Thermoresponsive Pyrrolidone Block Copolymer Organogels from 3D Micellar Networks.

ABSTRACT: A new series of amphiphilic pyrrolidone diblock copolymers poly[N-(2-methacrylaoyxyethyl)pyrrolidone]-block-poly(methyl methacrylate) (PNMP _m -b-PMMA _n ; where m is fixed at 37 and n is varied from 45 to 378) is developed. Spontaneously situ-gelling behaviors are observed in isopropanol when n varies from 117 to 230, whereas only dissolution or precipitation appears when n is beyond this region. Further analysis reveals that uniform thermoinduced reversible gel-sol transitions are observed in those organogels, which is attributed to the disassembly from micellar networks to micelles as confirmed by electron microscopy and other techniques. The gel-sol transition temperature is highly dependent on n and increases as n increases. Conformational interactions analyzed using ¹H NMR and 2D Noesy NMR suggest that the thermoinduced stretch of solvophilic PNMP segments within micelles and the sequencing variation in the isopropanol molecules are the major cause of the gel-sol transitions.

Quest for Thermoresponsive Block Copolymer Nanoparticles with Liquid-Crystalline Surfactant Cores.

Project description:This work reports on the preparation and characterization of anisotropic composition nanoparticles based on the electrostatic binding of dodecyltrimethylammonium surfactant to poly(acrylic acid) blocks of diblock copolymers with poly(ethylene oxide) (PEO) and poly(N-isopropyl acrylamide) (PNIPAm). These nanoparticles form kinetically stable dispersions and display liquid-crystalline cores with a micellar cubic structure, as determined by small-angle X-ray scattering. Mixtures with different proportions of the two block copolymers and stoichiometric amounts of C12TA+ were prepared and their behavior was compared with that of the parent nanoparticles. Upon heating, dilute dispersions (0.01 and 0.1 wt %) analyzed by dynamic light scattering display a slight decrease in the hydrodynamic radius, consistent with the dehydration of PNIPAm and mixed PNIPAm-PEO blocks at the shell. At higher concentrations, 2 wt %, the nanoparticles with pure PNIPAm shell undergo macroscopic phase separation above 32 °C. Nanoparticles with a pure PEO shell do not display temperature sensitivity. For the mixtures, no visual change is observed, but the dynamic light scattering results evidence the formation of clusters, whose size and reversibility depend on the PEO/PNIPAm proportion. This indicates the formation of mixed nanoparticles containing both PEO and PNIPAm blocks. Nuclear Overhauser enhancement spectroscopy NMR analyses of the mixtures do not show the correlation peak expected for PEO and PNIPAm blocks in close proximity, suggesting their segregation at the nanoparticle shell. On the basis of these results, we discuss the possibilities of the neutral blocks distribution on the shell of mixed nanoparticles. Overall, we have confirmed that these nanoparticles may display a temperature-controlled reversible aggregation while preserving their internal liquid-crystalline structures.

Thermoresponsive Block Copolymer Core-Shell Nanoparticles with Tunable Flow Behavior in Porous Media.

Project description:With the purpose of investigating new polymeric materials as potential flow modifiers for their future application in enhanced oil recovery (EOR), a series of amphiphilic poly(di(ethylene glycol) methyl ether methacrylate-co-oligo(ethylene glycol) methyl ether methacrylate) [P(DEGMA-co-OEGMA)]-based core-shell nanoparticles were prepared by aqueous reversible addition-fragmentation chain transfer-mediated polymerization-induced self-assembly. The developed nano-objects were shown to be thermoresponsive, demonstrating a reversible lower-critical solution temperature (LCST)-type phase transition with increasing solution temperature. Characterization of their thermoresponsive nature by variable-temperature UV-vis and dynamic light scattering analyses revealed that these particles reversibly aggregate when heated above their LCST and that the critical transition temperature could be accurately tuned by simply altering the molar ratio of core-forming monomers. Sandpack experiments were conducted to evaluate their pore-blocking performance at low flow rates in a porous medium heated at temperatures above their LCST. This analysis revealed that particles aggregated in the sandpack column and caused pore blockage with a significant reduction in the porous medium permeability. The developed aggregates and the increased pressure generated by the blockage were found to remain stable under the injection of brine and were observed to rapidly dissipate upon reducing the temperature below the LCST of each formulation. Further investigation by double-column sandpack analysis showed that the blockage was able to reform when re-heated and tracked the thermal front. Moreover, the rate of blockage formation was observed to be slower when the LCST of the injected particles was higher. Our investigation is expected to pave the way for the design of "smart" and versatile polymer technologies for EOR applications in future studies.

Temperature-modulated separation of vascular cells using thermoresponsive-anionic block copolymer-modified glass.

Project description:IntroductionVascular tissue engineering is a key technology in the field of regenerative medicine. In tissue engineering, the separation of vascular cells without cell modification is required, as cell modifications affect the intrinsic properties of the cells. In this study, we have developed an effective method for separating vascular cells without cell modification, using a thermoresponsive anionic block copolymer.MethodsA thermoresponsive anionic block copolymer, poly(acrylic acid)-b-poly(N-isopropylacryl-amide) (PAAc-b-PNIPAAm), with various PNIPAAm segment lengths, was prepared in two steps: atom transfer radical polymerization and subsequent deprotection. Normal human umbilical vein endothelial cells (HUVECs), normal human dermal fibroblasts, and human aortic smooth muscle cells (SMCs) were seeded onto the prepared thermoresponsive anionic block copolymer brush-modified glass. The adhesion behavior of cells on the copolymer brush was observed at 37 °C and 20 °C.ResultsA thermoresponsive anionic block copolymer, poly(acrylic acid)-b-poly(N-isopropylacrylamide) (PAAc-b-PNIPAAm), with various PNIPAAm segment lengths was prepared. The prepared copolymer-modified glass exhibited anionic properties attributed to the bottom PAAc segment of the copolymer brush. On the PAAc-b-PNIPAAm, which had a moderate PNIPAAm length, a high adhesion ratio of HUVECs and low adhesion ratio of SMCs were observed at 37 °C. By reducing temperature from 37 °C to 20 °C, the adhered HUVECs were detached, whereas the SMCs maintained adhesion, leading to the recovery of purified HUVECs by changing the temperature.ConclusionsThe prepared thermoresponsive anionic copolymer-modified glass could be used to separate HUVECs and SMCs by changing the temperature without modifying the cell surface. Therefore, the developed cell separation method will be useful for vascular tissue engineering.

Dual Reduction/Acid-Responsive Disassembly and Thermoresponsive Tunability of Degradable Double Hydrophilic Block Copolymer.

Project description:We report a thermoresponsive double hydrophilic block copolymer degradable in response to dual reduction and acidic pH at dual locations. The copolymer consists of a poly(ethylene oxide) block covalently connected through an acid-labile acetal linkage with a thermoresponsive polymethacrylate block containing pendant oligo(ethylene oxide) and disulfide groups. The copolymer undergoes temperature-driven self-assembly in water to form nanoassemblies with acetal linkages at the core/corona interface and disulfide pendants in the core, exhibiting dual reduction/acid responses at dual locations. The physically assembled nanoaggregates are converted to disulfide-core-crosslinked nanogels through disulfide-thiol exchange reaction, retaining enhanced colloidal stability, yet degraded to water-soluble unimers upon reduction/acid-responsive degradation. Further, the copolymer exhibits improved tunability of thermoresponsive property upon the cleavage of junction acetal and pendant disulfide linkages individually and in combined manner. This work suggests that dual location dual reduction/acid-responsive degradation is a versatile strategy toward effective drug delivery exhibiting disulfide-core-crosslinking capability and disassembly as well as improved thermoresponsive tunability.

Convoluted micellar morphological transitions driven by tailorable mesogenic ordering effect from discotic mesogen-containing block copolymer.

Project description:Solution-state self-assemblies of block copolymers to form nanostructures are tremendously attractive for their tailorable morphologies and functionalities. While incorporating moieties with strong ordering effects may introduce highly orientational control over the molecular packing and dictate assembly behaviors, subtle and delicate driving forces can yield slower kinetics to reveal manifold metastable morphologies. Herein, we report the unusually convoluted self-assembly behaviors of a liquid crystalline block copolymer bearing triphenylene discotic mesogens. They undergo unusual multiple morphological transitions spontaneously, driven by their intrinsic subtle liquid crystalline ordering effect. Meanwhile, liquid crystalline orderedness can also be built very quickly by doping the mesogens with small-molecule dopants, and the morphological transitions are dramatically accelerated and various exotic micelles are produced. Surprisingly, with high doping levels, the self-assembly mechanism of this block copolymer is completely changed from intramolecular chain shuffling and rearrangement to nucleation-growth mode, based on which self-seeding experiments can be conducted to produce highly uniform fibrils.

3D-Positioning of Nanoparticles in High-Curvature Block Copolymer Domains.

Project description:The defined assembly of nanoparticles in polymer matrices is an important precondition for next-generation functional materials. Here we demonstrate that a defined three-dimensional nanoparticle assembly within the unit cells can be realized by directly linking the nanoparticles to block copolymers. We show that for a range of nearly symmetric to unsymmetric block copolymers there are only two formed structures, a hexagonal lattice of P6/mmm-symmetry, where the nanoparticles are located in 1D-arrays within the cylindrical domains, and a cubic lattice of Im3m-symmetry, where the nanoparticles are located in the octahedral voids of a BCC-lattice, corresponding to the structure of ferrite steel. We observe the block length ratio and thus the interfacial curvature to be the most important parameter determining the lattice type. This is rationalized in terms of minimal chain extension such that domain topologies with large positive curvature are highly preferred. Already volume fractions of only one percent are sufficient to destabilize a lamellar structure and favor the formation of highly curved interfaces. The study thus demonstrates how nanoparticles can be located on well-defined positions in three-dimensional unit cells of block copolymer nanocomposites. This opens the way to functional 3D-nanocomposites where the nanoparticles need to be located on defined matrix positions.

Green Synthesis and the Evaluation of a Functional Amphiphilic Block Copolymer as a Micellar Curcumin Delivery System.

Project description:Polymer micelles represent one of the most attractive drug delivery systems due to their design flexibility based on a variety of macromolecular synthetic methods. The environmentally safe chemistry in which the use or generation of hazardous materials is minimized has an increasing impact on polymer-based drug delivery nanosystems. In this work, a solvent-free green synthetic procedure was applied for the preparation of an amphiphilic diblock copolymer consisting of biodegradable hydrophobic poly(acetylene-functional carbonate) and biocompatible hydrophilic polyethylene glycol (PEG) blocks. The cyclic functional carbonate monomer 5-methyl-5-propargyloxycarbonyl-1,3-dioxane-2-one (MPC) was polymerized in bulk using methoxy PEG-5K as a macroinitiator by applying the metal-free organocatalyzed controlled ring-opening polymerization at a relatively low temperature of 60 °C. The functional amphiphilic block copolymer self-associated in aqueous media into stable micelles with an average diameter of 44 nm. The copolymer micelles were physico-chemically characterized and loaded with the plant-derived anticancer drug curcumin. Preliminary in vitro evaluations indicate that the functional copolymer micelles are non-toxic and promising candidates for further investigation as nanocarriers for biomedical applications.

Silica nanoparticle-loaded thermoresponsive block copolymer vesicles: a new post-polymerization encapsulation strategy and thermally triggered release.

Project description:A thermoresponsive amphiphilic diblock copolymer that can form spheres, worms or vesicles in aqueous media at neutral pH by simply raising the dispersion temperature from 1 °C (spheres) to 25 °C (worms) to 50 °C (vesicles) is prepared via polymerization-induced self-assembly (PISA). Heating such an aqueous copolymer dispersion from 1 °C up to 50 °C in the presence of 19 nm glycerol-functionalized silica nanoparticles enables this remarkable 'shape-shifting' behavior to be exploited as a new post-polymerization encapsulation strategy. The silica-loaded vesicles formed at 50 °C are then crosslinked using a disulfide-based dihydrazide reagent. Such covalent stabilization enables the dispersion to be cooled to room temperature without loss of the vesicle morphology, thus aiding characterization and enabling the loading efficiency to be determined as a function of both copolymer and silica concentration. Small-angle X-ray scattering (SAXS) analysis indicated a mean vesicle membrane thickness of approximately 20 ± 2 nm for the linear vesicles and TEM studies confirmed encapsulation of the silica nanoparticles within these nano-objects. After removal of the non-encapsulated silica nanoparticles via multiple centrifugation-redispersion cycles, thermogravimetric analysis indicated that vesicle loading efficiencies of up to 86% can be achieved under optimized conditions. Thermally-triggered release of the silica nanoparticles is achieved by cleaving the disulfide bonds at 50 °C using tris(2-carboxyethyl)phosphine (TCEP), followed by cooling to 20 °C to induce vesicle dissociation. SAXS is also used to confirm the release of silica nanoparticles by monitoring the disappearance of the structure factor peak arising from silica-silica interactions.

Surface Attachment of Gold Nanoparticles Guided by Block Copolymer Micellar Films and Its Application in Silicon Etching.

Project description:Patterning metallic nanoparticles on substrate surfaces is important in a number of applications. However, it remains challenging to fabricate such patterned nanoparticles with easily controlled structural parameters, including particle sizes and densities, from simple methods. We report on a new route to directly pattern pre-formed gold nanoparticles with different diameters on block copolymer micellar monolayers coated on silicon substrates. Due to the synergetic effect of complexation and electrostatic interactions between the micellar cores and the gold particles, incubating the copolymer-coated silicon in a gold nanoparticles suspension leads to a monolayer of gold particles attached on the coated silicon. The intermediate micellar film was then removed using oxygen plasma treatment, allowing the direct contact of the gold particles with the Si substrate. We further demonstrate that the gold nanoparticles can serve as catalysts for the localized etching of the silicon substrate, resulting in nanoporous Si with a top layer of straight pores.

Cross-Linked Micellar Spherical Nucleic Acids from Thermoresponsive Templates.

Project description:A one-pot synthesis of micellar spherical nucleic acid (SNA) nanostructures using Pluronic F127 as a thermoresponsive template is reported. These novel constructs are synthesized in a chemically straightforward process that involves intercalation of the lipid tails of DNA amphiphiles (CpG motifs for TLR-9 stimulation) into the hydrophobic regions of Pluronic F127 micelles, followed by chemical cross-linking and subsequent removal of non-cross-linked structures. The dense nucleic acid shell of the resulting cross-linked micellar SNA enhances their stability in physiological media and facilitates their rapid cellular internalization, making them effective TLR-9 immunomodulatory agents. These constructs underscore the potential of SNAs in regulating immune response and address the relative lack of stability of noncovalent constructs.
