Reusable Xerogel Containing Quantum Dots with High Fluorescence Retention.

ABSTRACT: Although various analytical methods have been established based on quantum dots (QDs), most were conducted in solution, which is inadequate for storage/transportation and rapid analysis. Moreover, the potential environmental problems caused by abandoned QDs cannot be ignored. In this paper, a reusable xerogel containing CdTe with strong emission is established by introducing host⁻guest interactions between QDs and polymer matrix. This xerogel shows high QDs loading capacity without decrease or redshift in fluorescence (the maximum of loading is 50 wt % of the final xerogel), which benefits from the steric hindrance of β-cyclodextrin (βCD) molecules. Host⁻guest interactions immobilize QDs firmly, resulting in the excellent fluorescence retention of the xerogel. The good detecting performance and reusability mean this xerogel could be employed as a versatile analysis platform (for quantitative and qualitative analyses). In addition, the xerogel can be self-healed by the aid of water.

Carbon Quantum Dots for Zebrafish Fluorescence Imaging.

Project description:Carbon quantum dots (C-QDs) are becoming a desirable alternative to metal-based QDs and dye probes owing to their high biocompatibility, low toxicity, ease of preparation, and unique photophysical properties. Herein, we describe fluorescence bioimaging of zebrafish using C-QDs as probe in terms of the preparation of C-QDs, zebrafish husbandry, embryo harvesting, and introduction of C-QDs into embryos and larvae by soaking and microinjection. The multicolor of C-QDs was validated with their imaging for zebrafish embryo. The distribution of C-QDs in zebrafish embryos and larvae were successfully observed from their fluorescence emission. the bio-toxicity of C-QDs was tested with zebrafish as model and C-QDs do not interfere to the development of zebrafish embryo. All of the results confirmed the high biocompatibility and low toxicity of C-QDs as imaging probe. The absorption, distribution, metabolism and excretion route (ADME) of C-QDs in zebrafish was revealed by their distribution. Our work provides the useful information for the researchers interested in studying with zebrafish as a model and the applications of C-QDs. The operations related zebrafish are suitable for the study of the toxicity, adverse effects, transport, and biocompatibility of nanomaterials as well as for drug screening with zebrafish as model.

Polymersomes containing quantum dots for cellular imaging.

Project description:Quantum dots (QDs) are highly fluorescent and stable probes for cellular and molecular imaging. However, poor intracellular delivery, stability, and toxicity of QDs in biological compartments hamper their use in cellular imaging. To overcome these limitations, we developed a simple and effective method to load QDs into polymersomes (Ps) made of poly(dimethylsiloxane)-poly(2-methyloxazoline) (PDMS-PMOXA) diblock copolymers without compromising the characteristics of the QDs. These Ps showed no cellular toxicity and QDs were successfully incorporated into the aqueous compartment of the Ps as confirmed by transmission electron microscopy, fluorescence spectroscopy, and fluorescence correlation spectroscopy. Ps containing QDs showed colloidal stability over a period of 6 weeks if stored in phosphate-buffered saline (PBS) at physiological pH (7.4). Efficient intracellular delivery of Ps containing QDs was achieved in human liver carcinoma cells (HepG2) and was visualized by confocal laser scanning microscopy (CLSM). Ps containing QDs showed a time- and concentration-dependent uptake in HepG2 cells and exhibited better intracellular stability than liposomes. Our results suggest that Ps containing QDs can be used as nanoprobes for cellular imaging.

A visual dual-aptamer logic gate for sensitive discrimination of prion diseases-associated isoform with reusable magnetic microparticles and fluorescence quantum dots.

Project description:Molecular logic gates, which have attracted increasing research interest and are crucial for the development of molecular-scale computers, simplify the results of measurements and detections, leaving the diagnosis of disease either "yes" or "no". Prion diseases are a group of fatal neurodegenerative disorders that happen in human and animals. The main problem with a diagnosis of prion diseases is how to sensitively and selectively discriminate and detection of the minute amount of PrP(Res) in biological samples. Our previous work had demonstrated that dual-aptamer strategy could achieve highly sensitive and selective discrimination and detection of prion protein (cellular prion protein, PrP(C), and the diseases associated isoform, PrP(Res)) in serum and brain. Inspired by the advantages of molecular logic gate, we further conceived a new concept for dual-aptamer logic gate that responds to two chemical input signals (PrP(C) or PrP(Res) and Gdn-HCl) and generates a change in fluorescence intensity as the output signal. It was found that PrP(Res) performs the "OR" logic operation while PrP(C) performs "XOR" logic operation when they get through the gate consisted of aptamer modified reusable magnetic microparticles (MMPs-Apt1) and quantum dots (QDs-Apt2). The dual-aptamer logic gate simplifies the discrimination results of PrP(Res), leaving the detection of PrP(Res) either "yes" or "no". The development of OR logic gate based on dual-aptamer strategy and two chemical input signals (PrP(Res) and Gdn-HCl) is an important step toward the design of prion diseases diagnosis and therapy systems.

High-purity C<sub>3</sub>N quantum dots for enhancing fluorescence detection of metal ions.

Project description:A new type of two-dimensional layered material, namely C3N, has been fabricated by polymerization and recommended to have great potential in various applications such as the development of electronic devices or photo-detectors, due to its enhanced conductivity, electronegativity, and unique optical properties. Actually, most of the present research on C3N is limited in the scope of theoretical calculation, while experimental research is blocked by inefficient synthesis with low purity and homogeneity. Here, we report an optimized efficient synthesis method of high-purity C3N QDs in aqueous solution by polymerization of DAP with combined centrifugation and filtration of products, with the synthesis yield up to 33.1 ± 3.1%. Subsequently, the C3N QDs have been used as novel metal ion probes exhibiting a sensitive fluorescent response to various metal ions including monovalent alkaline metals (Li+, Na+, and K+), divalent alkaline-earth metals (Mg2+, Ca2+, and Sr2+), and multivalent transition metals (Cu2+, Co2+, Ni2+, and Au3+, Fe3+, Cr3+) due to the high electronegativity of the C3N surface. Particularly, the fluorescent quenching response of Al3+, Ga3+, In3+, and Sc3+ ions is significantly different from the fluorescent enhanced response of most other carbon-based QDs, which is promising for enriching the detection methods of these metal ions and beneficial to improve the accuracy of ion recognition.

Incorporation of quantum dots in silk biomaterials for fluorescence imaging.

Project description:Tracking the distribution and degradation of biomaterials after in vivo implantation or injection is important for tissue engineering and drug delivery. Intrinsic and externally labeled fluorescence has been widely used for these purposes. In the present study, 3-mercaptopropionic acid (MPA)-coated CdTe quantum dots (QDs) were incorporated into silk materials via strong interactions between QDs and silk, likely involving the hydrophobic beta-sheet structures in silk. MPA-QDs were pre-mixed with silk solution, followed by ultrasonication to induce silk gelation or by blending with polyvinyl alcohol (PVA) to generate silk microspheres. Silk structural changes and hydrogel/microsphere morphologies were examined by ATR-FTIR and SEM, respectively. The fluorescence of QDs-incorporated silk hydrogels and microspheres remained stable in PBS pH 7.4 for more than 4 days. The amount of QDs released from the materials during the incubation was dependent on loading; no QDs were released when loading was below 0.026 nmol/mg silk. After subcutaneous injection in mice, the fluorescence of QDs-incorporated silk microspheres was quenched within 24 h, similar to that of free QDs. In contrast, the QDs-incorporated silk hydrogels fluoresced for more than 4 days in vivo.

Fluorescence modulation of cadmium sulfide quantum dots by azobenzene photochromic switches.

Project description:We have investigated the attachment of azobenzene photochromic switches on the modified surface of cadmium sulfide (CdS) quantum dots (QDs). The modification of CdS QDs is done by varying the concentration of the capping agent (mercaptoacetic acid) and NH3 in order to control the size of the QDs. The X-ray diffraction studies revealed that the crystallite size of CdS QDs ranged from 6 to 10 nm. The azobenzene photochromic derivatives bis(4-hydroxybenzene-1-azo)4,4'(1,1' diphenylmethane) (I) and 4,4'-diazenyldibenzoic acid (II) were synthesized and attached with surface-modified CdS QDs to make fluorophore-photochrome CdS-(I) and CdS-(II) dyad assemblies. Upon UV irradiation, the photochromic compounds (I) and (II) undergo a reversible trans-cis isomerization. The photo-induced trans-cis transformation helps to transfer photo-excited electrons from the conduction band of the CdS QDs to the lowest unoccupied molecular orbital of cis isomer of photochromic compounds (I) and (II). As a result, the fluorescence of CdS-(I) and CdS-(II) dyads is suppressed approximately five times compared to bare CdS QDs. The fluorescence modulation in such systems could help to design luminescent probes for bioimaging applications.

Resonance Fluorescence of GaAs Quantum Dots with Near-Unity Photon Indistinguishability.

Project description:Photonic quantum technologies call for scalable quantum light sources that can be integrated, while providing the end user with single and entangled photons on demand. One promising candidate is strain free GaAs/AlGaAs quantum dots obtained by aluminum droplet etching. Such quantum dots exhibit ultra low multi-photon probability and an unprecedented degree of photon pair entanglement. However, different to commonly studied InGaAs/GaAs quantum dots obtained by the Stranski-Krastanow mode, photons with a near-unity indistinguishability from these quantum emitters have proven to be elusive so far. Here, we show on-demand generation of near-unity indistinguishable photons from these quantum emitters by exploring pulsed resonance fluorescence. Given the short intrinsic lifetime of excitons and trions confined in the GaAs quantum dots, we show single photon indistinguishability with a raw visibility of [Formula: see text], without the need for Purcell enhancement. Our results represent a milestone in the advance of GaAs quantum dots by demonstrating the final missing property standing in the way of using these emitters as a key component in quantum communication applications, e.g., as quantum light sources for quantum repeater architectures.

Nanobubbles loaded with carbon quantum dots for ultrasonic fluorescence dual detection.

Project description:To increase the efficiency and accuracy of clinical tumor detection, we explored multiple imaging by preparing carbon quantum dot (CQD)-loaded nanobubbles for ultrasonic fluorescence dual detection. In this experiment, we prepared 1,2-dioleoyl3-trimethylammonium-propane chloride (DOTAP) cationic liposomes using the film dispersion method and chose perfluoropentane as the core gas material of the nanobubbles. The nanobubbles were coupled with the negatively charged CQDs through the charge effect to prepare the testing agent for two-way diagnosis with ultrasound contrast and fluorescence detection. The formulation and preparation of the loaded CQD liposome nanobubbles were screened. In vivo experiments showed that nanobubbles can be enriched to the tumor site within 5 min, which enables clearer ultrasound imaging and is conducive to tumor detection. We expect CQD-loaded liposome (Lip-CQD) nanobubbles to become a new ultrasonic contrast agent for clinical applications that can provide a basis for early tumor diagnosis and thus earlier treatment.

Fluorescence dynamics of the biosynthesized CdSe quantum dots in Candida utilis.

Project description:Organisms served as factories of bio-assembly of nanoparticles attracted a lot of attentions due to the safe, economic and environmental-benignity traits, especially the fabrication of the super fluorescence properties quantum dots (QDs). However, information about the developmental dynamics of QDs in living organisms is still lacking. In this work, we synthesized cadmium-selenium (CdSe) QDs in Candida utilis WSH02-08, and then tracked and quantitatively characterized the developmental dynamics (photoactivation, photostable and photobleaching processes) of bio-QDs by translating fluorescence microscopy movies into visual quantitative curve. These findings shed light on the fluorescence properties of the bio-assembled QDs and are expected to accelerate the applications of the synthesized QDs in vivo. It provided a new way to screen bio-QDs and monitor the quality of QDs in vivo.
