Separation of spermatozoa from erythrocytes using their tumbling mechanism in a pinch flow fractionation device.

ABSTRACT: Men suffering from azoospermia can father a child, by extracting spermatozoa from a testicular biopsy sample. The main complication in this procedure is the presence of an abundance of erythrocytes. Currently, the isolation of the few spermatozoa from the sample is manually performed due to ineffectiveness of filtering methods, making it time consuming and labor intensive. The spermatozoa are smaller in both width and height than any other cell type found in the sample, with a very small difference compared with the erythrocyte for the smallest, making this not the feature to base the extraction on. However, the length of the spermatozoon is 5× larger than the diameter of an erythrocyte and can be utilized. Here we propose a microfluidic chip, in which the tumbling behavior of spermatozoa in pinched flow fractionation is utilized to separate them from the erythrocytes. We show that we can extract 95% of the spermatozoa from a sample containing 2.5% spermatozoa, while removing around 90% of the erythrocytes. By adjusting the flow rates, we are able to increase the collection efficiency while slightly sacrificing the purity, tuning the solution for the available sample in the clinic.

Project description:Cellulose nanocrystals (CNCs) are renewable, naturally derived polymeric nanomaterials receiving substantial attention for a wide range of potential applications. The recent availability of high quality reference materials will facilitate the development and validation of measurement methods needed to advance the scientific and commercial use of CNCs. In the present study, we demonstrate an optimized method to fractionate CNCs with narrow size dispersion based on asymmetrical-flow field-flow fractionation (AF4) coupled with on-line multi-angle light scattering (MALS), dynamic light scattering (DLS), and differential refractometry (dRI). A stable suspension of CNC (Certified Reference Material CNCD-1, National Research Council-Canada) in deionized water was prepared using a dispersion method provided by NRC and adopted from a protocol originally developed at the National Institute of Standards and Technology. The as-prepared material was initially characterized in batch mode to validate the NRC dispersion method. AF4 was then optimized for channel and cross flow, mobile phase composition, and injection volume, among other parameters. Additionally, suspensions containing (1.25-10) mg mL-1 CNC were injected directly into the dRI detector (off-line), yielding a dn/dc value of 0.148 ± 0.003 mL g-1. dRI was then used as an on-line mass sensitive detector to quantify recovery. Results show that maximum recovery (≈ 99%) was achieved under optimized conditions. The weight-averaged molar mass (Mw) was estimated at roughly 107 Da from a partial Zimm analysis. The optical radius of gyration, Rg, and the hydrodynamic radius, Rh, were measured during elution. The shape factor (Rg/Rh) ranged from 1.5 to 1.9 for the fractionated material, supporting an elongated or rod-like structure. To our knowledge, this is the first time that both the morphology and molar mass of CNCs have been directly measured for the full distribution of species. Finally, we developed and demonstrated a semi-preparatory fractionation method to separate CNCs at the milligram scale for off-line research and analysis.

Project description:Polymeric particles are ideal drug delivery systems due to their cellular uptake-relevant size. Microparticles could be developed for direct injection of drug formulations into a diseased site, such as a tumor, allowing for drug retention and slow drug exposure over time through sustained release mechanisms. Bombyx mori silk fibroin has shown promise as a biocompatible biomaterial both in research and the clinic. Silk has been previously used to make particles using an emulsion-based method with poly(vinyl alcohol) (PVA). In this study, polydimethylsiloxane-based microfluidic devices were designed, fabricated, and characterized to produce silk particles through self-association of silk when exposed to PVA. Three main variables resulted in differences in particle size and size distribution, or polydispersity index (PDI). Utilizing a co-flow microfluidic device decreased the PDI of the silk particles as compared to an emulsion-based method (0.13 versus 0.65, respectively). With a flow-focusing microfluidics device, lowering the silk flow rate from 0.80 to 0.06 mL/h resulted in a decrease in the median particle size from 6.8 to 3.0 μm and the PDI from 0.12 to 0.05, respectively. Lastly, decreasing the silk concentration from 12% to 2% resulted in a decrease in the median particle size from 5.6 to 2.8 μm and the PDI from 0.81 to 0.25, respectively. Binding and release of doxorubicin, a cytotoxic drug commonly used for cancer treatment, with the fabricated silk particles was evaluated. Doxorubicin loading in the silk particles was approximately 41 µg/mg; sustained doxorubicin release occurred over 23 days. When the cytotoxicity of the released doxorubicin was tested on KELLY neuroblastoma cells, significant cell death was observed. To demonstrate the potential for internalization of the silk particles, both KELLY and THP-1-derived macrophages were exposed to fluorescently labelled silk particles for up to 24 h. With the macrophages, internalization of the silk particles was observed. Additionally, THP-1 derived macrophages exposure to silk particles increased TNF-α secretion. Overall, this microfluidics-based approach for fabricating silk particles utilizing PVA as a means to induce phase separation and silk self-assembly is a promising approach to control particle size and size distribution. These silk particles may be utilized for a variety of biomedical applications including drug delivery to multiple cell types within a tumor microenvironment.
