Combined application of biochar and PGPR consortia for sustainable production of wheat under semiarid conditions with a reduced dose of synthetic fertilizer.

ABSTRACT: This study investigates the combined effect of locally adopted plant growth promoting rhizobacteria (PGPR), biochar, and synthetic fertilizer on the wheat crop for the production and economic returns. A total of 20 PGPR strains were isolated from three different ecological zones of Pakistan and were evaluated. Of them, three isolates were selected for further studies. The treatments included (i) control with a full dose of the recommended fertilizer, (ii) control with half a dose of the fertilizer, (iii) PGPR consortia with half a dose of the fertilizer, (iv) biochar with half a dose of the fertilizer, and (v) PGPR + biochar with half a dose of the fertilizer. The study was repeated at three different locations. The data collected for leaf area index (LAI), grain yield, biological yield, straw yield, and harvest index (HI) revealed significant differences (P ≤ 0.05) for the locations and treatments, but the interaction of location and treatments was not significant. Based on the productivity and economic returns, the treatment with PGPR + biochar with half a dose of the fertilizer proved to be the best. Thus, the use of the PGPR consortia and biochar can improve the yield and profit of wheat crop with reduced synthetic fertilization. Graphical abstract ᅟ.

Project description:Phosphorous (P) is a limiting macronutrient for crop growth. Its deficiency prevents plant development leading to an extensive use of phosphatic fertilizers globally. Bio-organic phosphate (BOP) fertilizer provides a sustainable approach to optimize nutrient availability, enhance crop yield, and mitigate the negative impacts of chemical fertilizers on the environment. Therefore, the present study integrates the application of heat-tolerant phosphate-solubilizing bacteria, rock phosphate, and organic materials for the development of BOP. For this purpose, potential heat-tolerant phosphate-solubilizing bacteria (PSB) were isolated from major wheat-growing areas of southern Punjab. Five isolates were the efficient phosphate solubilizers based on in vitro phosphate-solubilizing activity (291-454 μg ml-1 and 278-421 μg ml-1) with a concomitant decrease in pH (up to 4.5) at 45°C and 50°C, respectively. These PSB were used for the development of potential consortia that are compatible and showed high P solubilization. In planta evaluation of these PSB consortia in a pot experiment under net house conditions showed that consortium-2 had a favorable impact on growth parameter with enhanced grain yield (9.63 g plant-1) and soil available P (10 μg g-1) as compared with 80% uninoculated control. The microcosm study was conducted to evaluate PSB consortium-2 integrated with carrier material (plant material and filter mud) and rock phosphate as BOP increased total phosphorous (14%) as compared with uninoculated controls. Plant-based BOP showed higher viable count (3.5 × 108\u00B0CFU) as compared with filter mud-based BOP. Furthermore, the effect of BOP on wheat growth parameters revealed that BOP showed a promising influence on grain yield (4.5 g plant-1) and soil available P (10.7 μg g-1) as compared with uninoculated 80 and 100% controls. Principle component analysis (PCA) further validates a positive correlation between BOP with grain weight and plant height and soil available P as compared with both 80 and 100% controls. For the first time, this study reports the combined application of bio-organic phosphate fertilizer and heat-tolerant PSB, which offers an eco-friendly option to harvest better wheat yield with low fertilizer input.

Project description:BackgroundSoil application of biochar and straw alone or their combinations with nitrogen (N) fertilizer are becoming increasingly common, but little is known about their agronomic and environmental performance in semiarid environments. This study was conducted to investigate the effect(s) of these amendments on soil properties, nitrous oxide (N2O) and methane (CH4) emissions and grain and biomass yield of spring wheat (Triticum aestivum L.), and to produce background dataset that may be used to inform nutrient management guidelines for semiarid environments.MethodsThe experiment involved the application of biochar, straw or urea (46% nitrogen [N]) alone or their combinations. The treatments were: CN0-control (zero-amendment), CN50 -50 kg ha-1 N, CN100-100 kg ha-1 N, BN0 -15 t ha-1 biochar, BN50-15 t ha-1 biochar + 50 kg ha-1 N, BN100-15 t ha-1 biochar + 100 kg ha-1 N, SN0 -4.5 t ha-1 straw, SN50 -4.5 t ha-1 straw + 50 kg ha-1 N and SN100-4.5 t ha-1 straw + 100 kg ha-1 N. Fluxes of N2O, CH4 and grain yield were monitored over three consecutive cropping seasons between 2014 and 2016 using the static chamber-gas chromatography method.ResultsOn average, BN100reported the highest grain yield (2054 kg ha-1), which was between 25.04% and 38.34% higher than all other treatments. In addition, biomass yield was much higher under biochar treated plots relative to the other treatments. These findings are supported by the increased in soil organic C by 17.14% and 21.65% in biochar amended soils (at 0-10 cm) compared to straw treated soils and soils without carbon respectively. The BN100treatment also improved bulk density and hydraulic properties (P < 0.05), which supported the above results. The greatest N2O emissions and CH4 sink were recorded under the highest rate of N fertilization (100 kg N ha-1). Cumulative N2O emissions were 39.02% and 48.23% lower in BN100 compared with CN0 and CN100, respectively. There was also a ≈ 37.53% reduction in CH4 uptake under BN100compared with CN0-control and CN50. The mean cumulative N2O emission from biochar treated soils had a significant decrease of 10.93% and 38.61% compared to straw treated soils and soils without carbon treatment, respectively. However, differences between mean cumulative N2O emission between straw treated soils and soils without carbon were not significant. These results indicate the dependency of crop yield, N2O and CH4 emissions on soil quality and imply that crop productivity could be increased without compromising on environmental quality when biochar is applied in combination with N-fertilizer. The practice of applying biochar with N fertilizer at 100 kg ha-1 N resulted in increases in crop productivity and reduced N2O and CH4soil emissions under dryland cropping systems.

Project description:Soil salinity has emerged as a major obstacle to meet world food demands. Halo-tolerant plant growth promoting rhizobacteria (PGPR) are potential bioinoculants to enhance crop productivity in saline agriculture. Current work was aimed at studying individual or synergetic impact of salt tolerant PGPR on wheat growth and yield under saline conditions. A pot experiment was conducted on two wheat genotypes (Aas-11; salt tolerant and Galaxy-13; salt sensitive) inoculated with Pseudomonas fluorescence, Bacillus pumilus, and Exiguobacterium aurantiacum alone and in consortium. The salt tolerant variety (Aas-11) exhibited maximum root fresh (665.2%) and dry biomass (865%), free proline (138.12%) and total soluble proteins (155.9%) contents, CAT (41.7%) activity and shoot potassium uptake (81.08%) upon inoculation with B. pumilus, while improved shoot dry weight (70.39%), water (23.49%) and osmotic (29.65%) potential, POD (60.51%) activity, enhanced root potassium (286.36%) and shoot calcium (400%) were manifested by E. aurantiacum. Highest shoot length (14.38%), fresh weight (72.73%), potassium (29.7%) and calcium (400%) acquisition as well as glycinebetaine (270.31%) content were found in plants treated with PGPR consortium. On the other hand, in the salt sensitive variety (Galaxy-13), P. fluorescens treated plants showed significantly improved leaf-water relations, glycinebetaine (10.78%) content, shoot potassium (23.07%), root calcium (50%) uptake, and yield parameters, respectively. Plant root length (71.72%) and potassium content (113.39%), root and shoot fresh and dry biomass, turgor potential (231.02%) and free proline (317.2%) content were maximum upon PGPR inoculation in consortium. Overall, Aas-11 (salt tolerant variety) showed significantly better performance than Galaxy-13 (salt sensitive variety). This study recommends B. pumilus and E. aurantiacum for the salt tolerant (Aas-11) and P. fluorescens for the salt sensitive (Galaxy-13) varieties, as potential bioinoculants to augment their growth and yield through modulation of morpho-physiological and biochemical attributes under saline conditions.
