An RNAi supplemented diet as a reverse genetics tool to control bluegreen aphid, a major pest of legumes.

ABSTRACT: Aphids are important agricultural pests causing major yield losses worldwide. Since aphids can rapidly develop resistance to chemical insecticides there is an urgent need to find alternative aphid pest management strategies. Despite the economic importance of bluegreen aphid (Acyrthosiphon kondoi), very few genetic resources are available to expand our current understanding and help find viable control solutions. An artificial diet is a desirable non-invasive tool to enable the functional characterisation of genes in bluegreen aphid and discover candidate target genes for future use in RNA interference (RNAi) mediated crop protection against aphids. To date no artificial diet has been developed for bluegreen aphid, so we set out to develop a suitable diet by testing and optimising existing diets. Here, we describe an artificial diet for rearing bluegreen aphid and also provide a proof of concept for the supplementation of the diet with RNAi molecules targeting the salivary gland transcript C002 and gap gene hunchback, resulting in bluegreen aphid mortality which has not yet been documented in this species. Managing this pest, for example via RNAi delivery through artificial feeding will be a major improvement to test bluegreen aphid candidate target genes for future pest control and gain significant insights into bluegreen aphid gene function.

Project description:Understanding the spatiotemporal dynamics of pesticide resistance at the landscape scale is essential to anticipate the evolution and spread of new resistance phenotypes. In crop mosaics, host plant specialization in pest populations is likely to dampen the spread of pesticide resistance between different crops even in mobile pests such as aphids. Here, we assessed the contribution of host-based genetic differentiation to the dynamics of resistance alleles in Myzus persicae, a major aphid pest which displays several insecticide resistance mechanisms. We obtained a representative sample of aphids from a crop mosaic through a suction trap for 7 years and from various crops as a reference collection. We genotyped these aphids at 14 microsatellite markers and four insecticide-resistant loci, analyzed the genetic structure, and assigned host-based genetic groups from field-collected aphids. Four well-defined genetic clusters were found in aerial samples, three of which with strong association with host-plants. The fourth group was exclusive to aerial samples and highly divergent from the others, suggesting mixture with a closely related taxon of M. persicae associated with unsampled plants. We found a sharp differentiation between individuals from peach and herbaceous plants. Individuals from herbaceous hosts were separated into two genetic clusters, one more strongly associated with tobacco. The 4-loci resistance genotypes showed a strong association with the four genetic clusters, indicative of barriers to the spread of resistance. However, we found a small number of clones with resistant alleles on multiple host-plant species, which may spread insecticide resistance between crops. The 7-year survey revealed a rapid turn-over of aphid genotypes as well as the emergence, frequency increase and persistence of clones with resistance to several families of insecticides. This study highlights the importance of considering landscape-scale population structure to identify the risk of emergence and spread of insecticide resistance for a particular crop.

Project description:BackgroundThe Russian wheat aphid, Diuraphis noxia Kurdjumov, is one of the most important pests of small grains throughout the temperate regions of the world. This phytotoxic aphid causes severe systemic damage symptoms in wheat, barley, and other small grains as a direct result of the salivary proteins it injects into the plant while feeding.ResultsWe sequenced and de novo assembled the genome of D. noxia Biotype 2, the strain most virulent to resistance genes in wheat. The assembled genomic scaffolds span 393 MB, equivalent to 93% of its 421 MB genome, and contains 19,097 genes. D. noxia has the most AT-rich insect genome sequenced to date (70.9%), with a bimodal CpG(O/E) distribution and a complete set of methylation related genes. The D. noxia genome displays a widespread, extensive reduction in the number of genes per ortholog group, including defensive, detoxification, chemosensory, and sugar transporter groups in comparison to the Acyrthosiphon pisum genome, including a 65% reduction in chemoreceptor genes. Thirty of 34 known D. noxia salivary genes were found in this assembly. These genes exhibited less homology with those salivary genes commonly expressed in insect saliva, such as glucose dehydrogenase and trehalase, yet greater conservation among genes that are expressed in D. noxia saliva but not detected in the saliva of other insects. Genes involved in insecticide activity and endosymbiont-derived genes were also found, as well as genes involved in virus transmission, although D. noxia is not a viral vector.ConclusionsThis genome is the second sequenced aphid genome, and the first of a phytotoxic insect. D. noxia's reduced gene content of may reflect the influence of phytotoxic feeding in shaping the D. noxia genome, and in turn in broadening its host range. The presence of methylation-related genes, including cytosine methylation, is consistent with other parthenogenetic and polyphenic insects. The D. noxia genome will provide an important contrast to the A. pisum genome and advance functional and comparative genomics of insects and other organisms.
