Rheological and NMR spectral data sets of quaternary ammonium derivative of Cassia tora Linn. seed gum.

ABSTRACT: The present study has been carried out to investigate the rheological and spectral data of galactomannan and its quaternary ammonium derivative obtained from the endosperm of Cassia tora Linn. seed. The characterization of native and quaternized Cassia tora gum (CTG) was done by employing 2D NMR spectroscopy including HSQC and HMBC spectra. The data was analysed to identify the quaternary ammonium moiety introduced onto the galactomannan chain. The correlation signal of carbon and hydrogen of quaternary ammonium groups introduced onto the galactomannan chain was observed at 56 and 2.7 ppm respectively in HSQC spectrum. Further, the rheological data of CTG and derivatized product has been discussed. The data is related to the research article "Synthesis and characterization of quaternized Cassia tora gum using Taguchi's L'16 approach" [1].

Sequencing and phylogenetic analysis of the complete chloroplast genome of Cassia tora Linn.

Project description:Cassia tora Linn. is widely distributed in South-East Asia and South-West Pacific as an important weed. It has many pharmacological activities including anti-allergic, anti-hepatotoxic, and remedy in skin diseases. In this study, we assembled and characterized the complete chloroplast genome sequence of C. tora from high-throughput sequencing data. The chloroplast genome was 162,426 bp in length, consisting of large single-copy (LSC) and small single-copy (SSC) regions of 90,843 bp and 18,001 bp, respectively, which were separated by a pair of 26,791 bp inverted repeat (IR) regions. The genome is predicted to contain 131 genes, including 84 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. The overall GC content of the genome is 36.0%. A phylogenetic tree reconstructed by 32 chloroplast genomes reveals that C. tora is mostly related to Senna occidentalis. The work reported the firstly complete chloroplast genome of C. tora which may provide useful information to the evolution of Cassieae Bronn.

Flexible Ternary Combination of Gellan Gum, Sodium Carboxymethyl Cellulose, and Silicon Dioxide Nanocomposites Fabricated by Quaternary Ammonium Silane: Rheological, Thermal, and Antimicrobial Properties.

Project description:Gellan gum-sodium carboxymethyl cellulose (GC)-based composite films with various concentrations of silicon dioxide (SiO2) nanoparticles and octadecyldimethyl-(3-triethoxy silylpropyl)ammonium chloride (ODDMAC) were successfully prepared by the traditional solution casting method to improve the antimicrobial and water repellent properties. Fourier transform infrared (FT-IR) spectra confirm the formation of hydrogen bonds between the GC and nano-SiO2. The microstructure and physicochemical properties were investigated by FT-IR, wide-angle X-ray diffraction, and scanning electron microscopy (SEM) analyses. The rheological properties of the GC-SiO2 hydrogel were also characterized. The results show that the inclusion of SiO2 nanoparticles significantly improved the viscosity and viscoelastic properties of the GC hydrogel. The GC-SiO2 hydrogel exhibited shear-thinning behavior and its viscosity decreased at high shear rates. The storage and loss moduli of the GC composites increased as the frequency and SiO2 concentration increased. The tensile strength and elongation at break of the GC composites increased by 75.9 and 62%, respectively, with the addition of SiO2 and ODDMAC. In addition, nano-SiO2 decreased the water vapor permeability and increased the hydrophobic properties of the GC-SiO2 composites. Thermogravimetric analysis showed that the T 5% loss was in the range of 99.4-128.6 °C and the char yield was in the range of 20.1-29.9%, which was significantly enhanced by the incorporation of SiO2 nanoparticles. The GC-SiO2 (ODDMAC) nanocomposites effectively shielded the UV light and exhibited high antimicrobial activity against six different pathogens. The simple and cost-effective GC-SiO2 (ODDMAC) nanocomposites gained importance in food packaging and biomedical applications.

Anticancer activity and toxicity of new quaternary ammonium geldanamycin derivative salts and their mixtures with potentiators.

Project description:Geldanamycin (GDM) has been modified by different type neutral/acidic/basic substituents (1-7) and by quinuclidine motif (8), transformed into ammonium salts (9-13) at C(17). These compounds have been characterised by spectroscopic and x-ray methods. Derivative 8 shows better potency than GDM in MCF-7, MDA-MB-231, A549 and HeLa (IC50s = 0.09-1.06 µM). Transformation of 8 into salts 9-13 reduces toxicity (by 11-fold) at attractive potency, e.g. MCF-7 cell line (IC50∼2 µM). Our studies show that higher water solubility contributes to lower toxicity of salts than GDM in healthy CCD39Lu and HDF cells. The use of 13 mixtures with potentiators PEI and DOX enhanced anticancer effects from IC50∼2 µM to IC50∼0.5 µM in SKBR-3, SKOV-3, and PC-3 cancer cells, relative to 13. Docking studies showed that complexes between quinuclidine-bearing 8-13 and Hsp90 are stabilised by extra hydrophobic interactions between the C(17)-arms and K58 or Y61 of Hsp90.

Population authentication of the traditional medicinal plant Cassia tora L. based on ISSR markers and FTIR analysis.

Project description:Cassia tora is a plant of medicinal importance. Medicinal plants from different localities are believed to differ in their therapeutic potency. In this study, six populations of C. tora with different eco-geographical origins were investigated genotypically (ISSR) and phytochemically (FTIR) to establish an integrated approach for population discrimination and authentication of the origin of this medicinal herb. CHS gene expression analysis and determination of flavonoid content were carried out to substantiate the study. A total of 19 population-specific authentication bands were observed in 11 ISSR fingerprints. Authentication codes were generated using six highly polymorphic bands, including three authentication bands. FTIR spectra revealed that the peaks at wavenumber 1623 cm-1 (carbonyl group) and 1034 cm-1 (>CO- group) were powerful in separating the populations. These peaks are assigned to flavonoids and carbohydrates, respectively, were more intense for Ranchi (highland) population. Variation in the transcript level of CHS gene was observed. The findings of FTIR and RT-PCR analyses were in agreement with the TFC analysis, where, the lowest amount of flavonoids observed for Lucknow (lowland) population. All the populations of C. tora have been authenticated accurately by ISSR analyses and FTIR fingerprinting, and the Ranchi site was observed to be more suitable for the potential harvesting of therapeutic bioactive compounds.

Rheological behaviours of guar gum derivatives with hydrophobic unsaturated long-chains.

Project description:Three guar gum derivatives with hydrophobic unsaturated long-chains were successfully synthesized, including oleic guar gum (OGG), linoleic guar gum (LGG) and erucic guar gum (EGG). The guar gum derivatives were characterized by Fourier transform infrared (FTIR) spectroscopy and thermogravimetric analysis (TGA). Moreover, comparative studies on the rheological behaviours of guar gum (GG) and three guar gum derivatives were performed. Regardless of their hydrophobic side groups, all sample solutions showed the properties of non-Newtonian shear-thinning fluids, and their complex viscosities (η*) increased significantly with increasing concentration. In contrast to GG, OGG or EGG, LGG exhibited the strongest shear-thinning property when the concentration is 1 wt%. It was found that the frequency-dependent viscous (G'') and elastic modulus (G') curves of GG and its derivatives aqueous solutions cross at a specific frequency. This crossover frequency decreases with increasing sample concentration, indicating that the viscoelasticity is highly dependent on polymer concentration. In addition, when the molar substitution (MS) of the repeating sugar units in the guar gum derivatives is less than 0.08, the LGG solution showed more obvious solid-like rheological properties than the OGG and EGG solutions at the same concentration.

Rheological Response of Polylactic Acid Dispersions in Water with Xanthan Gum.

Project description:In this work, the rheological behavior of stable poly(lactic acid) (PLA) dispersions in water, intended for coating applications, was investigated. The newly prepared dispersion consists of PLA particles with an average diameter of 222 ± 2 nm based on dynamic light scattering (DLS) and scanning electron microscopy (SEM) analyses, at concentrations varying in the 5-22 wt % range. Xanthan gum (XG), a bacterial polysaccharide, was used as a thickening agent to modulate the viscosity of the formulations. The rheological properties of the PLA dispersions with different XG and PLA contents were studied in steady shear, amplitude sweep, and frequency sweep experiments. Under steady shear conditions, the viscosity of all the formulations showed a shear-thinning behavior similar to XG solutions in the whole investigated 1-1000 s-1 range, with values dependent on both PLA particles and XG concentrations. Amplitude and frequency sweep data revealed a weak-gel behavior except in the case of the most diluted sample, with moduli dependent on both PLA and XG contents. A unified scaling parameter was identified in the volume fraction (ϕ) of the PLA particles, calculated by considering the dependence of the continuous phase density on the XG concentration. Accordingly, a master curve at different volume fractions was built using the time-concentration-superposition approach. The master curve describes the rheological response of the system over a wider frequency window than the experimentally accessible one and reveals the presence of a superimposed β relaxation process in the high-frequency region.

Thermal, spectroscopic, SEM and rheological datasets of native and quaternized guar gum.

Project description:The manuscript reports TGA, DTA, SEM and NMR datasets of native and quaternized guar gum as a tool for their characterization. The TGA and DTA data was acquired in temperature range 35-500 °C. Based on the TGA experimental values, activation energy plots were drawn to study the stability of native and quaternized polysaccharides (Cai and Bi, 2008 [1]). The data demonstrates the thermal behaviour of quaternized guar gum vis-a-vis native guar gum. The surface morphology of native and quaternized galactomannan was represented by SEM imaging. The 1H-1H COSY was acquired to understand structural changes by quaternization. The rheological measurements of native and modified products were carried out to obtain the viscosity profile of the respective samples. The datasets support the research article 'Synthesis of quaternized guar gum using Taguchi L (16) orthogonal array' (Tyagi et al., 2020 [2]).

New Quaternary Ammonium Derivatives Based on Citrus Pectin.

Project description:New citrus pectin derivatives carrying pendant N,N-dimethyl-N-alkyl-N-(2-hydroxy propyl) ammonium chloride groups were achieved via polysaccharide derivatization with a mixture of N,N-dimethyl-N-alkyl amine (alkyl = ethyl, butyl, benzyl, octyl, dodecyl) and epichlorohydrin in aqueous solution. The structural characteristics of the polymers were examined via elemental analysis, conductometric titration, Fourier Transform Infrared spectroscopy (FTIR) and 1D (1H and 13C) nuclear magnetic resonance (NMR). Capillary viscosity measurements allowed for the study of viscometric behavior as well as the determination of viscosity-average molar mass for pristine polysaccharide and intrinsic viscosity ([η]) values for pectin and its derivatives. Dynamic light scattering measurements (DLS) showed that pectin-based polymers formed aggregates in aqueous solution with a unimodal distribution. Critical aggregation concentration (cac) for the hydrophobic pectin derivatives were determined using fluorescence spectroscopy. Atom force microscopy (AFM) images allowed for the investigation of the morphology of polymeric populations obtained in aqueous solution, consisting of flocs and aggregates for crude pectin and its hydrophilic derivatives and well-organized aggregates for lipophilic pectin derivatives. Antimicrobial activity, examined using the disc diffusion method, proved that all polymers were active against Staphylococcus aureus bacterium and Candida albicans yeast.
