PET-RAFT Polymerization Catalyzed by Small Organic Molecule under Green Light Irradiation.

ABSTRACT: Photocatalyzed polymerization using organic molecules as catalysts has attracted broad interest because of its easy operation in ambient environments and low toxicity compared with metallic catalysts. In this work, we reported that 4,7-di(thiophen-2-yl)benzo[c][1,2,5]thiadiazole (DTBT) can act as an efficient photoredox catalyst for photoinduced electron transfer-reversible addition-fragmentation chain transfer (PET-RAFT) polymerization under green light irradiation. Well-defined (co)polymers can be obtained using this technique without any additional additives like noble metals and electron donors or acceptors. The living characteristics of polymerization were verified by kinetic study and the narrow dispersity (Đ) of the produced polymer. Excellent chain-end fidelity was demonstrated through chain extension as well. In addition, this technique showed great potential for various RAFT agents and monomers including acrylates and acrylamides.

Guiding the Design of Organic Photocatalyst for PET-RAFT Polymerization: Halogenated Xanthene Dyes.

Project description:By examining structurally similar halogenated xanthene dyes, this study establishes a guiding principle for resolving structure-property- performance relationships in the photocontrolled PET-RAFT polymerization system (PET-RAFT: photoinduced electron/energy transfer-reversible addition-fragmentation chain transfer). We investigated the effect of the halogen substituents on the photophysical and electrochemical properties of the xanthene dyes acting as photocatalysts and their resultant effect on the performance of PET-RAFT polymerization. Consideration of the structure- property-performance relationships allowed design of a new xanthene photocatalyst, where its photocatalytic activity (oxygen tolerance and polymerization rate) was successfully optimized for PET-RAFT polymerization. We expect that this study will serve as a theoretical framework in broadly guiding the design of high performance photocatalysts for organic photocatalysis.

Light and magnetism dual-gated photoinduced electron transfer-reversible addition-fragmentation chain transfer (PET-RAFT) polymerization.

Project description:A novel raspberry-like γ-Fe2O3@carbon dot (CD) nanocatalyst was prepared and applied for photoinduced electron transfer-reversible addition-fragmentation chain transfer (PET-RAFT) polymerization. The nanocatalyst was found to be an efficient photocatalyst in visible light-regulated PET-RAFT polymerization owing to the oxidative quenching mechanism between the photoexcited γ-Fe2O3@CDs and the RAFT agent in the PET process. Notably, polymerization can be reversibly ceased in the absence of light or under an external magnetic field. The superparamagnetic nature and high saturation magnetization value (∼30.4 emu g-1) of the nanocatalyst contribute to convenient recycling of the nanocatalyst after polymerization. The PET-RAFT polymerization with the nanocatalyst before and after recycling was investigated, which displayed all the characteristics of controlled/living polymerization systems.

Automated PET-RAFT Polymerization Towards Pharmaceutical Amorphous Solid Dispersion Development.

Project description:In pharmaceutical oral drug delivery development, about 90% of drugs in the pipeline have poor aqueous solubility leading to severe challenges with oral bioavailability and translation to effective and safe drug products. Amorphous solid dispersions (ASDs) have been utilized to enhance the oral bioavailability of poorly soluble active pharmaceutical ingredients (APIs). However, a limited selection of regulatory-approved polymer excipients exists for the development and further understanding of tailor-made ASDs. Thus, a significant need exists to better understand how polymers can be designed to interact with specific API moieties. Here, we demonstrate how an automated combinatorial library approach can be applied to the synthesis and screening of polymer excipients for the model drug probucol. We synthesized a library of 25 random heteropolymers containing one hydrophilic monomer (2-hydroxypropyl acrylate (HPA)) and four hydrophobic monomers at varied incorporation. The performance of ASDs made by a rapid film casting method was evaluated by dissolution using ultra-performance liquid chromatography (UPLC) sampling at various time points. This combinatorial library and rapid screening strategy enabled us to identify a relationship between polymer hydrophobicity, monomer hydrophobic side group geometry, and API dissolution performance. Remarkably, the most effective synthesized polymers displayed slower drug release kinetics compared to industry standard polymer excipients, showing the ability to modulate the drug release profile. Future coupling of high throughput polymer synthesis, high throughput screening (HTS), and quantitative modeling would enable specification of designer polymer excipients for specific API functionalities.

Green and Effective Removal of Aqueous Graphene Oxide under UV-Light Irradiation.

Project description:The potential extensive application of graphene oxide (GO) in various fields results in the possibility of its release into the natural environment with negative impacts on humans and the ecosystem. The UV-induced removal behavior of aqueous GO was evaluated in this study, and the effect of various parameters (including initial GO concentration, initial solution pH and co-existing ions) on removal rate of GO were investigated in detail. The results showed that UV-light induced a maximum removal rate of GO of 99.1% after 32 h irradiation without any additives, and that the photo-induced removal process in all cases fitted well with pseudo-first-order kinetics. Under optimal conditions, GO was completely removed, with initial GO concentrations of 10 mg/L while adjusting solution pH to 3 or adding Ca2+-containing salt. The GO and photoreduced graphene oxide (prGO) were characterized using High-resolution Transmission Microscopy (HRTEM), X-ray Photoelectron Spectroscopy (XPS), and Fourier-transform Infrared Spectroscopy (FT-IR). The radical species trapping experiments and Electron Spin Resonance (ESR) tests indicated that self-reduction of GO upon UV-light exposure could be achieved via photogenerated electrons from a GO semiconductor. Further mechanism study showed that the high efficiency of UV-induced GO removal came from UV-induced photoreduction, and pH-induced or cation-induced coagulation. This study provided a green and effective method to remove GO from aqueous solutions.

Synthesis of Polymer Bioconjugates via Photoinduced Atom Transfer Radical Polymerization under Blue Light Irradiation.

Project description:A rapid blue-light-induced atom transfer radical polymerization (ATRP) was conducted in a biologically friendly environment. Well-controlled polymerization of oligo(ethylene oxide) methyl ether methacrylate (OEOMA) was successfully performed in aqueous media (1X PBS) under irradiation by blue LED strips. With 10.0 mW/cm2 intensity output at 450 nm, >90% conversion was achieved in 2 h in the presence of a system comprising glucose, glucose oxidase, and sodium pyruvate. Poly-(OEOMA) was synthesized with predetermined M n and low dispersities using low ppm of Cu catalysts. Importantly, secondary structures of proteins, as analyzed by circular dichroism (CD), were preserved under blue-light irradiation due to its lower energy output. The aqueous blue-light ATRP technique was applied to biological systems by synthesizing well-defined protein-polymer and DNA-polymer hybrids by the "grafting-from" method.

Porphyrin-based donor-acceptor COFs as efficient and reusable photocatalysts for PET-RAFT polymerization under broad spectrum excitation.

Project description:Covalent organic frameworks (COFs) are crystalline and porous organic materials attractive for photocatalysis applications due to their structural versatility and tunable optical and electronic properties. The use of photocatalysts (PCs) for polymerizations enables the preparation of well-defined polymeric materials under mild reaction conditions. Herein, we report two porphyrin-based donor-acceptor COFs that are effective heterogeneous PCs for photoinduced electron transfer-reversible addition-fragmentation chain transfer (PET-RAFT). Using density functional theory (DFT) calculations, we designed porphyrin COFs with strong donor-acceptor characteristics and delocalized conduction bands. The COFs were effective PCs for PET-RAFT, successfully polymerizing a variety of monomers in both organic and aqueous media using visible light (λ max from 460 to 635 nm) to produce polymers with tunable molecular weights (MWs), low molecular weight dispersity, and good chain-end fidelity. The heterogeneous COF PCs could also be reused for PET-RAFT polymerization at least 5 times without losing photocatalytic performance. This work demonstrates porphyrin-based COFs that are effective catalysts for photo-RDRP and establishes design principles for the development of highly active COF PCs for a variety of applications.

Photocatalytic degradation of dissolved organic matter under ZnO-catalyzed artificial sunlight irradiation system.

Project description:This study investigates the photocatalytic degradation of dissolved organic matter (DOM) under ZnO-assisted artificial sunlight system at various conditions (ZnO dosage, pH, and the presence of Cl-, SO42-, and HCO3-). The results show that the degradation of DOM follows a pseudo-first-order kinetics. Fluorescence excitation-emission matrices coupled with parallel factor (EEM-PARAFAC) analysis decomposes DOM into two fluorophores (C1 and C2). The total removals and photodegradation rates calculated with DOC, UV254, and the Fmax of C1 are similar, increasing with higher ZnO dosages and being highest in pH 7 and lowest in pH 4. ZnO dosage has a similar effect on DOM degradation when assessed using C2, as with C1, but pH effect is not consistent. As for the anions, HCO3- shows the strongest inhibition for DOC, UV254 and C1 while Cl- has the strongest facilitation effect for C2. The total removal and photodegradation rates calculated with the Fmax of C1 and C2 are higher than those calculated using DOC and UV254. This study demonstrates that the successful application of EEM-PARAFAC analysis in addition to traditional parameters can provide further insight into the photocatalytic degradation mechanisms associated with DOM in conjunction with a ZnO catalyst under artificial sunlight.

An Oxygen-Tolerant PET-RAFT Polymerization for Screening Structure-Activity Relationships.

Project description:The complexity of polymer-protein interactions makes rational design of the best polymer architecture for any given biointerface extremely challenging, and the high throughput synthesis and screening of polymers has emerged as an attractive alternative. A porphyrin-catalysed photoinduced electron/energy transfer-reversible addition-fragmentation chain-transfer (PET-RAFT) polymerisation was adapted to enable high throughput synthesis of complex polymer architectures in dimethyl sulfoxide (DMSO) on low-volume well plates in the presence of air. The polymerisation system shows remarkable oxygen tolerance, and excellent control of functional 3- and 4-arm star polymers. We then apply this method to investigate the effect of polymer structure on protein binding, in this case to the lectin concanavalin A (ConA). Such an approach could be applied to screen the structure-activity relationships for any number of polymer-protein interactions.

Self-Healing Hydrogel Scaffolds through PET-RAFT Polymerization in Cellular Environment.

Project description:Photo electron/energy transfer-reversible addition-fragmentation chain transfer (PET-RAFT) has emerged as a powerful reversible-deactivation radical polymerization technique, enabling oxygen-tolerant polymerizations with exquisite spatiotemporal control through irradiation with visible light. In contrast to traditional free radical photo-polymerization, which often requires the use of DNA-damaging UV irradiation, PET-RAFT offers a more cytocompatible alternative for the preparation of polymeric materials in cell culture environments. Herein, we report the use of PET-RAFT for the fabrication of self-healing hydrogels using commercially available monomers, reaching high monomer conversions and cell encapsulation efficiencies. Our hydrogels showed the expected rheological and mechanical properties for the systems considered, together with excellent cytocompatibility and spatiotemporal control over the polymerization process. Moreover, hydrogels prepared through this method could be cut and healed again by simply adding further monomer and irradiating the system with visible light, even in the presence of mammalian cells. This study demonstrates for the first time the potential of PET-RAFT polymerization as a viable methodology for the synthesis of self-healing hydrogel scaffolds for cell encapsulation.

A Green Platform for Preparation of the Well-Defined Polyacrylonitrile: <sup>60</sup>Co γ-ray Irradiation-Initiated RAFT Polymerization at Room Temperature.

Project description:60Co γ-ray irradiation-initiated reversible addition⁻fragmentation chain transfer (RAFT) polymerization at room temperature with 2-cyanoprop-2-yl 1-dithionaphthalate (CPDN) as the chain transfer agent was first applied to acrylonitrile (AN) polymerization, providing a "green" platform for preparing polyacrylonitrile (PAN)-based carbon fibers using an environment-friendly energy source. Various effects of dose rate, molar ratio of the monomer to the chain transfer agent, monomer concentration and reaction time on the AN polymerization behaviors were performed to improve the controllability of molecular the weight and molecular weight distribution of the obtained PAN. The feature of the controlled polymerization was proven by the first-order kinetics, linear increase of the molecular weight with the monomer conversion and a successful chain-extension experiment. The molecular weight and molecular weight distribution of PAN were characterized by size exclusion chromatography (SEC). ¹H NMR and Matrix assisted laser desorption ionization/time of flight mass spectra (MALDI-TOF-MS) confirmed the chain-end functionality of PAN, which also was supported by the successful chain-extension experiments of original PANs with acrylonitrile and styrene as the second monomers respectively.
