First Descriptions of Larva and Pupa of Bagous claudicans Boheman, 1845 (Curculionidae, Bagoinae) and Systematic Position of the Species Based on Molecular and Morphological Data.

ABSTRACT: In this paper, the mature larva and pupa of Bagous claudicans are described and illustrated for the first time. Measurements of younger larval instars are also given. The biology of the species is discussed in association with larval morphology and feeding habits. Overall larval and pupal morphological characters of the genus Bagous are presented. Confirmation of the larva identification as Bagous claudicans species was conducted by cytochrome oxidase I (COI) sequencing. DNA barcoding was useful for specimen identification of larval stages. The systematic position of the species within the Bagous collignensis-group, based on morphological and molecular results, is also discussed.

The complete mitochondrial genome of Lixus subtilis Boheman, 1835 (Coleoptera, Curculionidae) and its phylogenetic implications.

Project description:The first complete mitochondrial genome of Lixus subtilis Boheman is reported in this study. The circular genome is 15,223 bp long, including a standard set of 21 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs), 13 protein-coding genes, and a non-coding control region. The trnI gene was not found in the L. subtilis mitogenome. All tRNAs had the typical cloverleaf structure, except for trnS1, which lacked the dihydrouridine arm. The phylogenetic tree of 13 Curculionidae species based on the concatenated nucleotide sequences of complete mitochondrial genomes strongly supported that L. subtilis is closely related to Curculioninae and Molytinae.

Review of the Parasa undulata (Cai, 1983) species group with the first conifer-feeding larva for Limacodidae and descriptions of two new species from China and Taiwan (Lepidoptera, Limacodidae).

Project description:Although the caterpillars are well-known for the stings and magnificent coloration, the systematics of Limacodidae is historically neglected and chaotic due to the difficulty in matching the larval with adult stages as well as the very conservative and convergent adult morphology. One of the biggest taxonomic problems surrounds a collective group from Southeastern Asia, termed the "green limacodid moths", which harbours at least 90 species placed in the genus Parasa Walker, 1859 and 14 "subunits". The P. undulata group was previously composed of 3 species from China and Taiwan, and characterized only by wing pattern. This species group is extensively studied herein with two new species described, i.e. P. viridiflamma sp. n. (Taiwan) and P. minwangi sp. n. (S. China), and discovery of female genitalia of three species, presenting new phylogenetic insights in this potentially paraphyletic genus. In addition, one limacodid larva was found to be feeding exclusively on Picea (Pinaceae) in Taiwan. Its identity, Parasa pygmy Solovyev, 2010 in P. undulata group, is confirmed through matching its COI sequence to the adult. This discovery is also biologically significant because the previous known host breadth of Parasa was of polyphagy on various angiosperm plant families. This case, therefore, represents the first record of conifer-feeding behavior in this family as well as the first of specialized herbivory in the genus. Meanwhile, the background match between Picea leaves and larval coloration is shared with other Picea-feeding insects. This phenomenon is worth of further investigation in the aspect of convergent evolution of crypsis associated with a particular plant.

Project description:Species of the genus Pseudocneorhinus occurring in or near China are reviewed, with description of five new species, Pseudocneorhinusangustus sp. nov., P.glaber sp. nov., P.hlavaci sp. nov., P.obliquehumeralis sp. nov., and P.setosicallus sp. nov. from the provinces of Beijing, Gansu, Shaanxi, Sichuan, and Zhejiang. They are illustrated and compared with similar species, and a key is provided to all presently known species of the genus. Lectotypes of the following species are designated: Callirhopalussubcallosus Voss, 1956 [current name Pseudocneorhinussubcallosus (Voss, 1956)] and P.squamosus Marshall, 1934. Pseudocneorhinussquameus Morimoto, 2015 is confirmed for the fauna of China.

Project description:The adult, pupa and larva of a new species, Gnaptorina (Gnaptorina) lhorongica Li, sp. nov., from northeastern Xizang, China are described and illustrated. The species was identified using molecular phylogenetic analyses based on three mitochondrial fragments and one nuclear gene fragment (COI, Cytb, 16S, and 28S-D2). The taxonomic status of the new species is confirmed using a combination of molecular and morphological datasets. This study provides valuable molecular and morphological data for phylogenetic studies of the tribe Blaptini.

A revision of the North American genus Proctorus (Coleoptera, Curculionidae, Ellescini) with descriptions of two new species.

Project description:The rarely collected North American endemic genus Proctorus (Coleoptera, Curculionidae, Ellescini) has hitherto contained two described species, P.armatus LeConte, 1876 and P.decipiens (LeConte, 1876). Here, Proctorus is revised and two new species, namely P.emarginatus sp. nov. and P.truncatus sp. nov., are described. Lectotypes for P.armatus and P.decipiens are designated from known syntypes. All four species in the genus are associated with Salicaceae, but, in addition to differences in external and genital morphology, there is also evidence of differing host plant usage between the species. A photographic key to the four species is provided to facilitate identification.

Insight into weevil biology from a reference quality genome of the boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae).

Project description:The boll weevil, Anthonomus grandis grandis Boheman, is one of the most historically impactful insects due to its near destruction of the US cotton industry in the early 20th century. Contemporary efforts to manage this insect primarily use pheromone baited traps for detection and organophosphate insecticides for control, but this strategy is not sustainable due to financial and environmental costs. We present a high-quality boll weevil genome assembly, consisting of 306 scaffolds with approximately 24,000 annotated genes, as a first step in the identification of gene targets for novel pest control. Gene content and transposable element distribution are similar to those found in other Curculionidae genomes; however, this is the most contiguous and only assembly reported to date for a member in the species-rich genus Anthonomus. Transcriptome profiles across larval, pupal, and adult life stages led to identification of several genes and gene families that could present targets for novel control strategies.

Two Splice Isoforms of Leptinotarsa Ecdysis Triggering Hormone Receptor Have Distinct Roles in Larva-Pupa Transition.

Project description:Insect ecdysis triggering hormone (ETH) receptors (ETHRs) are rhodopsin-like G protein-coupled receptors. Upon binding its ligand ETH, ETHR initiates a precisely programed ecdysis behavior series and physiological events. In Drosophila melanogaster, the ethr gene produces two functionally distinct splicing isoforms, ethra and ethrb. ETH/ETHRA activates eclosion hormone (EH), kinin, crustacean cardioactive peptide (CCAP), and bursicon (burs and pburs) neurons, among others, in a rigid order, to elicit the behavioral sequences and physiological actions for ecdysis at all developmental stages, whereas ETH/ETHRB is required at both pupal and adult ecdysis. However, the role of ETHRB in regulation of molting has not been clarified in any non-drosophila insects. In the present paper, we found that 20-hydroxyecdysone (20E) signaling triggers the expression of both ethra and ethrb in a Coleopteran insect pest, the Colorado potato beetle Leptinotarsa decemlineata. RNA interference (RNAi) was performed using double-stranded RNAs (dsRNAs) targeting the common (dsethr) or isoform-specific (dsethra, dsethrb) regions of ethr. RNAi of dsethr, dsethra, or dsethrb by the final-instar larvae arrested larva development. The arrest was not rescued by feeding 20E. All the ethra depleted larvae stopped development at prepupae stage; the body cavity was expanded by a large amount of liquid. Comparably, more than 80% of the ethrb RNAi larvae developmentally halted at the prepupae stage. The remaining Ldethrb hypomorphs became pupae, with blackened wings and highly-expressed burs, pburs and four melanin biosynthesis genes. Therefore, ETHRA and ETHRB play isoform-specific roles in regulation of ecdysis during larva-pupa transition in L. decemlineata.

Immature stages of Palearctic Mecinus species (Coleoptera, Curculionidae, Curculioninae): morphological characters diagnostic at genus and species levels.

Project description:The immature stages of ten Mecinus species are described for the first time and those of two other species are redescribed, adding important chaetotaxy characters that were missing from previous descriptions. These species belong to six of the nine assemblages of Mecinus species previously established according to a phylogenetic analysis. All these groupings are confirmed on the basis of several characters of mature larvae and pupae. Moreover, all the species show several characters that are useful for distinguishing them from each other, including cryptic species that previously had few differential characters. Some characters that may be useful for separating Mecinus from other genera in the tribe are suggested. To confirm the taxonomic identification of some larvae, the mtCOII gene was obtained and compared with sequences from identified adult specimens. The most important characters for separating the immature stages of the genera and species groups in Mecinus are the number of palpomeres of the labial palpi (1 or 2), the number of air tubes of the thoracic and abdominal spiracles (unicameral or bicameral), and the number of epipharyngeal setae. The species studied herein were compared with those known from other genera in the tribe Mecinini. Two keys, one to the described larvae and the other to the pupae, are provided. Detailed biological data, several of which are new, on some species are reported.

Redescription of the forgotten New Caledonian weevil genus Callistomorphus Perroud, 1865 (Coleoptera, Curculionidae, Eugnomini) with descriptions of eight new species.

Project description:Callistomorphus is one of the "forgotten" genera of the tribe Eugnomini inhabiting rain forest in New Caledonia. In this paper, the genus Callistomorphus and the type species C.farinosus are redescribed. Eight new species, Callistomorphusfundatus sp. n., C.gibbus sp. n., C.malleus sp. n., C.minimus sp. n., C.rutai sp. n., C.szoltysi sp. n., C.torosus sp. n. and C.turbidus sp. n., are described, originating from the main island of New Caledonia. Illustrations and SEM photographs of the external morphology and the male and female terminalia are provided, as well as dorsal habitus colour photographs of the adults, a key to the species, a distribution map, and a discussion of the systematic position of Callistomorphus within the tribe.
