Biodiversity data rescue in the framework of a long-term Kenya-Belgium cooperation in marine sciences.

ABSTRACT: The Kenya-Belgium data collection includes about 111,800 biotic observations on benthos, algae, fish, zooplankton, phytoplankton, birds and mangroves which cover more than 400 unique locations that were sampled between 1873 and 1999. The scope of this data digitization project was to recover data in theses and reports resulting from marine and coastal research activities in the Eastern African region conducted between 1984 and 1999. Data were digitized and quality checked in the frame of the Belgian LifeWatch project. The dataset provides a better insight into the different types of research conducted between 1985 and 1996 in frame of the Kenya-Belgium cooperation in marine sciences (KBP) project and can facilitate further coastal biodiversity research in Kenya.

Project description:Humans have created an accelerating, increasingly connected, globalized economy, resulting in a more globalized, shared flora. The prevention of new, establishing species is less costly, both economically and ecologically, and is more manageable than eradicating nonnative invasive species once they are widespread and negatively impactful. We ask if international trade hubs and points-of-entry with high-volume trade, constant disturbance, and propagule rain have a higher number of nonnative species compared to surrounding areas and if they may serve as initial establishment sites and refugia of nonnative, invasive populations. Therefore, we partnered with various federal, state, and private interests to evaluate the floristic composition at the Garden City Terminal of the Port of Savannah, Georgia, USA. We conducted the following study to demonstrate the collaborative relationship-building between researchers and industry and to develop a framework for biodiversity conservation. In our study, we collected all reproductive vascular plants in the secured areas of the Garden City Terminal during four major seasonal time points over two years. The percent of nonnative species and number of nonnative plant species per hectare at this industrial location exceeded all other comparison floras. The mean coefficient of conservatism was lowest among the comparison floras, indicating a highly disturbed habitat with nonnative, weedy native, and other native species tolerant of disturbance. Our study represents one of the first inventories of an Industrialized Flora and indicates that such areas are hot-spots of nonnative plant diversity and possible sources of emergent plant invasions. We posit that industrial sites and international points-of-entry should be considered laboratories for research on species transport and introduction, adaptability, and taxonomic delineation to better understand the mechanisms and consequences of biotic homogenization due to the volume and frequency of anthropogenic activities.

Project description:Cooperation based on the production of costly common goods is observed throughout nature. This is puzzling, as cooperation is vulnerable to exploitation by defectors which enjoy a fitness advantage by consuming the common good without contributing fairly. Depletion of the common good can lead to population collapse and the destruction of cooperation. However, population collapse implies small population size, which, in a structured population, is known to favor cooperation. This happens because small population size increases variability in cooperator frequency across different locations. Since individuals in cooperator-dominated locations (which are most likely cooperators) will grow more than those in defector-dominated locations (which are most likely defectors), cooperators can outgrow defectors globally despite defectors outgrowing cooperators in each location. This raises the possibility that defectors can lead to conditions that sometimes rescue cooperation from defector-induced destruction. We demonstrate multiple mechanisms through which this can occur, using an individual-based approach to model stochastic birth, death, migration, and mutation events. First, during defector-induced population collapse, defectors occasionally go extinct before cooperators by chance, which allows cooperators to grow. Second, empty locations, either preexisting or created by defector-induced population extinction, can favor cooperation because they allow cooperator but not defector migrants to grow. These factors lead to the counterintuitive result that the initial presence of defectors sometimes allows better survival of cooperation compared to when defectors are initially absent. Finally, we find that resource limitation, inducible by defectors, can select for mutations adaptive to resource limitation. When these mutations are initially present at low levels or continuously generated at a moderate rate, they can favor cooperation by further reducing local population size. We predict that in a structured population, small population sizes precipitated by defectors provide a "built-in" mechanism for the persistence of cooperation.

Project description:Long-term potentiation (LTP) is an activity-dependent and persistent increase in synaptic transmission. Currently available techniques to measure LTP are time-intensive and require highly specialized expertise and equipment, and thus are not well suited for screening of multiple candidate treatments, even in animal models. To expand and facilitate the analysis of LTP, here we use a flow cytometry-based method to track chemically induced LTP by detecting surface AMPA receptors in isolated synaptosomes: fluorescence analysis of single-synapse long-term potentiation (FASS-LTP). First, we demonstrate that FASS-LTP is simple, sensitive, and models electrically induced LTP recorded in intact circuitries. Second, we conducted FASS-LTP analysis in two well-characterized Alzheimer's disease (AD) mouse models (3xTg and Tg2576) and, importantly, in cryopreserved human AD brain samples. By profiling hundreds of synaptosomes, our data provide the first direct evidence to support the idea that synapses from AD brain are intrinsically defective in LTP. Third, we used FASS-LTP for drug evaluation in human synaptosomes. Testing a panel of modulators of cAMP and cGMP signaling pathways, FASS-LTP identified vardenafil and Bay-73-6691 (phosphodiesterase-5 and -9 inhibitors, respectively) as potent enhancers of LTP in synaptosomes from AD cases. These results indicate that our approach could provide the basis for protocols to study LTP in both healthy and diseased human brains, a previously unattainable goal.Significance statementLearning and memory depend on the ability of synapses to strengthen in response to activity. Long-term potentiation (LTP) is a rapid and persistent increase in synaptic transmission that is thought to be affected in Alzheimer's disease (AD). However, direct evidence of LTP deficits in human AD brain has been elusive, primarily due to methodological limitations. Here, we analyze LTP in isolated synapses from AD brain using a novel approach that allows testing LTP in cryopreserved brain. Our analysis of hundreds of synapses supports the idea that AD-diseased synapses are intrinsically defective in LTP. Further, we identified pharmacological agents that rescue LTP in AD, thus opening up a new avenue for drug screening and evaluation of strategies for alleviating memory impairments.
