An Effective Screening Method and a Reliable Screening Trait for Salt Tolerance of Brassica napus at the Germination Stage.

ABSTRACT: Salinity is a major and complex abiotic stress that inhibits plant growth and reduces crop yield. Given the global increase in soil salinity, there is a need to develop salt-tolerant species. Brassica napus L. is an important oilseed crop with some level of salt tolerance. However, few studies have evaluated its salt tolerance thoroughly or screened for traits that can be reliably evaluated for salt tolerance. Here, we evaluated salt tolerance in 549 B. napus inbred lines with different genetic backgrounds using the membership function value (MFV) of certain traits, including the germination rate, root and shoot length, root and shoot fresh weight, and total fresh weight. According to the evaluation criteria-mean MFV, 50 highly salt-tolerant, 115 salt-tolerant, 71 moderately salt-tolerant, 202 salt-sensitive, and 111 highly salt-sensitive inbred lines were screened at the germination stage. We also developed a mathematical evaluation model and identified that the salt tolerance index of shoot fresh weight is a single trait that reliably represents the salt tolerance of B. napus germplasm at the germination stage. These results are useful for evaluating and breeding salt-tolerant B. napus germplasm.

Project description:Cotton is an important cash crop with a certain salt tolerance, but its germination stage is very susceptible to the damage of salt stress, causing significant yield loss. However, few studies have evaluated the cotton salt tolerance and selected salt tolerance traits at germination stage. Therefore, in this study, 16 cotton samples with geographical representation were randomly selected from 308 cotton germplasms to determine the optimal 200 mmol·L-1 NaCl in cotton germination experiments. On this basis, the salt tolerance of 308 upland cotton varieties and the growth, ion distribution and transport of highly salt-tolerant and non salt-tolerant cotton germplasms were analyzed. The results showed that the 308 germplasms were classified into five classes through cluster analysis, i.e, (1) highly salt-tolerant germplasms (HST, 49), (2) salt-tolerant germplasms (ST, 169), (3) moderately salt-tolerant class (MST,43), (4) lowly salt-tolerant germplasms (LST, 16), and (5) non-salt-tolerant germplasms (NST, 31). By calculating the salt tolerance index (STI) of various cotton germination and growth parameters and principal component analysis, combined with the correlation analysis and linear regression between mean membership function value (MFV) and STI, the key indexes of cotton germination and growth under salt stress, including total fresh weight, shoot fresh weight, and shoot length, were determined. In addition, three salt tolerance evaluation models constructed with different variables (6 variables in Model 1; 3 variables in Model 2; 1 variable in Model 3) found that the total fresh weight was the most reliable trait for the salt tolerance evaluation. In practical application, the variable selection for modelling could be adjusted based on the experimental workload. The comparison of the K+, Na+, and Ca2+ contents between HST and NST found that the higher the salt tolerance of cotton germplasms, the lower the Na+ content in the root system. Besides, the ion ratios and ion selective transport coefficients (ST) was found to be significantly positively correlated with the salt tolerance of cotton. This study will provide a basis for evaluating and breeding salt-tolerant cotton germplasms.

Project description:Abiotic stresses such as drought and salt stresses have a negative effect on the growth and productivity of plants. Improvement of stress tolerance through genetic engineering in plants has been reported in intense studies. Transcription factors play vital roles in plant adaptation to stresses by regulating expression of a great deal of target genes. A family of Arabidopsis basic region leucine zipper (bZIP) transcription factors that can recognize and bind to the abscisic acid (ABA)-responsive elements (ABREs) in promoter is named as ABRE binding factors (ABFs)/ABRE binding proteins (AREBs). They play a key role in the regulation of expression of downstream stress-responsive genes in ABA signalling. Genetic transformation of ABF/ABRE transcription factors has been suggested to be an effective approach for engineering stress-tolerant plants. However, whether the ABF/ABRE transcription factors are able to be used for generating stress-tolerant rapeseed plants has not yet been studied.BnaABF2, encoding a bZIP transcription factor, was cloned from rapeseed in this study. Subcellular localization and transactivation analyses showed that BnaABF2 was localized to the nucleus with transactivation activity in plant cells. BnaABF2 gene expression was induced by drought and salt stresses and BnaABF2 positively functions in ABA signalling during the vegetative stage. Overexpression of BnaABF2 was found to render drought and salt tolerance to Arabidopsis plants. The resistance of the BnaABF2-expressing transgenic plants to drought and salt stresses is due to reduced water-loss rate and expression of stress-responsive genes such as RD29B, RAB18 and KIN2. The expression of RD29B, RAB18 and KIN2 regulated by BnaABF2 is involved in an ABA-dependent stress signalling.Identification of the positive role of rapeseed BnaABF2 in plant tolerance to drought and salt provides evidence for ability of engineering stress-tolerant rapeseed plants by genetic transformation of BnaABF2.
