A Comparative Analysis of Floral Scent Compounds in Intraspecific Cultivars of Prunus mume with Different Corolla Colours.

ABSTRACT: Prunus mume is the only fragrant flowering species of Prunus. According to the previous studies, benzyl acetate and eugenol dominate its floral scent. However, the diversity of its floral scents remains to be elucidated. In this work, the floral volatiles emitted from eight intraspecific cultivars of P. mume with white, pink and red flowers, were collected and analyzed using headspace solid-phase microextraction combined with gas chromatograms-mass spectrometry (HS-SPME-GC-MS). In total, 31 volatile compounds were identified, in which phenylpropanoids/benzenoids accounted for over 95% of the total emission amounts. Surprisingly, except for benzyl acetate and eugenol, several novel components, such as benzyl alcohol, cinnamyl acohol, cinnamy acetate, and benzyl benzoate were found in some cultivars. The composition of floral volatiles in cultivars with white flowers was similar, in which benzyl acetate was dominant, while within pink flowers, there were differences of floral volatile compositions. Principal component analysis (PCA) showed that the emissions of benzyl alcohol, cinnamyl alcohol, benzyl acetate, eugenol, cinnamyl acetate, and benzyl benzoate could make these intraspecific cultivars distinguishable from each other. Further, hierarchical cluster analysis indicated that cultivars with similar a category and amount of floral compounds were grouped together. Our findings lay a theoretical basis for fragrant plant breeding in P. mume.

Project description:Background and aimsAmong the various floral traits involved in pollinator attraction and potentially under selection mediated by pollinators, floral scent/fragrance has been less investigated than other components of floral phenotype. Whether or not pollinator-mediated selection impacts floral scents depends on the heritability of scent/fragrance and the occurrence of some variation within species. Although most studies have investigated how scent varies among species, growing amounts of data are available on variation at the intraspecific level.MethodsThe results of 81 studies investigating intraspecific variation of floral scents in 132 taxa were reviewed. For each study, whether variation was found in either identity, proportion or absolute quantities of volatile organic compounds (VOCs) was recorded, as well as information with the potential to explain variation, such as methodology, plant origin or pollination biology.Key resultsVariation was found for almost all investigated species, both among individuals (among and sometimes within populations) and within individuals across different temporal scales. Cases in which such variation is a possible result of pollinator-mediated selection were analysed, by discussing separately selection related to variation in pollinator identity/behaviour among populations or across time, deceit pollination and sex-specific selection. Not surprisingly, in many cases, pollinator-mediated selection alone does not explain the observed variation in floral scent. This led us to review current knowledge on less investigated factors, such as selection mediated by natural enemies, genetic drift and gene flow, environmental constraints, phylogenetic inertia, or biochemical constraints that could be invoked to explain scent variation.ConclusionsThis review highlights the great potential of analysing floral scent variation and including it in integrated studies of floral phenotypes. We also have identified the current gaps in our understanding of this complex signal and we propose several methodological and conceptual future directions in this research area.

Project description:Bud dormancy is a critical developmental process for perennial plant survival, and also an important physiological phase that affects the next season’s growth of temperate fruit trees. Bud dormancy is regulated by multiple genetic factors, and affected by various environmental factors, tree age and vigor. To understand molecular mechanism of bud dormancy in Japanese apricot (Prunus mume Sieb. et Zucc.), we constructed a custom oligo DNA microarray covering the Japanese apricot dormant bud ESTs referring to peach (P. persica) genome sequence. Because endodormancy release is a chilling temperature-dependent physiological event, genes showing chilling-mediated differential expression patterns are candidates to control endodormancy release. Using the microarray constructed in this study, we monitored gene expression changes of dormant vegetative buds of Japanese apricot during prolonged artificial chilling exposure. In addition, we analyzed seasonal gene expression changes. ‘Nanko’ vegetative buds collected in November, and those exposed to chilling for 40 or 60 days were used as microarray samples. Among the 58539 different unigene probes, 2345 and 1059 genes were identified as being more than two-fold up-regulated and down-regulated, respectively, following chilling exposure for 60 days (P value < 0.05). The down-regulated genes included P. mume DORMANCY-ASSOCIATED MADS-box genes, which supported the previous quantitative RT-PCR and EST analyses showing that these genes are repressed by prolonged chilling treatments. The genes encoding lipoxygenase were remarkably up-regulated by prolonged chilling. Cluster analysis suggested that the expression of the genes showing expression changes by artificial chilling exposure were coordinately regulated by seasonal changes. Our parametric analysis of gene set enrichment suggested that genes related to jasmonic acid (JA) and oxylipin biosynthesis and metabolic processes were significantly up-regulated by prolonged chilling, whereas genes related to circadian rhythm were significantly down-regulated. The results obtained from the microarray analyses were verified by quantitative RT-PCR analysis of selected genes. Taken together, this study raised the possibility that the microarray platform constructed in this study is applicable for deeper understanding of molecular network related to agronomically important bud phisiologies including dormancy release.
