Towards triptycene functionalization and triptycene-linked porphyrin arrays.

ABSTRACT: Herein, 9,10-diethynyltriptycene is investigated for its use as a rigid isolating unit in the synthesis of multichromophoric arrays. Sonogashira cross-coupling conditions are utilized to attach various porphyrins and boron dipyrromethenes (BODIPYs) to the triptycene scaffold. While there are previous examples of triptycene porphyrin complexes, this work reports the first example of a linearly connected porphyrin dimer, linked through the bridgehead carbons of triptycene. Symmetric and unsymmetric examples of these complexes are demonstrated and single crystal X-ray analysis of an unsymmetrically substituted porphyrin dimer highlights the evident linearity in these systems. Moreover, initial UV-vis and fluorescence studies show the promise of triptycene as a linker for electron transfer studies, showcasing its isolating nature.

Singly and Triply Linked Magnetic Porphyrin Lanthanide Arrays.

Project description:The introduction of paramagnetic metal centers into a conjugated π-system is a promising approach toward engineering spintronic materials. Here, we report an investigation of two types of spin-bearing dysprosium(III) and gadolinium(III) porphyrin dimers: singly meso-meso-linked dimers with twisted conformations and planar edge-fused β,meso,β-linked tapes. The rare-earth spin centers sit out of the plane of the porphyrin, so that the singly linked dimers are chiral, and their enantiomers can be resolved, whereas the edge-fused tape complexes can be separated into syn and anti stereoisomers. We compare the crystal structures, UV-vis-NIR absorption spectra, electrochemistry, EPR spectroscopy, and magnetic behavior of these complexes. Low-temperature SQUID magnetometry measurements reveal intramolecular antiferromagnetic exchange coupling between the GdIII centers in the edge-fused dimers (syn isomer: J = -51 ± 2 MHz; anti isomer: J = -19 ± 3 MHz), whereas no exchange coupling is detected in the singly linked twisted complex. The phase-memory times, Tm, are in the range of 8-10 μs at 3 K, which is long enough to test quantum computational schemes using microwave pulses. Both the syn and anti Dy2 edge-fused tapes exhibit single-molecule magnetic hysteresis cycles at temperatures below 0.5 K with slow magnetization dynamics.

Spatioselective functionalization of gold nanopillar arrays.

Project description:A process combining electrochemical nanofabrication by hard templating with the use of a masking strategy and surface functionalization methods, is developed to produce arrays of gold nanopillars of spatially-controlled surface chemistry. Therefore, a gold nanopillar array is first fabricated by performing metal electrochemical deposition into a track-etched membrane supported on a gold substrate. After dissolution of the membrane, a protective polymer layer is deposited on the array and partially etched to specifically reveal the top of the nanopillars. Then, a polythiolactone-based copolymer is grafted on the upper part of the nanopillars. Afterwards, the sacrificial polymer layer is dissolved to reveal the non-functionalized surface corresponding to the lower part of the gold nanopillars and the background surface. This surface is subsequently modified by a self-assembled monolayer (SAM) of alkylthiol molecules which leads to nanostructured surfaces with spatio-selective surface chemistry. The grafting of gold nanoparticles and of a bioadhesive peptide on the top and on the background of the nanopillar array, respectively, is performed to prove the versatility of the approach to produce bifunctionalized nanopillar arrays for biological, biosensing or (bio)catalysis applications.

Self-complementary double-stranded porphyrin arrays assembled from an alternating pyridyl-porphyrin sequence.

Project description:Oligomeric porphyrin arrays with an alternating pyridyl-porphyrin sequence were synthesized to explore double-strand formation through self-complementary pyridyl-to-zinc axial coordination bonds. Competitive titration experiments revealed the thermodynamic aspects involved in the zipper effect within double-strand formation. Multiple axial coordination bonds defined the stacked conformation, despite a marginal contribution to the stability of the double-strands. Thus, the zipper cooperativity was the dominant factor for the remarkable stability. Moreover, the dimeric and trimeric porphyrin arrays were independently assembled into double-strands by self-sorting from a binary mixture. Double-strand formation engineered discretely stacked π-systems. Successive slipped-cofacial stacks of the porphyrin rings progressively extended the π-system via exciton coupling over the double-strand while keeping a relatively high fluorescence quantum yield.

Single-Acetylene Linked Porphyrin Nanorings.

Project description:The synthesis of ethyne-linked porphyrin nanorings has been achieved by template-directed Sonogashira coupling. The cyclic hexamer and octamer are predicted by density functional theory to adopt low symmetry conformations, due to dihedral twists between neighboring porphyrin units, but their symmetries are effectively D6h and D8h, respectively, in solution by 1H NMR. The fluorescence spectra indicate that the singlet excited states of these nanorings are highly delocalized.

Ordering, flexibility and frustration in arrays of porphyrin nanorings.

Project description:The regular packing of atoms, molecules and nanoparticles provides the basis for the understanding of structural order within condensed phases of matter. Typically the constituent particles are considered to be rigid with a fixed shape. Here we show, through a combined experimental and numerical study of the adsorption of cyclic porphyrin polymers, nanorings, on a graphite surface, that flexible molecules can exhibit a rich and complex packing behaviour. Depending on the number of porphyrin sub-units within the nanoring we observe either a highly ordered hexagonal phase or frustrated packing driven by directional interactions which for some arrangements is combined with the internal deformation of the cyclic polymer. Frustration and deformation occur in arrays of polymers with ten sub-units since close packing and co-alignment of neighbouring groups cannot be simultaneously realised for nanorings with this internal symmetry.

Polydopamine surface functionalization of 3D printed resin material for enhanced polystyrene adhesion towards insulation layers for 3D microelectrode arrays (3D MEAs).

Project description:3D printing involves the use of photopolymerizable resins, which are toxic and typically have incompatible properties with materials such as polystyrene (PS), which present limitations for biomedical applications. We present a method to dramatically improve the poor adhesion between the PS insulative layer on 3D printed Microelectrode Array (MEA) substrates by functionalizing the resin surface with polydopamine (PDA), a mussel-inspired surface chemistry derivative. A commercial 3D printing prepolymer resin, FormLabs Clear (FLC), was printed using a digital light processing (DLP) printer and then surface functionalized with PDA by alkali-induced aqueous immersion deposition and self-polymerization. It was observed that the adhesion of the PS to FLC was improved due to the precision emanating from the DLP method and further improved after the functionalization of DLP printed substrates with PDA at 1, 12, and 24 h time intervals. The adhesion of PS was evaluated through scotch tape peel testing and instron measurements of planar substrates and incubation testing with qualitative analysis of printed culture wells. The composition and topology of the samples were studied to understand how the properties of the surface change after PDA functionalization and how this contributes to the overall improvement in PS adhesion. Furthermore, the surface energies at each PDA deposition time were calculated from contact angle studies as it related to adhesion. Finally, biocompatibility assays of the newly modified surfaces were performed using mouse cardiac cells (HL-1) to demonstrate the biocompatibility of the PDA functionalization process. PDA surface functionalization of 3D DLP printed FLC resin resulted in a dramatic improvement of thin film PS adhesion and proved to be a biocompatible solution for improving additive manufacturing processes to realize biosensors such as in vitro MEAs.

Mechanochemical functionalization of disulfide linked hydrogels.

Project description:Poly(ethylene glycol) hydrogels with disulfide linkages are functionalized through applied force. Compression or tension induces bond rupture at the relatively weak disulfide linkages, which will subsequently react through Michael-type addition with an acceptor molecule within the gel. We demonstrate the utility of this approach by patterning cell adhesion proteins through compression of a lithographically structured stamp, where cells predominately adhere to the compressed regions.

Functionalization of cobalt porphyrin-phospholipid bilayers with his-tagged ligands and antigens.

Project description:Methods to attach polypeptides to lipid bilayers are often indirect and ineffective, and can represent a substantial bottleneck in the formation of functionalized lipid-based materials. Although the polyhistidine tag (his-tag) has been transformative in its simplicity and efficacy in binding to immobilized metals, the successful application of this approach has been challenging in physiological settings. Here we show that lipid bilayers containing porphyrin-phospholipid conjugates that are chelated with cobalt, but not with other metals, can effectively capture his-tagged proteins and peptides. The binding follows a Co(II) to Co(III) transition and occurs within the sheltered hydrophobic bilayer, resulting in an essentially irreversible attachment in serum or in a million fold excess of competing imidazole. Using this approach we anchored homing peptides into the bilayer of preformed and cargo-loaded liposomes to enable tumour targeting without disrupting the bilayer integrity. As a further demonstration, a synthetic protein fragment derived from the human immunodeficiency virus was bound to immunogenic liposomes for potent antibody generation for an otherwise non-antigenic peptide.

Robust Functionalization of Large Microelectrode Arrays by Using Pulsed Potentiostatic Deposition.

Project description:Surface modification of microelectrodes is a central step in the development of microsensors and microsensor arrays. Here, we present an electrodeposition scheme based on voltage pulses. Key features of this method are uniformity in the deposited electrode coatings, flexibility in the overall deposition area, i.e., the sizes and number of the electrodes to be coated, and precise control of the surface texture. Deposition and characterization of four different materials are demonstrated, including layers of high-surface-area platinum, gold, conducting polymer poly(ethylenedioxythiophene), also known as PEDOT, and the non-conducting polymer poly(phenylenediamine), also known as PPD. The depositions were conducted using a fully integrated complementary metal-oxide-semiconductor (CMOS) chip with an array of 1024 microelectrodes. The pulsed potentiostatic deposition scheme is particularly suitable for functionalization of individual electrodes or electrode subsets of large integrated microelectrode arrays: the required deposition waveforms are readily available in an integrated system, the same deposition parameters can be used to functionalize the surface of either single electrodes or large arrays of thousands of electrodes, and the deposition method proved to be robust and reproducible for all materials tested.
