First Detection of the Adventive Egg Parasitoid of Halyomorpha halys (Stal) (Hemiptera: Pentatomidae) Trissolcus mitsukurii (Ashmead) (Hymenoptera: Scelionidae) in France.

Project description:We report the first detection of Trissolcus mitsukurii in France. More than 1860 sentinel egg masses of Halyomorpha halys (BMSB) were exposed in the field during the 2018-2020 period, and 12 specimens of T. mitsukurii emerged from one egg mass. Their taxonomic identification was confirmed both by morphological and molecular analysis. Trissolcus mitsukurii, similar to T. japonicus, is an egg parasitoid of BMSB in its area of origin in Asia, and both species are considered to be candidates for a classical biological control strategy against BMSB. Trissolcus mitsukurii was previously recorded in Italy where it is well established and widespread, and this may be the source of the French population. Possible permanent establishment and dispersion of T. mitsukurii in France should be monitored with emphasis on its potential effect on BMSB populations.

Field releases of the exotic parasitoid Trissolcus japonicus (Hymenoptera: Scelionidae) and survey of native parasitoids attacking Halyomorpha halys (Hemiptera: Pentatomidae) in Michigan.

Project description:An adventive population of the exotic parasitoid wasp, Trissolcus japonicus (Ashmead) (Hymenoptera: Scelionidae), discovered in Michigan in 2018, is a promising biological control agent of the invasive Halyomorpha halys (Stål) (Hemiptera: Pentatomidae). Following its discovery, field releases of Tr. japonicus were conducted over 2 yr in southern Michigan, to test how release size or release frequency impacts establishment. Sentinel eggs of H. halys and of the native Podisus maculiventris (Say) (Hemiptera: Pentatomidae) were used alongside yellow sticky cards to monitor parasitoids. In 2019 and 2020, 7,200 Tr. japonicus were released at 16 sites. Monitoring between 2019 and 2021 yielded only 49 individuals. The captures suggest reproductive activity and overwintering success in the field but do not allow for evaluation of best release methods. Parasitism by native parasitoids was below 7%, which is similar to other states and unlikely to provide sufficient control of H. halys. The placement of sentinel eggs or sticky traps either in the lower or middle canopy of trees did not influence parasitoid capture rates. Frozen and fresh H. halys sentinel eggs were attacked at the same rate, but more native parasitoids emerged from frozen eggs. We did not find signs of nontarget effects on P. maculiventris thus parasitism rates overall were very low. These results could indicate dispersal of Tr. japonicus from the release sites or slow population growth. The latter may be due to the relatively low densities of H. halys in Michigan or may stem from the small founding size of our laboratory colony.

Surveys in northern Utah for egg parasitoids of Halyomorpha halys (Stal) (Hemiptera: Pentatomidae) detect Trissolcus japonicus (Ashmead) (Hymenoptera: Scelionidae).

Project description:The highly polyphagous and invasive brown marmorated stink bug, Halyomorpha halys (Stål) (Hemiptera: Pentatomidae), has become a significant insect pest in North America since its detection in 1996. It was first documented in northern Utah in 2012 and reports of urban nuisance problems and plant damage have since increased. Biological control is the preferred solution to managing H. halys in North America and other invaded regions due to its alignment with integrated pest management and sustainable practices. Native and non-native biological control agents, namely parasitoid wasps, have been assessed for efficacy. Trissolcus japonicus (Ashmead) (Hymenoptera: Scelionidae) is an effective egg parasitoid of H. halys in its native range of southeast Asia and has recently been documented parasitising H. halys eggs in North America and Europe. Field surveys for native and exotic egg parasitoids using wild (in situ) and lab-reared H. halys egg masses were conducted in suburban and agricultural sites in northern Utah from June to September 2017-2019. Seven native wasp species in the families Eupelmidae and Scelionidae were discovered guarding H. halys eggs and adult wasps from five of these species completed emergence. Native species had low mean rates of adult emergence from wild (0.5-3.7%) and lab-reared (0-0.4%) egg masses. In 2019, an adventive population of T. japonicus was discovered for the first time in Utah, emerging from 21 of the 106 wild H. halys egg masses found that year, and none from lab-reared eggs. All T. japonicus emerged from egg masses collected on Catalpa speciosa (Warder). Our results support other studies that have observed biological control of H. halys from T. japonicus and improved parasitoid wasp detection with wild as compared to lab-reared H. halys egg masses.

The complete mitochondrial genome of Trissolcus japonicus (Hymenoptera: Scelionidae), the candidate for the biological control of Halyomorpha halys (Hemiptera: Pentatomidae).

Project description:The samurai wasp Trissolcus japonicus (Ashmead, 1904) is a parasitoid hymenopteran that came into the limelight as the natural enemy of Halyomorpha halys. Here, we present the complete sequence of the mitochondrial genome of the CREATJ laboratory strain, naturally recovered in Italy in 2018. The molecule conforms to the typical model of animal mitochondrial genomes. Gene order is identical to that of its congeneric Trissolcus basalis. Phylogenetic analysis confirms its placement within monophyletic Scelionidae and Telenominae as the sister group of T. basalis.

Halyomorpha halys (Hemiptera: Pentatomidae) Genetic Diversity in North America and Europe.

Project description:The brown marmorated stink bug, Halyomorpha halys (Hemiptera: Pentatomidae), is an invasive species in North America and Europe that damages many different host plants. Substantial work has been conducted on the genetic diversity and invasion pathways of H. halys in some of the countries where it has been found, based on mitochondrial sequences. The main objective of the present study was to further explore the genetic diversity of invasive populations of H. halys exploiting both mitochondrial and nuclear markers. We used two molecular markers: the mitochondrial Cytochrome Oxidase I (COI) gene, an ideal standardized molecular marker for distinguishing closely related species, and the ribosomal Internal Transcribed Spacer 1 (ITS1), because only a few sequences of H. halys exist to this point in global databases. We used specimens from eight populations from Greece, Italy, Canada, and the US. Among the 14 haplotypes retrieved based on the mtCOI gene, two of them (H162-H163) were detected for the first time. These two haplotypes were found in specimens from Canada, Italy, and the US. Concerning the ITS1 region, 24 haplotypes were identified, with 15 being unique for a sampled population. In Greece and the US, 14 and 12 haplotypes were found, respectively, with 7 and 6 of them being unique for Greece and the US, respectively. Our analysis of the nuclear genes of H. halys indicates high genetic diversity of the invading populations in North America and Europe.

Insights into the saliva of the brown marmorated stink bug Halyomorpha halys (Hemiptera: Pentatomidae).

Project description:We examined the salivary gland structure of the brown marmorated stink bug (Pentatomidae: Halyomorpha halys) and developed methods for independent collection of watery saliva and sheath saliva. This stink bug has become a serious invasive pest of agriculture in the United States and its saliva is largely responsible for the damage it causes. We determined by protein gel analysis and shotgun proteomics that the suite of proteins comprising the sheath and watery saliva are very distinct. Our results indicate that a substantial amount of sheath proteins are derived from tomato when stink bugs feed on tomato fruit. Consequently, the sheath saliva is comprised of both insect and plant-derived proteins. Both sheath and watery saliva possessed amylase activities, but polyphenol oxidase and glucose oxidase activities were not detected in either saliva. Peroxidase activity was only detected in salivary sheaths, but only when stink bugs fed on tomato. Proteomic analysis indicated that the peroxidase was likely of plant origin. We also determined that sheath saliva, but not watery saliva elicited the jasmonate inducible defense gene proteinase inhibitor 2 (Pin2), but this induction was only observed when sheaths had been collected from tomato. This indicates that the eliciting factor of the saliva is likely of plant origin. Lastly, neither watery or sheath saliva affected the expression of the salicylate inducible gene pathogenesis related gene (Pr1a-P4).

Attraction of Halyomorpha halys (Hemiptera: Pentatomidae) haplotypes in North America and Europe to baited traps.

Project description:Halyomorpha halys is a global invasive species, native to Southeast Asia, that is threatening agriculture in invaded regions. Our objectives were to: 1) establish the attractiveness of semiochemical stimuli paired with field-deployed traps in Europe (Greece, Hungary, Italy, and Switzerland), compared with Maryland, USA, and 2) identify H. halys haplotypes recovered from traps at each location. We found qualitatively identical patterns of capture between sites located across Europe and in Maryland, USA. In both regions, captures of H. halys adults indicated a synergistic response to traps baited with the two component H. halys aggregation pheromone, and pheromone synergist, methyl (2E, 4E, 6Z)-decatrienoate when compared with either individually. Haplotype diversity in Europe based on trapped specimens was much greater than the USA, with five new haplotypes described here, probably indicating ongoing invasion and re-introduction of H. halys. By contrast, a single, previously identified haplotype was trapped in Maryland, USA, representing a single introduction. All H. halys haplotypes responded to each semiochemical in apparent proportion to their frequency in the overall population based on independently derived information from prior work. Taken together, these data suggest that pheromone-based technology will be of global utility for the monitoring of this important invasive species.

Monitoring and Biosurveillance Tools for the Brown Marmorated Stink Bug, Halyomorpha halys (Stal) (Hemiptera: Pentatomidae).

Project description:Halyomorpha halys (Stål) (Hemiptera: Pentatomidae) is an invasive pest of numerous agricultural crops with an increasing global distribution. Finding simple and reliable monitoring tools for H. halys agricultural and surveillance programs is imperative. In 2016, we compared standard pyramid traps to clear sticky cards attached atop wooden stakes and evaluated two commercially formulated lures (Trécé and AgBio) with low and high rates of the H. halys aggregation pheromone (PHER) and pheromone synergist (MDT) at 12 sites (low: 5 mg PHER + 50 mg MDT; high: 20 mg PHER + 200 mg MDT). In 2017, we reevaluated lure efficacy using only the clear sticky traps at six locations. Sites were classified as having low, moderate, or high relative population densities of H. halys in 2016, and as very low or low densities of H. halys in 2017. Although clear sticky traps captured fewer adults and nymphs than pyramid traps, their captures were generally correlated at all population levels indicating that clear sticky traps can reliably monitor H. halys presence and relative abundance regardless of relative population density. During both years, adult and nymphal captures were significantly greater in traps baited with Trécé lures than with AgBio lures. Captures were greater in traps baited with high loading rate lures for each lure type, and with the exception of traps baited with AgBio lures at high relative density sites in 2016, H. halys captures in traps with low and high loading rates of each lure type were correlated for both years. Comparison of yellow and clear sticky cards indicated they performed equally, but yellow cards captured more nontargets. In summary, clear sticky traps attached atop wooden posts and baited with H. halys pheromone and pheromone synergist lures are an effective option for this pest monitoring and detection.

Utilizing immunomarking techniques to track Halyomorpha halys (Hemiptera: Pentatomidae) movement and distribution within a peach orchard.

Project description:In this study we focus on the invasive brown marmorated stink bug, Halyomorpha halys (Stål) (Hemiptera: Pentatomidae), which has a strong dispersal capacity and has had a significant impact on several cropping systems, including peach (Prunus persica (L.)). Management of H. halys has relied on intensive insecticide use, and thus a better understanding of its dispersal behavior may assist in developing improved management strategies. In order to investigate H. halys movement and distribution patterns within a peach orchard we applied ecologically safe, food protein markers to the trees along the orchard border (chicken egg albumin in the form of liquid egg whites) and to the trees within the orchard interior (bovine casein in the form of cow's milk). We used enzyme-linked immunosorbent assays (ELISA) to assess whether collected H. halys were "marked" with either of the two protein markers, revealing where in the orchard the bugs had visited. From the density data we determined that H. halys is a perimeter-driven pest in peaches, with a significantly higher density of bugs collected along the orchard border. Interestingly, this trend is primarily driven by the distribution of male bugs. The protein marking data revealed that a small proportion of male H. halys move equally between the orchard border and interior, while a small proportion of females move predominately to the border after visiting the interior. The verification of a strong edge-effect, although potentially sex-specific, implies that H. halys displays a dispersal behavior that may also be exploited for management, which may help growers more efficiently and more effectively manage H. halys.
