precursor, iron (III) diethylenetriaminepentaacetic acid complex. Physicochemical analyses revealed that the core of the synthesized particles consists of ferromagnetic Fe3O4 material ranging several hundred nanometers, embedded in nitrogen-doped graphitic carbon with a thickness of ~120 nm. Because of their photothermal activity (absorption of near-infrared [NIR] light), the Fe3O4@C particles have been investigated for photothermal therapeutic applications. An example of one such application would be the use of Fe3O4@C particles in human adenocarcinoma A549 cells by means of NIR-triggered cell death. In this system, the Fe3O4@C can rapidly generate heat, causing >98% cell death within 10 minutes under 808 nm NIR laser irradiation (2.3 W cm(-2)). These Fe3O4@C particles provided a superior photothermal therapeutic effect by intratumoral delivery and NIR irradiation of tumor xenografts. These results demonstrate that one-pot synthesis of carbon-coated magnetic particles could provide promising materials for future clinical applications and encourage further investigation of this simple method.

AIEgen-loaded nanofibrous membrane as photodynamic/photothermal antimicrobial surface for sunlight-triggered bioprotection.

Project description:The outbreak of infectious diseases such as COVID-19 causes an urgent need for abundant personal protective equipment (PPE) which leads to a huge shortage of raw materials. Additionally, the inappropriate disposal and sterilization of PPE may result in a high risk of cross-contamination. Therefore, the exploration of antimicrobial materials possessing both microbe interception and self-decontamination effects to develop reusable and easy-to-sterilize PPE is of great importance. Herein, an aggregation-induced emission (AIE)-active luminogen-loaded nanofibrous membrane (TTVB@NM) sharing sunlight-triggered photodynamic/photothermal anti-pathogen functions are prepared using the electrospinning technique. Thanks to its porous nanostructure, TTVB@NM shows excellent interception effects toward ultrafine particles and pathogenic aerosols. Benefiting from the superior photophysical properties of the AIE-active dopants, TTVB@NM exhibits integrated properties of wide absorption in visible light range, efficient ROS generation, and moderate photothermal conversion performance. A series of antimicrobial evaluations reveal that TTVB@NM could effectively inactivate pathogenic aerosols containing bacteria (inhibition rate: >99%), fungi (~88%), and viruses (>99%) within only 10 min sunlight irradiation. This study represents a new strategy to construct reusable and easy-to-sterilize hybrid materials for potential bioprotective applications.

Isocorrole-Loaded Polymer Nanoparticles for Photothermal Therapy under 980 nm Light Excitation.

Project description:Photothermal therapy (PTT) is a promising treatment option for diseases, including cancer, arthritis, and periodontitis. Typical photothermal agents (PTAs) absorb light in the near-infrared (NIR)-I region of 650-900 nm with a predominant focus around 800 nm, as these wavelengths are minimally absorbed by water and blood in the tissue. Recently, interest has grown in developing nanomaterials that offer more efficient photothermal conversion and that can be excited by light close to or within the NIR-II window of 1000-1700 nm, which offers less absorption by melanin. Herein, we report on the development of 5,5-diphenyl isocorrole (5-DPIC) complexes containing either Zn(II) or Pd(II) (Zn[5-DPIC] and Pd[5-DPIC], respectively) that absorb strongly across the 850-1000 nm window. We also show that poly(lactic-co-glycolic acid) (PLGA) nanoparticles loaded with these designer isocorroles exhibit low toxicity toward triple-negative breast cancer (TNBC) cells in the dark but enable efficient heat production and photothermal cell ablation upon excitation with 980 nm light. These materials represent an exciting new platform for 980 nm activated PTT and demonstrate the potential for designer isocorroles to serve as effective PTAs.

Polydopamine coated shape memory polymer: enabling light triggered shape recovery, light controlled shape reprogramming and surface functionalization.

Project description:Photo-active shape memory polymers (SMPs) are considered as a promising candidate for converting light into mechanical energy. However, most known SMPs are only thermo-responsive. To achieve photo-activity, photo-responsive choromophores or fillers usually have to be incorporated from the very beginning of the material synthesis. Here, we introduce a novel post-synthesis approach to endow normal SMPs with photo-active properties using mussel-inspired surface chemistry. Without changing the original properties, the resultant polydopamine (PDA) coated SMPs show an efficient photo-active performance. The coating can be easily patterned and erased, which allows flexible light-triggered 3-D shape deformation of a planar SMP sheet. Moreover, owing to the high chemical activity, the PDA coating also provides a platform to optimize the surface properties of the photo-responsive SMPs through secondary surface modification.

Polymer particles that switch shape in response to a stimulus.

Project description:Particle engineering for biomedical applications has unfolded the roles of attributes such as size, surface chemistry, and shape for modulating particle interactions with cells. Recently, dynamic manipulation of such key properties has gained attention in view of the need to precisely control particle interaction with cells. With increasing recognition of the pivotal role of particle shape in determining their biomedical applications, we report on polymeric particles that are able to switch their shape in real time in a stimulus-responsive manner. The shape-switching behavior was driven by a subtle balance between polymer viscosity and interfacial tension. The balance between the two forces was modulated by application of an external stimulus chosen from temperature, pH, or chemical additives. The dynamics of shape switch was precisely controlled over minutes to days under physiological conditions. Shape-switching particles exhibited unique interactions with cells. Elliptical disk-shaped particles that are not phagocytosed by macrophages were made to internalize through shape switch, demonstrating the ability of shape-switchable particles in modulating interaction with cells.

Photothermal conversion triggered thermal asymmetric catalysis within metal nanoparticles loaded homochiral covalent organic framework.

Project description:For seeking high enantiopurity, the previously reported thermal asymmetric catalysis is usually carried out at low temperature sometimes with limited yield, that is, the high enantiomeric excess (ee) usually at the cost of high yield. Thus, the achieving both high stereoselectivity and yield is an enormous challenge. We report herein two metal nanoparticle (M NP)-loaded and porphyrin-containing homochiral covalent organic framework (CCOF)-based composite catalysts, and their application in the thermally-driven asymmetric one-pot Henry and A3-coupling reactions. All the reactions are conducted at elevated temperatures with both excellent stereoselectivity and yield which resulted from the synergy of CCOF confinement effect and M NP catalytic activation. Notably, the needed thermal energy for the asymmetric reactions herein is derived from the photothermal conversion via porphyrin-based CCOF upon irradiation with visible light. Remarkably, the CCOF confinement effect can be effectively maintained up to 100 °C for the asymmetric one-pot Henry and A3-coupling reactions herein.

Entanglement-Based Thermoplastic Shape Memory Polymeric Particles with Photothermal Actuation for Biomedical Applications.

Project description:Triggering shape-memory functionality under clinical hyperthermia temperatures could enable the control and actuation of shape-memory systems in clinical practice. For this purpose, we developed light-inducible shape-memory microparticles composed of a poly(d,l-lactic acid) (PDLLA) matrix encapsulating gold nanoparticles (Au@PDLLA hybrid microparticles). This shape-memory polymeric system for the first time demonstrates the capability of maintaining an anisotropic shape at body temperature with triggered shape-memory effect back to a spherical shape at a narrow temperature range above body temperature with a proper shape recovery speed (37 < T < 45 °C). We applied a modified film-stretching processing method with carefully controlled stretching temperature to enable shape memory and anisotropy in these micron-sized particles. Accordingly, we achieved purely entanglement-based shape-memory response without chemical cross-links in the miniaturized shape-memory system. Furthermore, these shape-memory microparticles exhibited light-induced spatiotemporal control of their shape recovery using a laser to trigger the photothermal heating of doped gold nanoparticles. This shape-memory system is composed of biocompatible components and exhibits spatiotemporal controllability of its properties, demonstrating a potential for various biomedical applications, such as tuning macrophage phagocytosis as demonstrated in this study.
