Do Foliar Endophytes Matter in Litter Decomposition?

ABSTRACT: Litter decomposition rates are affected by a variety of abiotic and biotic factors, including the presence of fungal endophytes in host plant tissues. This review broadly analyzes the findings of 67 studies on the roles of foliar endophytes in litter decomposition, and their effects on decomposition rates. From 29 studies and 1 review, we compiled a comprehensive table of 710 leaf-associated fungal taxa, including the type of tissue these taxa were associated with and isolated from, whether they were reported as endo- or epiphytic, and whether they had reported saprophytic abilities. Aquatic (i.e., in-stream) decomposition studies of endophyte-affected litter were significantly under-represented in the search results (p < 0.0001). Indicator species analyses revealed that different groups of fungal endophytes were significantly associated with cool or tropical climates, as well as specific plant host genera (p < 0.05). Finally, we argue that host plant and endophyte interactions can significantly influence litter decomposition rates and should be considered when interpreting results from both terrestrial and in-stream litter decomposition experiments.

Project description:Organic matter input regulates the rate and temperature sensitivity (expressed as Q 10) of soil organic matter (SOM) decomposition by changing microbial composition and activities. It remains unclear how the incorporation of litter-made biochar instead of litter affects the Q 10 of SOM decomposition. Using a unique combination of two-and three-source partitioning methods (isotopic discrimination between C3/C4 pathways and 14C labeling), we investigated: (1) how maize litter versus litter-made biochar (of C4 origin) addition influenced the Q 10 of SOM (C3 origin) under 10°C warming, and (2) how the litter or biochar amendments affected the Q 10 of 14C-labeled fresh organic matter (FOM) after long-term incubation. Compared with biochar addition, litter increased the rates and Q 10 of mass-specific respiration, SOM and FOM decomposition, as well as the contents of SOM-derived dissolved organic C (DOC) and total phospholipid fatty acids (PLFA). Litter-amended soils have much higher activities (V max) of β-glucosidase, N-acetyl-β-glucosaminidase, and leucine aminopeptidase, suggesting larger enzyme pools than in soils with biochar. The Q 10 of enzyme V max (1.6-2.0) and K m (1.2-1.4) were similar between litter-and biochar-amended soils, and remained stable with warming. However, warming reduced microbial biomass (PLFA) and enzyme activity (V max), suggesting decreased enzyme production associated with smaller microbial biomass or faster enzyme turnover at higher temperatures. Reductions in PLFA content and enzyme V max due to warming were larger in litter-amended soils (by 31%) than in the control and biochar-amended soils (by 4-11%), implying the active litter-feeding microorganisms have a smaller degree of heat tolerance than the inactive microorganisms under biochar amendments. The reduction in enzyme activity (V max) by warming was lower in soils with biochar than in the control soil. Our modeling suggested that the higher Q 10 in litter-amended soils was mainly caused by faster C loss under warming, linked to reductions in microbial biomass and growth efficiency, rather than the slightly increased SOM-originated substrate availability (DOC). Overall, using straw-made biochar instead of straw per se as a soil amendment lowers the Q 10 of SOM and FOM by making microbial communities and enzyme pools more temperature-tolerant, and consequently reduces SOM losses under warming.

Project description:Abundant secondary metabolites, such as condensed tannins, and their interpopulation genotypic variation can remain through plant leaf senescence and affect litter decomposition. Whether the intrapopulation genotypic variation of a more diverse assortment of secondary metabolites equally persists through leaf senescence and litter decomposition is not well understood. We analyzed concentrations of intracellular phenolics, epicuticular flavonoid aglycones, epicuticular triterpenoids, condensed tannins, and lignin in green leaves, senescent leaves and partly decomposed litter of silver birch, Betula pendula. Broad-sense heritability (H2) and coefficient of genotypic variation (CVG) were estimated for metabolites in senescent leaves and litter using 19 genotypes selected from a B. pendula population in southern Finland. We found that most of the secondary metabolites remained through senescence and decomposition and that their persistence was related to their chemical properties. Intrapopulation H2 and CVG for intracellular phenolics, epicuticular flavonoid aglycones and condensed tannins were high and remarkably, increased from senescent leaves to decomposed litter. The rank of genotypes in metabolite concentrations was persistent through litter decomposition. Lignin was an exception, however, with a diminishing genotypic variation during decomposition, and the concentrations of lignin and condensed tannins had a negative genotypic correlation in the senescent leaves. Our results show that secondary metabolites and their intrapopulation genotypic variation can for the most part remain through leaf senescence and early decomposition, which is a prerequisite for initial litter quality to predict variation in litter decomposition rates. Persistent genotypic variation also opens an avenue for selection to impact litter decomposition in B. pendula populations through acting on their green foliage secondary chemistry. The negative genotypic correlations and diminishing heritability of lignin concentrations may, however, counteract this process.

Project description:Mixing plant litters of multiple species can alter litter decomposition, a key driver of carbon and nutrient cycling in terrestrial ecosystems. Changes in microbial decomposer communities is proposed as one of the mechanisms explaining this litter-mixture effect, but the underlying mechanism is unclear. In a microcosm litterbag experiment, we found that, at the early stage of decomposition, litter mixing promoted tomato root litter decomposition, thus generating a synergistic nonadditive litter-mixture effect. The transplanting decomposer community experiment showed that changes in microbial decomposer communities contributed to the nonadditive litter-mixture effect on tomato root litter decomposition. Moreover, litter mixing altered the abundance and diversity of bacterial and fungal communities on tomato root litter. Litter mixing also stimulated several putative keystone operational taxonomic units (OTUs) in the microbial correlation network, such as Fusarium sp. fOTU761 and Microbacterium sp. bOTU6632. Then, we isolated and cultured representative isolates of these two taxa, named Fusarium sp. F13 and Microbacterium sp. B26. Subsequent in vitro tests found that F13, but not B26, had strong decomposing ability; moreover, these two isolates developed synergistic interaction, thus promoted litter decomposition in coculture. Addition of F13 or B26 both promoted the decomposing activity of the resident decomposer community on tomato root litter, confirming their importance for litter decomposition. Overall, litter mixing promoted tomato root litter decomposition through altering microbial decomposers, especially through stimulating certain putative keystone taxa. IMPORTANCE Microbial decomposer community plays a key role in litter decomposition, which is an important regulator of soil carbon and nutrient cycling. Though changes in decomposer communities has been proposed as one of the potential underlying mechanisms driving the litter-mixture effects, direct evidence is still lacking. Here, we demonstrated that litter mixing stimulated litter decomposition through altering microbial decomposers at the early stage of decomposition. Moreover, certain putative keystone taxa stimulated by litter mixing contributed to the nonadditive litter-mixture effect. In vitro culturing validated the role of these taxa in litter decomposition. This study also highlights the possibility of regulating litter decomposition through manipulating certain microbial taxa.
