Development of a Low-Cost UV-Vis Spectrophotometer and Its Application for the Detection of Mercuric Ions Assisted by Chemosensors.

ABSTRACT: Detection of an environmental contaminant requires the use of expensive measurement equipment, which limits the realization of in situ tests because of their high cost, their limited portability, or the extended time duration of the tests. This paper presents in detail the development of a portable low-cost spectrophotometer which, by using a specialized chemosensor, allows detection of mercuric ions (Hg2+), providing effective and accurate results. Design specifications for all the stages assembling the spectrophotometer and the elements selected to build them are presented along with the process to synthesize the chemosensor and the tests developed to validate its performance in comparison with a high-precision commercial laboratory spectrophotometer.

Optofluidic UV-Vis spectrophotometer for online monitoring of photocatalytic reactions.

Project description:On-chip integration of optical detection units into the microfluidic systems for online monitoring is highly desirable for many applications and is also well in line with the spirit of optofluidics technology-fusion of optics and microfluidics for advanced functionalities. This paper reports the construction of a UV-Vis spectrophotometer on a microreactor, and demonstrates the online monitoring of the photocatalytic degradations of methylene blue and methyl orange under different flow rates and different pH values by detecting the intensity change and/or the peak shift. The integrated device consists of a TiO2-coated glass substrate, a PDMS micro-sized reaction chamber and two flow cells. By comparing with the results of commercial equipment, we have found that the measuring range and the sensitivity are acceptable, especially when the transmittance is in the range of 0.01-0.9. This integrated optofluidic device can significantly cut down the test time and the sample volume, and would provide a versatile platform for real-time characterization of photochemical performance. Moreover, its online monitoring capability may enable to access the usually hidden information in biochemical reactions like intermediate products, time-dependent processes and reaction kinetics.

Rapid and Low-Cost Quantification of Adulteration Content in <i>Camellia</i> Oil Utilizing UV-Vis-NIR Spectroscopy Combined with Feature Selection Methods.

Project description:This study aims to explore the potential use of low-cost ultraviolet-visible-near infrared (UV-Vis-NIR) spectroscopy to quantify adulteration content of soybean, rapeseed, corn and peanut oils in Camellia oil. To attain this aim, test oil samples were firstly prepared with different adulterant ratios ranging from 1% to 90% at varying intervals, and their spectra were collected by an in-house built experimental platform. Next, the spectra were preprocessed using Savitzky-Golay (SG)-Continuous Wavelet Transform (CWT) and the feature wavelengths were extracted using four different algorithms. Finally, Support Vector Regression (SVR) and Random Forest (RF) models were developed to rapidly predict adulteration content. The results indicated that SG-CWT with decomposition scale of 25 and the Iterative Variable Subset Optimization (IVSO) algorithm can effectively improve the accuracy of the models. Furthermore, the SVR model performed best for predicting adulteration of camellia oil with soybean oil, while the RF models were optimal for camellia oil adulterated with rapeseed, corn, or peanut oil. Additionally, we verified the models' robustness by examining the correlation between the absorbance and adulteration content at certain feature wavelengths screened by IVSO. This study demonstrates the feasibility of using low-cost UV-Vis-NIR spectroscopy for the authentication of Camellia oil.

Development of detection system for lead ions in mixture solutions using UV-Vis measurements with peptide immobilized microbeads.

Project description:Environmental pollution caused by heavy metals are problems worldwide. In particular, pollution and poisoning by lead ions (Pb2+) continue to be common and serious problems. Hence, there is a need for a widely usable method to easily detect Pb2+ from solutions containing organic materials from environmental water such as seas, ponds, etc. Here, we established a system to easily detect Pb2+ from such mixture solutions using Pb2+ binding peptide immobilized beads (peptidyl beads) and ultraviolet (UV) absorption measurements. This method could detect Pb2+ at low concentrations equivalent to inductively coupled plasmon-atomic emission spectroscopy (ICP-AES). Using the detected values to create a calibration curve, it was found that there was a positive correlation between the concentration of Pb2+ and absorbance, which also made it possible to quantify sub-µM Pb2+ in the solutions. Furthermore, Pb2+ was detected and quantified under mixed conditions of environmental water such as seas, rivers, and ponds. This method is expected to become a versatile and easy-to-use Pb2+ detection method for end-users worldwide.

UV Lithography-Assisted Fabrication of Low-Cost Copper Electrodes Modified with Gold Nanostructures for Improved Analyte Detection.

Project description:An in-house UV lithography setup has been optimized to fabricate low-cost disposable electrochemical sensing Cu electrodes using a copper clad board. In view of the high oxidation probability of copper, the low-cost electrodes were modified using different gold nanostructures and a conducing polymer PEDOT:PSS to attain maximal signal output and improved shelf-life. Zero-dimensional (0D) gold nanoparticles (∼40 nm) and three-dimensional (3D) gold nanoflowers (∼38 nm) mixed with PEDOT:PSS were used as signal-enhancing conductors for the ultrasensitive detection of our model contaminant, methylene blue dye (MB). The bare copper electrode was sensitive to MB, linearly within the range of 4-100 μM, with a limit of detection of 3.49 μM. While for gold nanoparticle-PEDOT:PSS-modified electrode, the sensitivity of the electrode was found to increase linearly in the range of 0.01-0.1 μM, and for gold nanoflowers-PEDOT:PSS, the sensitivity achieved was 0.01-0.1 μM with the LOD as 0.0022 μM. For a PEDOT:PSS-modified Cu electrode, used as a comparative to study the contributing role of gold nanostructures towards improved sensitivity, the linearity was found to be in the range of 0.1-1.9 μM with the LOD as 0.0228 μM. A 6 times improvement in signal sensitivity for the nanoflower-PEDOT:PSS electrode compared to the nanoparticle-PEDOT:PSS-modified electrode indicates the influence of nanoparticle shape on the electrode efficiency. 3D gold nanoflowers with a large surface area-to-volume ratio and a high catalytic activity prove to be a superior choice for electrode modification.

Efficient Optical and UV-Vis Chemosensor Based on Chromo Probes-Polymeric Nanocomposite Hybrid for Selective Recognition of Fluoride Ions.

Project description:A novel colorimetric sensor based on the TiO2/poly(acrylamide-co-methylene bis acrylamide-co-2-(3-(4-nitro-phenyl)thioureido)ethyl methacrylate) nanocomposite was synthesized via a surface modification strategy; methacryloxypropyltrimethoxysilane was used to provide reactive vinyl groups on the surface of TiO2 nanoparticles for the successful surface polymerization of Am (acrylamide), MBA (methylenbisacrylamide), and NPhM (2-(3-(4-nitrophenyl)thioureido)ethyl methacrylate) components. The successful preparation of nanocomposites was confirmed using Fourier transform infrared, 1H NMR, 13C NMR, scanning electron microscopy, transmission electron microscopy, thermogravimetry analysis, and X-ray diffraction methods, and the sensing ability of the probe toward fluoride ions was investigated using naked-eye detection and UV-vis measurement. The interaction of the prepared polymeric nanocomposite with fluoride ions elicited a significant visible change in color from pale yellow to orange and was further affirmed by a clean interconversion of the two absorption bands at 330 and 485 nm. The selective binding ability of the polymeric nanocomposite towards fluoride over other anions, such as I-, Cl-, Br-, AcO-, H2PO4 -, and H2SO4 - was further explored; the prepared chemosensor could detect fluoride ions in acetonitrile with a detection limit of 3 μM.

Ultraviolet Imaging with Low Cost Smartphone Sensors: Development and Application of a Raspberry Pi-Based UV Camera.

Project description:Here, we report, for what we believe to be the first time, on the modification of a low cost sensor, designed for the smartphone camera market, to develop an ultraviolet (UV) camera system. This was achieved via adaptation of Raspberry Pi cameras, which are based on back-illuminated complementary metal-oxide semiconductor (CMOS) sensors, and we demonstrated the utility of these devices for applications at wavelengths as low as 310 nm, by remotely sensing power station smokestack emissions in this spectral region. Given the very low cost of these units, ≈ USD 25, they are suitable for widespread proliferation in a variety of UV imaging applications, e.g., in atmospheric science, volcanology, forensics and surface smoothness measurements.

A Simple, Fast, Low Cost, HPLC/UV Validated Method for Determination of Flutamide: Application to Protein Binding Studies.

Project description:The main goal of this study was development of a reverse phase high performance liquid chromatography (RP-HPLC) method for flutamide quantitation which is applicable to protein binding studies.Ultrafilteration method was used for protein binding study of flutamide. For sample analysis, flutamide was extracted by a simple and low cost extraction method using diethyl ether and then was determined by HPLC/UV. Acetanilide was used as an internal standard. The chromatographic system consisted of a reversed-phase C8 column with C8 pre-column, and the mobile phase of a mixture of 29% (v/v) methanol, 38% (v/v) acetonitrile and 33% (v/v) potassium dihydrogen phosphate buffer (50 mM) with pH adjusted to 3.2.Acetanilide and flutamide were eluted at 1.8 and 2.9 min, respectively. The linearity of method was confirmed in the range of 62.5-16000 ng/ml (r(2) > 0.99). The limit of quantification was shown to be 62.5 ng/ml. Precision and accuracy ranges found to be (0.2-1.4%, 90-105%) and (0.2-5.3 %, 86.7-98.5 %) respectively. Acetanilide and flutamide capacity factor values of 1.35 and 2.87, tailing factor values of 1.24 and 1.07 and resolution values of 1.8 and 3.22 were obtained in accordance with ICH guidelines.Based on the obtained results a rapid, precise, accurate, sensitive and cost-effective analysis procedure was proposed for quantitative determination of flutamide.

UV-adVISor: Attention-Based Recurrent Neural Networks to Predict UV-Vis Spectra.

Project description:Ultraviolet-visible (UV-Vis) absorption spectra are routinely collected as part of high-performance liquid chromatography (HPLC) analysis systems and can be used to identify chemical reaction products by comparison to the reference spectra. Here, we present UV-adVISor as a new computational tool for predicting the UV-Vis spectra from a molecule's structure alone. UV-Vis prediction was approached as a sequence-to-sequence problem. We utilized Long-Short Term Memory and attention-based neural networks with Extended Connectivity Fingerprint Diameter 6 or molecule SMILES to generate predictive models for the UV spectra. We have produced two spectrum datasets (dataset I, N = 949, and dataset II, N = 2222) using different compound collections and spectrum acquisition methods to train, validate, and test our models. We evaluated the prediction accuracy of the complete spectra by the correspondence of wavelengths of absorbance maxima and with a series of statistical measures (the best test set median model parameters are in parentheses for model II), including RMSE (0.064), R2 (0.71), and dynamic time warping (DTW, 0.194) of the entire spectrum curve. Scrambling molecule structures with the experimental spectra during training resulted in a degraded R2, confirming the utility of the approaches for prediction. UV-adVISor is able to provide fast and accurate predictions for libraries of compounds.

Assessment of the Irradiation Exposure of PET Film with Swift Heavy Ions Using the Interference-Free Transmission UV-Vis Transmission Spectra.

Project description:This paper presents the results of a study of polyethylene terephthalate (PET) films irradiated with Ar and Kr ions at both normal orientation and an angle of 40° to the normal. Normal irradiation was performed using Ar8+ and Kr15+ ions with an energy of 1.75 MeV/au and fluences in the range (2-500) × 1010 cm-2 for Ar8+ ions and (1.6 - 6.5) × 1010 cm-2 for Kr15+ ions. Kr ions with an energy of 1.2 MeV/au and charges of 13+, 14+, and 15+ were used for angled irradiation. For each Kr ion charge value, three fluence values were used: 5 × 1010, 1 × 1011, and 2.5 × 1011 cm-2. It is well known that irradiation of PET films by swift heavy ions results in a red shift of the UV-vis transmission spectra absorption edge. The experimental transmission spectra exhibit well-defined interference fringes, which obscure the underlying transmission response. Using an existing technique to obtain interference-free transmission curves Tα(λ) for both pristine and irradiated PET film samples, we found that S, the total radiation-induced absorption of light by the PET film, is proportional to the logarithm of the fluence F. In addition to this dependence on the irradiating fluence, we also found that the charge of the irradiating ion has a significant influence on the position of the absorption edge in the UV-vis spectra. This provides experimentally independent evidence to confirm our previous results showing that ion charge has an effect on the post-irradiation state of PET films. We present a physical interpretation of the observed absorption edge red shift in irradiated PET films as being due to the growth of extended conjugated systems via the formation of intermolecular helical structures. Our investigations into the stability of irradiation-induced effects in PET films show that comparison of UV-vis transmission spectra before and after annealing can provide information about the structure of deep traps in PET.

A mobile microvolume UV/visible light spectrophotometer for the measurement of levofloxacin in saliva.

Project description:IntroductionTherapeutic drug monitoring (TDM) for personalized dosing of fluoroquinolones has been recommended to optimize efficacy and reduce acquired drug resistance in the treatment of MDR TB. Therefore, the aim of this study was to develop a simple, low-cost, robust assay for TDM using mobile UV/visible light (UV/VIS) spectrophotometry to quantify levofloxacin in human saliva at the point of care for TB endemic settings.MethodsAll experiments were performed on a mobile UV/VIS spectrophotometer. The levofloxacin concentration was quantified by using the amplitude of the second-order spectrum between 300 and 400 nm of seven calibrators. The concentration of spiked samples was calculated from the spectrum amplitude using linear regression. The method was validated for selectivity, specificity, linearity, accuracy and precision. Drugs frequently co-administered were tested for interference.ResultsThe calibration curve was linear over a range of 2.5-50.0 mg/L for levofloxacin, with a correlation coefficient of 0.997. Calculated accuracy ranged from -5.2% to 2.4%. Overall precision ranged from 2.1% to 16.1%. Application of the Savitsky-Golay method reduced the effect of interferents on the quantitation of levofloxacin. Although rifampicin and pyrazinamide showed analytical interference at the lower limit of quantitation of levofloxacin concentrations, this interference had no implication on decisions regarding the levofloxacin dose.ConclusionsA simple UV/VIS spectrophotometric method to quantify levofloxacin in saliva using a mobile nanophotometer has been validated. This method can be evaluated in programmatic settings to identify patients with low levofloxacin drug exposure to trigger personalized dose adjustment.
