A Simple and Efficient Device and Method for Measuring the Kinetics of Gas-Producing Reactions.

ABSTRACT: We present a new device for quantifying gases or gas mixtures based on the simple principle of bubble counting. With this device, we can follow reaction kinetics down to volume step sizes of 8-12 μL. This enables the accurate determination of both time and size of these gas quanta, giving a very detailed kinetic analysis. We demonstrate this method and device using ammonia borane hydrolysis as a model reaction, obtaining Arrhenius plots with over 300 data points from a single experiment. Our device not only saves time and avoids frustration, but also offers more insight into reaction kinetics and mechanistic studies. Moreover, its simplicity and low cost open opportunities for many lab applications.

A simple method for measuring organotypic tissue slice culture thickness.

Project description:This paper presents a simple method to measure tissue slice thicknesses using an ohmmeter. The circuit described here is composed of a metal probe, an ohmmeter, a counter electrode, culture medium or physiological buffer, and tissue slice. The probe and the electrode are on opposite interfaces of an organotypic hippocampal slice culture. The circuit closes when the metal probe makes contact with the surface of the tissue slice. The probe position is recorded and compared to its position when it makes contact with the insert membrane on which the tissue grows, thus yielding a thickness measurement. The method does not reduce the viability of slice cultures. Thicknesses of the slice cultures were measured under a number of culturing protocols. An initial drop in thickness occurred between 0 and 4 days in culture. Thicknesses are rather constant thereafter. The type of culture medium and the initial thickness of the tissue explant influence the thickness. Slice thicknesses were compared to a known technique by using optical measurements of slice cross-sections to obtain thicknesses. In contrast to this known technique, the proposed method does not sacrifice the slice culture for measurement purposes. The proposed measurement technique described is straightforward and rapid, about 1 min per culture.

We report here the development of a simple biocompatible, bioluminescent method for measuring DAO activity based on the highly selective condensation of D-Cys with 6-hydroxy-2-cyanobenzothiazole (CBT-OH) to form D-luciferin.

A simple method to measure methane emissions from indoor gas leaks.

Project description:From wellhead to burner tip, each component of the natural gas process chain has come under increased scrutiny for the presence and magnitude of methane leaks, because of the large global warming potential of methane. Top-down measures of methane emissions in urban areas are significantly greater than bottom-up estimates. Recent research suggests this disparity might in part be explained by gas leaks from one of the least understood parts of the process chain: behind the gas meter in homes and buildings. However, little research has been performed in this area and few methods and data sets exist to measure or estimate them. We develop and test a simple and widely deployable closed chamber method that can be used for quantifying indoor methane emissions with an order-of-magnitude precision which allows for screening of indoor large volume ("super-emitting") leaks. We also perform test applications of the method finding indoor leaks in 90% of the 20 Greater Boston buildings studied and indoor methane emissions between 0.02-0.51 ft3 CH4 day-1 (0.4-10.3 g CH4 day-1) with a mean of 0.14 ft3 CH4 day-1 (2.8 g CH4 day-1). Our method provides a relatively simple way to scale up indoor methane emissions data collection. Increased data may reduce uncertainty in bottom-up inventories, and can be used to find super-emitting indoor emissions which may better explain the disparity between top-down and bottom-up post-meter emissions estimates.

A simple and dynamic thermal gradient device for measuring thermal performance in small ectotherms.

Project description:The body temperature of ectothermic animals is heavily dependent on environmental temperature, impacting fitness. Laboratory exposure to favorable and unfavorable temperatures is used to understand these effects, as well as the physiological, biochemical, and molecular underpinnings of variation in thermal performance. Although small ectotherms, like insects, can often be easily reared in large numbers, it can be challenging and expensive to simultaneously create and manipulate several thermal environments in a laboratory setting. Here, we describe the creation and use of a thermal gradient device that can produce a wide range of constant or varying temperatures concurrently. Conservatively, this system as designed can operate between -6 °C and 40 °C. This device is composed of a solid aluminum plate and copper piping, combined with a pair of refrigerated circulators. As a simple proof-of-concept, we completed single experimental runs to produce a low-temperature survival curve for flies (Drosophila melanogaster) and explore the effects of daily thermal cycles of varying amplitude on growth rates of crickets (Gryllodes sigillatus). This approach avoids the use of multiple heating/cooling water or glycol baths or incubators for large-scale assessments of organismal thermal performance. It makes static or dynamic thermal experiments (e.g., creating a thermal performance or survival curves, quantifying responses to fluctuating thermal environments, or monitoring animal behavior across a range of temperatures) easier, faster, and less costly.

Highly Efficient Method for Suzuki Reactions in Aqueous Media.

Project description:Herein, we report the crystal structure and characterization of mono-6-(l-aminopropanol)-deoxy-β-cyclodextrin (L n @β-CD). A highly efficient, in situ generated catalyst, PdCl2(L n @β-CD), was synthesized for palladium-catalyzed cross-coupling reactions under mild reaction conditions, and the use of this catalyst in Suzuki cross-couplings was investigated. Low palladium loadings of 0.01 mol % PdCl2(L n @β-CD) (Pd accounted for approximately 8.4% of the catalyst by mass) were found to be highly efficient for Suzuki cross-couplings in water and afforded the corresponding biaryl compounds in excellent yields. The catalyst can be recycled and reused.

A multi-objective stacked regression method for distance based colour measuring device.

Project description:Identifying colour from a distance is challenging due to the external noise associated with the measurement process. The present study focuses on developing a colour measuring system and a novel Multi-target Regression (MTR) model for accurate colour measurement from distance. Herein, a novel MTR method, referred as Multi-Objective Stacked Regression (MOSR) is proposed. The core idea behind MOSR is based on stacking as an ensemble approach with multi-objective evolutionary learning using NSGA-II. A multi-objective optimization approach is used for selecting base learners that maximises prediction accuracy while minimising ensemble complexity, which is further compared with six state-of-the-art methods over the colour dataset. Classification and regression tree (CART), Random Forest (RF) and Support Vector Machine (SVM) were used as regressor algorithms. MOSR outperformed all compared methods with the highest coefficient of determination values for all three targets of the colour dataset. Rigorous comparison with state-of-the-art methods over 18 benchmarked datasets showed MOSR outperformed in 15 datasets when CART was used as a regressor algorithm and 11 datasets when RF and SVM were used as regressor algorithms. The MOSR method was statistically superior to compared methods and can be effectively used to measure accurate colour values in the distance-based colour measuring device.

A simple and efficient method to enhance audiovisual binding tendencies.

Project description:Individuals vary in their tendency to bind signals from multiple senses. For the same set of sights and sounds, one individual may frequently integrate multisensory signals and experience a unified percept, whereas another individual may rarely bind them and often experience two distinct sensations. Thus, while this binding/integration tendency is specific to each individual, it is not clear how plastic this tendency is in adulthood, and how sensory experiences may cause it to change. Here, we conducted an exploratory investigation which provides evidence that (1) the brain's tendency to bind in spatial perception is plastic, (2) that it can change following brief exposure to simple audiovisual stimuli, and (3) that exposure to temporally synchronous, spatially discrepant stimuli provides the most effective method to modify it. These results can inform current theories about how the brain updates its internal model of the surrounding sensory world, as well as future investigations seeking to increase integration tendencies.

Enhanced condensation kinetics in aqueous microdroplets driven by coupled surface reactions and gas-phase partitioning.

Project description:Although aqueous microdroplets have been shown to exhibit enhanced chemical reactivity compared to bulk solutions, mechanisms for these enhancements are not completely understood. Here we combine experimental measurements and kinetic modeling to show the strong coupling of interfacial reactions and gas/droplet partitioning in the condensation reaction of pyruvic acid (PA) to yield zymonic acid (ZA) in acidic aqueous microdroplets. Experimental analysis of single microdroplets reveals the substantial influence of evaporation of PA and partitioning of water on the size-, relative humidity (RH)- and temperature-dependent sigmoidal reaction kinetics for the condensation reaction. A newly developed diffusion-reaction-partitioning model is used to simulate the complex kinetics observed in the microdroplets. The model can quantitatively predict the size and compositional changes as the reaction proceeds under different environmental conditions, and provides insights into how microdroplet reactivity is controlled by coupled interfacial reactions and the gas-phase partitioning of PA and water. Importantly, the kinetic model best fits the data when an autocatalytic step is included in the mechanism, i.e. a reaction step where the product, ZA, catalyzes the interfacial condensation reaction. Overall, the dynamic nature of aqueous microdroplet chemistry and the coupling of interfacial chemistry with gas-phase partitioning are demonstrated. Furthermore, autocatalysis of small organic molecules at the air-water interface for aqueous microdroplets, shown here for the first time, has implications for several fields including prebiotic chemistry, atmospheric chemistry and chemical synthesis.

A simple and soft chemical deaggregation method producing single-digit detonation nanodiamonds.

Project description:Detonation nanodiamonds (DNDs) are a class of very small and spherical diamond nanocrystals. They are used in polymer reinforcement materials or as drug delivery systems in the field of nanomedicine. Synthesized by detonation, only the final deaggregation step down to the single-digit nanometer size (<10 nm) unfolds their full potential. Existing deaggregation methods mainly rely on mechanical forces, such as high-power sonication or bead milling. These techniques entail drawbacks such as contamination of the sample and the need for a specialized apparatus. In this paper, we report a purely chemical deaggregation method by simply combining oxidation in air followed by a boiling acid treatment, to produce highly stable single-digit DNDs in a suspension. The resulting DNDs are surface functionalized with carboxyl groups, the final boiling acid treatment removes primary metal contaminants such as magnesium, iron or copper and the nanoparticles remain dispersed over a wide pH range. Our method can be easily carried out in a standard chemistry laboratory with commonly available laboratory apparatus. This is a key step for many DND-based applications, ranging from materials science to biological or medical applications.

A label-free real-time method for measuring glucose uptake kinetics in yeast.

Project description:Glucose uptake assays commonly rely on the isotope-labeled sugar, which is associated with radioactive waste and exposure of the experimenter to radiation. Here, we show that the rapid decrease of the cytosolic pH after a glucose pulse to starved Saccharomyces cerevisiae cells is dependent on the rate of sugar uptake and can be used to determine the kinetic parameters of sugar transporters. The pH-sensitive green fluorescent protein variant pHluorin is employed as a genetically encoded biosensor to measure the rate of acidification as a proxy of transport velocity in real time. The measurements are performed in the hexose transporter-deficient (hxt0) strain EBY.VW4000 that has been previously used to characterize a plethora of sugar transporters from various organisms. Therefore, this method provides an isotope-free, fluorometric approach for kinetic characterization of hexose transporters in a well-established yeast expression system.
