Molecular and morphological evidence for the identity of two nominal species of Astegopteryx (Hemiptera, Aphididae, Hormaphidinae).

ABSTRACT: The morphology of many insect species is usually influenced by environmental factors and therefore high phenotypic variation exists even within a species. This causes difficulty and uncertainty in species taxonomy, which can be remedied by using molecular data and integrative taxonomy. Astegopteryxbambusae and A.bambucifoliae are currently regarded as two closely related aphid species with similar bamboo hosts and overlapping distributions in the oriental region. However, in practice it is hard to distinguish between them. By incorporating molecular data from four mitochondrial and nuclear genes as well as morphological information from an extensive collection of live specimens, the present study indicates that A.bambucifoliae is a junior synonym of A.bambusae. The data also indicate that large-scale geographic patterns of population differentiation may exist within this species.

Diversity of bacteria associated with Hormaphidinae aphids (Hemiptera: Aphididae).

Project description:Bacteria are ubiquitous inhabitants of animals. Hormaphidinae is a particular aphid group exhibiting very diverse life history traits. However, the microbiota in this group is poorly known. In the present study, using high-throughput sequencing of bacterial 16S ribosomal RNA gene amplicons, we surveyed the bacterial flora in hormaphidine aphids and explored whether the aphid tribe, host plant and geographical distribution are associated with the distribution of secondary symbionts. The most dominant bacteria detected in hormaphidine species are heritable symbionts. As expected, the primary endosymbiont Buchnera aphidicola is the most abundant symbiont across all species and has cospeciated with its host aphids. Six secondary symbionts were detected in Hormaphidinae. Arsenophonus is widespread in Hormaphidinae species, suggesting the possibility of ancient acquisition of this symbiont. Ordination analyses and statistical tests show that the symbiont composition does not seem to relate to any of the aphid tribes, host plants or geographical distributions, which indicate that horizontal transfers might occur for these symbionts in Hormaphidinae. Correlation analysis exhibits negative interference between Buchnera and coexisting secondary symbionts, while the interactions between different secondary symbionts are complicated. These findings display a comprehensive picture of the microbiota in Hormaphidinae and may be helpful in understanding the symbiont diversity within a group of aphids.

Project description:The genus Aleurodaphis van der Goot is reviewed. One new species Aleurodaphis sinojackiae Qiao & Jiang, sp. n. on Sinojackia xylocarpa from Jiangsu and Zhejiang, China is described. Aleurodaphis sinisalicis Zhang, 1982 is synonymised with Aleurodaphis blumeae van der Goot, 1917. Keys to species, morphological description and features of the new species, host plants, and distribution are provided. The specimens including types are deposited in British Natural History Museum, London (BMNH), Kôgakkan University, Japan and the National Zoological Museum of China, Institute of Zoology, Chinese Academy of Sciences, Beijing, China (NZMC).

The mitochondrial genome of a social aphid, Pseudoregma bambucicola (Hemiptera: Aphididae: Hormaphidinae).

Project description:In this study, the complete mitochondrial genome of the social aphid Pseudoregma bambucicola was assembled and annotated. The genome spans 16,631 bp in length with a high A + T content (85.1%), containing 13 protein-coding genes, 22 tRNA genes, two rRNA genes, one control region locating between srRNA and tRNAIle , and a repeat region located between tRNAGlu and tRNAPhe . All the tRNAs except tRNACys and tRNASer(gct) exhibit the typical clover-leaf like structures. The lengths of lrRNA and srRNA are 1301 bp and 758 bp, respectively. The order of genes between COIII and tRNAGlu exhibits significant difference for P. bambucicola. A phylogenetic analysis for P. bambucicola and 22 other aphids based on 13 protein-coding genes is also presented.

The complete mitochondrial genome of Schizoneuraphis gallarum van der Goot, 1917 (Hemiptera: Aphididae: Hormaphidinae).

Project description:In this study, the complete mitochondrial genome of Schizoneuraphis gallarum was sequenced and annotated. The mitogenome is 14,990 bp in length with an A + T content of 82.6%, including 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes and a control region. This is the shortest aphid mitogenome reported so far. All the protein-coding genes are initiated with ATN codon and terminated with TAA codon except cox1 and nad4 which are terminated with T. All the transfer RNA genes can form the typical clover-leaf secondary structure except trnS (AGN). The maximum-likelihood phylogenetic tree based on mitogenome sequences of 25 aphid species shows that S. gallarum is sister to Hormaphis betulae.

Taxonomic notes on the genus Chaitoregma (Hemiptera, Aphididae, Hormaphidinae), with description of a new species from China.

Project description:A new aphid species, Chaitoregmakirlia sp. nov., from Fujian and Guangdong, China, is described, which feeds on bamboo. The diagnostic morphological characteristics of the new species are described and illustrated. A key to apterous viviparous females of Chaitoregma species is provided. The COI barcode sequence of this new species is also provided. Due to its unique morphological characteristics, the diagnosis of the genus has been revised. Other species within the genus are also reviewed and discussed.

Insights Into the Species-Specific Microbiota of Greenideinae (Hemiptera: Aphididae) With Evidence of Phylosymbiosis.

Project description:Aphids and their symbionts represent an outstanding model for studies of insect-symbiont interactions. The aphid microbiota can be shaped by aphid species, geography and host plants. However, the relative importance of phylogenetic and ecological factors in shaping microbial community structures is not well understood. Using Illumina sequencing of the V3-V4 hypervariable region of the 16S rRNA gene, we characterized the microbial compositions of 215 aphid colonies representing 53 species of the aphid subfamily Greenideinae from different regions and plants in China, Nepal, and Vietnam. The primary endosymbiont Buchnera aphidicola and secondary symbiont Serratia symbiotica dominated the microbiota of Greenideinae. We simultaneously explored the relative contribution of host identity (i.e., aphid genus and aphid species), geography and host plant to the structures of bacterial, symbiont and secondary symbiont communities. Ordination analyses and statistical tests highlighted the strongest impact of aphid species on the microbial flora in Greenideinae. Furthermore, we found a phylosymbiosis pattern in natural Greenideinae populations, in which the aphid phylogeny was positively correlated with microbial community dissimilarities. These findings will advance our knowledge of host-associated microbiota assembly across both host phylogenetic and ecological contexts.

Comparison of two clones of Myzus Persicae (Sulzer) (Hemiptera: Aphididae)

Project description:The aim of this experiment was to compare the transciptome of the peach-potato aphid (Myzus persicae) clone 4106a (a laboratory insecticide-susceptible standard collected from potato in Scotland in 2000) with clone 5191A (an insecticide resistant aphid clone collected from tobacco in Greece in 2007) to identify which genes are over or underexpressed in the resistant phenotype. Two-condition experiment, 4106a vs. 5191a Myzus persicae clones. Biological replicates: 4 pools of RNA extracted from ten 15 day old aphids of each clone. Technical Replicates: Two technical reps incorporating a dye swap. Total replication: eight replicates for each clone.
