A neuronal correlate of insect stereopsis.

ABSTRACT: A puzzle for neuroscience-and robotics-is how insects achieve surprisingly complex behaviours with such tiny brains. One example is depth perception via binocular stereopsis in the praying mantis, a predatory insect. Praying mantids use stereopsis, the computation of distances from disparities between the two retinal images, to trigger a raptorial strike of their forelegs when prey is within reach. The neuronal basis of this ability is entirely unknown. Here we show the first evidence that individual neurons in the praying mantis brain are tuned to specific disparities and eccentricities, and thus locations in 3D-space. Like disparity-tuned cortical cells in vertebrates, the responses of these mantis neurons are consistent with linear summation of binocular inputs followed by an output nonlinearity. Our study not only proves the existence of disparity sensitive neurons in an insect brain, it also reveals feedback connections hitherto undiscovered in any animal species.

Insect stereopsis demonstrated using a 3D insect cinema.

Project description:Stereopsis - 3D vision - has become widely used as a model of perception. However, all our knowledge of possible underlying mechanisms comes almost exclusively from vertebrates. While stereopsis has been demonstrated for one invertebrate, the praying mantis, a lack of techniques to probe invertebrate stereopsis has prevented any further progress for three decades. We therefore developed a stereoscopic display system for insects, using miniature 3D glasses to present separate images to each eye, and tested our ability to deliver stereoscopic illusions to praying mantises. We find that while filtering by circular polarization failed due to excessive crosstalk, "anaglyph" filtering by spectral content clearly succeeded in giving the mantis the illusion of 3D depth. We thus definitively demonstrate stereopsis in mantises and also demonstrate that the anaglyph technique can be effectively used to deliver virtual 3D stimuli to insects. This method opens up broad avenues of research into the parallel evolution of stereoscopic computations and possible new algorithms for depth perception.

A novel dynamic random-dot stereopsis assessment to measure stereopsis in intermittent exotropia.

Project description:BackgroundTo investigate dynamic stereopsis in intermittent exotropia [X(T)] patients using a novel dynamic random-dot stereopsis assessment method.MethodsA novel dynamic random-dot stereopsis test was performed using novel self-programmed software, which consisted of red and green dots and four blocks on the screen. The test included motion + disparity (MD), motion (M), and disparity (D), where the D cues ranged from 200 to 1,200 arc-seconds. The characteristics of preoperative dynamic stereopsis in 83 X(T) patients and associations with clinical features were analysed, and the prognosis was followed up on the 1st day and at the 2nd, 6th and 12th months postoperatively.ResultsPreoperatively, the mean reciprocal values of near and far stereopsis were 0.013±0.0016 and 0.0011±0.0005 arc-seconds in static stereopsis patients, respectively, and the MD, M, and D values were 0.002±0.0002, 0.0018±0.0001, and 0.0012±0.0002 arc-seconds in dynamic stereopsis, respectively. The MD value was negatively correlated with the Newcastle control score, disease course, and prism deviations (P<0.05), M was correlated with disease course and the Newcastle control score (P<0.05), and D was not correlated with any clinical features. Postoperative dynamic stereopsis improved from the 1st day and gradually peaked at the 6th month, while static stereopsis showed a decreased tendency in near but not far stereopsis.ConclusionsThe dynamic stereopsis quantitative evaluation method based on random dots is a feasible test and shows that destruction of X(T) patients' dynamic stereopsis is affected by age at surgery, disease course, strabismus controllability and the strabismus degree.

Neuronal responses to omitted tones in the auditory brain: A neuronal correlate for predictive coding.

Project description:Prediction provides key advantages for survival, and cognitive studies have demonstrated that the brain computes multilevel predictions. Evidence for predictions remains elusive at the neuronal level because of the complexity of separating neural activity into predictions and stimulus responses. We overcome this challenge by recording from single neurons from cortical and subcortical auditory regions in anesthetized and awake preparations, during unexpected stimulus omissions interspersed in a regular sequence of tones. We find a subset of neurons that responds reliably to omitted tones. In awake animals, omission responses are similar to anesthetized animals, but larger and more frequent, indicating that the arousal and attentional state levels affect the degree to which predictions are neuronally represented. Omission-sensitive neurons also responded to frequency deviants, with their omission responses getting emphasized in the awake state. Because omission responses occur in the absence of sensory input, they provide solid and empirical evidence for the implementation of a predictive process.

Distinct C9orf72-Associated Dipeptide Repeat Structures Correlate with Neuronal Toxicity.

Project description:Hexanucleotide repeat expansions in C9orf72 are the most common inherited cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). The expansions elicit toxicity in part through repeat-associated non-AUG (RAN) translation of the intronic (GGGGCC)n sequence into dipeptide repeat-containing proteins (DPRs). Little is known, however, about the structural characteristics and aggregation propensities of the dipeptide units comprising DPRs. To address this question, we synthesized dipeptide units corresponding to the three sense-strand RAN translation products, analyzed their structures by circular dichroism, electron microscopy and dye binding assays, and assessed their relative toxicity when applied to primary cortical neurons. Short, glycine-arginine (GR)3 dipeptides formed spherical aggregates and selectively reduced neuronal survival compared to glycine-alanine (GA)3 and glycine-proline (GP)3 dipeptides. Doubling peptide length had little effect on the structure of GR or GP peptides, but (GA)6 peptides formed β-sheet rich aggregates that bound thioflavin T and Congo red yet lacked the typical fibrillar morphology of amyloids. Aging of (GA)6 dipeptides increased their β-sheet content and enhanced their toxicity when applied to neurons. We also observed that the relative toxicity of each tested dipeptide was proportional to peptide internalization. Our results demonstrate that different C9orf72-related dipeptides exhibit distinct structural properties that correlate with their relative toxicity.

Transfer of Perceptual Learning From Local Stereopsis to Global Stereopsis in Adults With Amblyopia: A Preliminary Study.

Project description:It has long been debated whether the analysis of global and local stereoscopic depth is performed by a single system or by separate systems. Global stereopsis requires the visual system to solve a complex binocular matching problem to obtain a coherent percept of depth. In contrast, local stereopsis requires only a simple matching of similar image features. In this preliminary study, we recruited five adults with amblyopia who lacked global stereopsis and trained them on a computerized local stereopsis depth task for an average of 12 h. Three out of five (60%) participants recovered fine global stereoscopic vision through training. Those who recovered global stereopsis reached a learning plateau more quickly on the local stereopsis task, and they tended to start the training with better initial local stereopsis performance, to improve more on local stereopsis with training, and to have less severe amblyopia. The transfer of learning from local stereopsis to global stereopsis is compatible with an interacting two-stage model.

Mapping residual stereopsis in macular degeneration.

Project description:Individuals with macular degeneration typically lose vision in the central region of one or both eyes. A binocular scotoma occurs when vision loss occurs in overlapping locations in both eyes, but stereopsis is impacted even in the non-overlapping region wherever the visual field in either eye is affected. We used a novel stereoperimetry protocol to measure local stereopsis across the visual field (up to 25° eccentricity) to determine how locations with functional stereopsis relate to the scotomata in the two eyes. Participants included those with monocular or binocular scotomata and age-matched controls with healthy vision. Targets (with or without depth information) were presented on a random dot background. Depth targets had true binocular disparity of 20' (crossed), whereas non-depth targets were defined by monocular cues such as contrast and dot density. Participants reported target location and whether it was in depth or flat. Local depth sensitivity (d') estimates were then combined to generate a stereopsis map. This stereopsis map was compared to the union of the monocular microperimetry estimates that mapped out the functional extent of the scotoma in each eye. The "union" prediction aligned with residual stereopsis, showing impaired stereopsis within this region and residual stereopsis outside this region. Importantly, the stereoblind region was typically more extensive than the binocular scotoma defined by the intersection (overlap) of the scotomata. This explains why individuals may have intact binocular visual fields but be severely compromised in tasks of daily living that benefit from stereopsis, such as eye-hand coordination and navigation.

Contextual Binocular Imbalance Impairs Local Stereopsis.

Project description:PurposeBinocular imbalance is known to inhibit stereopsis. This study investigates whether an imbalanced context around stereo stimuli also affects local stereopsis and explores the underlying mechanisms.MethodsThree experiments were conducted with normally sighted participants. Experiment 1 measured local stereo detection thresholds under three context conditions: binocular balance (0.5 vs. 0.5 contrast), left-eye dominance (0.8 vs. 0.2 contrast), and right-eye dominance (0.2 vs. 0.8 contrast). Experiment 2 assessed the modulation of the imbalance effect by context-target collinearity. Experiment 3 examined the imbalance effect with binocular fusion and rivalry context stimuli.ResultsIn experiment 1, the average stereo threshold was 62.4 arcsec in the binocular balance condition, elevated to 111.4 arcsec in the left-eye dominance (P = 0.003), and 114.7 arcsec in the right-eye dominance (P < 0.001), with no significant difference between the two imbalance conditions (P = 0.650). Experiment 2 showed that context-target collinearity modulated the imbalance effect, resulting in a smaller threshold elevation in the non-collinear condition (P = 0.011). Experiment 3 revealed significant main effects of imbalance (P = 0.031) and rivalry (P = 0.004), with no significant interaction (P = 0.966).ConclusionsContextual binocular imbalance inhibits local stereopsis, an effect modulated by collinearity and similarly observed in both binocular integrative and suppressive contexts. These findings suggest that lateral connectivity in the primary visual cortex (V1) plays a fundamental role in stereopsis generation, offering novel approaches for clinical interventions aimed at restoring binocular balance and stereopsis.

Balanced Binocular Inputs Support Superior Stereopsis.

Project description:PurposeOur visual system compares the inputs received from the two eyes to estimate the relative depths of features in the retinal image. We investigated how an imbalance in the strength of the input received from the two eyes affects stereopsis. We also explored the level of agreement between different measurements of sensory eye imbalance.MethodsWe measured the sensory eye imbalance and stereoacuity of 30 normally sighted participants. We made our measurements using a modified amblyoscope. The sensory eye imbalance was assessed through three methods: the difference between monocular contrast thresholds, the difference in dichoptic masking weight, and the contribution of each eye to a fused binocular percept. We referred them as the "threshold imbalance," "masking imbalance," and "fusion imbalance," respectively. The stereoacuity threshold was measured by having subjects discriminate which of four circles were displaced in depth. All of our tests were performed using stimuli of the same spatial frequency (2.5 cycles/degree).ResultsWe found a relationship between stereoacuity and sensory eye imbalance. However, this was only the case for fusion imbalance measurement (ρ = 0.52; P = 0.003). Neither the threshold imbalance nor the masking imbalance was significantly correlated with stereoacuity. We also found the threshold imbalance was correlated with both the fusion and masking imbalances (r = 0.46, P = 0.011 and r = 0.49, P = 0.005, respectively). However, a nonsignificant correlation was found between the fusion and masking imbalances.ConclusionsOur findings suggest that there exist multiple types of sensory eye dominance that can be assessed by different tasks. We find only imbalances in dominance that result in biases to fused percepts are correlated with stereoacuity.

Indoxacarb, an oxadiazine insecticide, blocks insect neuronal sodium channels.

Project description:1. Decarbomethoxyllated JW062 (DCJW), the active component of a new oxadiazine insecticide DPX-JW062 (Indoxacarb), was tested on action potentials and the inward sodium current recorded from short-term cultured dorsal unpaired median neurones of the cockroach Periplaneta americana. 2. Under whole-cell current-clamp conditions, 100 nM DCJW reduced the amplitude of action potentials and induced a large hyperpolarization of the resting membrane potential associated with a 41% increase in input resistance. 3. In voltage-clamp, DCJW resulted in a dose-dependent inhibition (IC(50) 28 nM) of the peak sodium current. Based on IC(50) values, the effect of DCJW was about 10 fold less potent than tetrodotoxin (TTX) but 1000 fold more potent than the local anaesthetic lidocaine. DCJW (100 nM) was without effect on activation properties of the sodium current, reversal potential, voltage dependence of sodium conductance and on both fast and slow steady-state inactivations. 4. TTX (2 nM) resulted in 48% inhibition of the peak inward sodium current. Co-application of TTX (2 nM) with various concentrations of DCJW produced an additional inhibition of the peak inward current, indicating that the blocking actions of DCJW and TTX were distinct. Co-application of lidocaine (IC(50) 30 microM) with various concentrations of DCJW produced a reduction of the apparent potency of DCJW, suggesting that DCJW and lidocaine acted at the same site. 5. DCJW (100 nM) did not affect inward calcium or outward potassium currents. 6. This study describes, for the first time, the action on insect neuronal voltage-dependent sodium channels of Indoxacarb, a new class of insecticides.

Cuttlefish use stereopsis to strike at prey.

Project description:The camera-type eyes of vertebrates and cephalopods exhibit remarkable convergence, but it is currently unknown whether the mechanisms for visual information processing in these brains, which exhibit wildly disparate architecture, are also shared. To investigate stereopsis in a cephalopod species, we affixed "anaglyph" glasses to cuttlefish and used a three-dimensional perception paradigm. We show that (i) cuttlefish have also evolved stereopsis (i.e., the ability to extract depth information from the disparity between left and right visual fields); (ii) when stereopsis information is intact, the time and distance covered before striking at a target are shorter; (iii) stereopsis in cuttlefish works differently to vertebrates, as cuttlefish can extract stereopsis cues from anticorrelated stimuli. These findings demonstrate that although there is convergent evolution in depth computation, cuttlefish stereopsis is likely afforded by a different algorithm than in humans, and not just a different implementation.
