Nanoparticles Based on Novel Carbohydrate-Functionalized Polymers.

ABSTRACT: Polymeric nanoparticles can be used for drug delivery systems in healthcare. For this purpose poly(lactic-co-glycolic acid) (PLGA) and poly(ethylene glycol) (PEG) offer an excellent polymeric matrix. In this work, PLGA and PEG polymers were functionalized with coumarin and carbohydrate moieties such as thymidine, glucose, galactose, and mannose that have high biological specificities. Using a single oil in water emulsion methodology, functionalized PLGA nanoparticles were prepared having a smooth surface and sizes ranging between 114-289 nm, a low polydispersity index and a zeta potential from -28.2 to -56.0 mV. However, for the corresponding PEG derivatives the polymers obtained were produced in the form of films due to the small size of the hydrophobic core.

Project description:Carbohydrates play prominent roles in immune surveillance and response to infection. Multivalency, molecular weight control, and molecular architecture control are properties that polymer science is well suited to address. Each of these properties has been demonstrated to impact the biological interaction of carbohydrate-bearing chains with their binding partners. This viewpoint highlights synthetic advances and potential applications of carbohydrate-based polymers for immune modulation. It also offers future directions in polymer science necessary for carbohydrate polymers to fulfill their potential as immune modulators.

Ionic Porous Organic Polymers Based on Functionalized Tetraarylborates.

Project description:Lithium tetrakis(4-boronatoaryl)borates were subjected to polycondensation reactions with selected polyhydroxyl monomers such as 2,3,6,7,10,11-hexahydroxytriphenylene (HHTP) and 2,3,6,7-tetrahydroxy-9,10-dimethylanthracene (THDMA). Obtained boronate-type ionic porous polymers TAB1-4 were characterized by PXRD, 6Li and 11B magic-angle spinning nuclear magnetic resonance (MAS NMR), FT-IR, SEM, and TGA. They exhibit relatively good sorption of H2 (up to 75 cm3/g STP), whereas N2 uptake at 77 K for lower pressure range is relatively poor (up to 50 cm3/g STP below P/P0 = 0.8). In addition, the effect of elongation of aryl arms in the tetraarylborate core on the materials' properties was studied. Thus, it was found that replacement of the 4-boronatophenyl with 4-boronatobiphenylyl group has a negative impact on the sorption characteristics.

Magnetite Nanoparticles Functionalized with Thermoresponsive Polymers as a Palladium Support for Olefin and Nitroarene Hydrogenation.

Project description:A thermoresponsive and recyclable nanomaterial was synthesized by surface modification of magnetite nanoparticles (MNPs) with poly(N-isopropylacrylamide-co-diethylaminoethyl methacrylate) (P(NIPAAm-co-DEAEMA)), having PNIPAAm as a thermoresponsive moiety and PDEAEMA for catalyst binding. Palladium (Pd) nanoparticles were incorporated into this material, and the resulting nanocatalyst was efficient in the hydrogenation of olefins and nitro compounds with turnover frequencies (TOFs) up to 750 h-1. Consistent catalytic activity in 10 consecutive runs was observed when performing the hydrogenation at 45 °C, i.e., above the lower critical solution temperature (LCST) of the copolymer (37 °C), followed by cooling to 15 °C, i.e., below the LCST of the copolymer.

Arylene Ethynylene-Functionalized Bithiazole-Based Zinc Polymers for Ultraefficient Photocatalytic Activity.

Project description:Polymers 4 containing poly(arylene ethynylene) were synthesized and characterized systematically. Among them, 4c exhibited a remarkable H2 evolution rate (14.32 mmol h-1 g-1) with visible-light irradiation, lasting 72 h in different water qualities; the corresponding apparent quantum yield was 11.6% at 450 nm.

Aptamer-functionalized lipid nanoparticles targeting osteoblasts as a novel RNA interference-based bone anabolic strategy.

Project description:Currently, major concerns about the safety and efficacy of RNA interference (RNAi)-based bone anabolic strategies still exist because of the lack of direct osteoblast-specific delivery systems for osteogenic siRNAs. Here we screened the aptamer CH6 by cell-SELEX, specifically targeting both rat and human osteoblasts, and then we developed CH6 aptamer-functionalized lipid nanoparticles (LNPs) encapsulating osteogenic pleckstrin homology domain-containing family O member 1 (Plekho1) siRNA (CH6-LNPs-siRNA). Our results showed that CH6 facilitated in vitro osteoblast-selective uptake of Plekho1 siRNA, mainly via macropinocytosis, and boosted in vivo osteoblast-specific Plekho1 gene silencing, which promoted bone formation, improved bone microarchitecture, increased bone mass and enhanced mechanical properties in both osteopenic and healthy rodents. These results indicate that osteoblast-specific aptamer-functionalized LNPs could act as a new RNAi-based bone anabolic strategy, advancing the targeted delivery selectivity of osteogenic siRNAs from the tissue level to the cellular level.

A novel probe based on phenylboronic acid functionalized carbon nanotubes for ultrasensitive carbohydrate determination in biofluids and semi-solid biotissues.

Project description:Carbohydrates are known to be involved in a wide range of biological and pathological processes. However, due to the presence of multiple hydroxyl groups, carbohydrate recognition is a particular challenge. Herein, we reported an ultrasensitive solid-phase microextraction (SPME) probe based on phenylboronic acid (PBA) functionalized carbon nanotubes (CNTs) for direct in vitro or in vivo recognition of carbohydrates in biofluids as well as semi-solid biotissues. The coating of the proposed probe possessed a 3D interconnected porous architecture formed by the stacking of CNTs. As a result, the binding capacity toward carbohydrates was excellent. The proposed approach was demonstrated to be much superior to most carbohydrate sensors, including higher sensitivity, wider linear range, and excellent qualitative ability in multi-carbohydrate systems. Thus, this approach opens up new avenues for the facile and efficient recognition of carbohydrates for important applications such as glycomics.

Synthesis of homo- and heteromultivalent carbohydrate-functionalized oligo(amidoamines) using novel glyco-building blocks.

Project description:We present the solid phase synthesis of carbohydrate-functionalized oligo(amidoamines) with different functionalization patterns utilizing a novel alphabet of six differently glycosylated building blocks. Highly efficient in flow conjugation of thioglycosides to a double-bond presenting diethylentriamine precursor is the key step to prepare these building blocks suitable for fully automated solid-phase synthesis. Introduction of the sugar ligands via functionalized building blocks rather than postfunctionalization of the oligomeric backbone allows for the straightforward synthesis of multivalent glycoligands with full control over monomer sequence and functionalization pattern. We demonstrate the potential of this building-block approach by synthesizing oligomers with different numbers and spacing of carbohydrates and also show the feasibility of heteromultivalent glycosylation patterns by combining building blocks presenting different mono- and disaccharides.

End-functionalized ROMP polymers for Biomedical Applications.

Project description:We present two novel allyl-based terminating agents that can be used to end-functionalize living polymer chains obtained by ring-opening metathesis polymerization (ROMP) using Grubbs' third generation catalyst. Both terminating agents can be easily synthesized and yield ROMP polymers with stable, storable activated ester groups at the chain-end. These end-functionalized ROMP polymers are attractive building blocks for advanced polymeric materials, especially in the biomedical field. Dye-labeling and surface-coupling of antimicrobially active polymers using these end-groups were demonstrated.

Mucus-responsive functionalized emulsions: design, synthesis and study of novel branched polymers as functional emulsifiers.

Project description:Mucus lines the moist cavities throughout the body, acting as barrier by protecting the underlying cells against the external environment, but it also hinders the permeation of drugs and drug delivery systems. As the rate of diffusion is low, the development of a system which could increase retention time at the mucosal surface would prove beneficial. Here, we have designed a range of branched copolymers to act as functional mucus-responsive oil-in-water emulsifiers comprising the hydrophilic monomer oligo(ethylene glycol) methacrylate and a hydrophobic dodecyl initiator. The study aimed to investigate the importance of chain end functionality on successful emulsion formation, by systematically replacing a fraction of the hydrophobic chain ends with a secondary poly(ethylene glycol) based hydrophilic initiator in a mixed-initiation strategy; a decrease of up to 75 mole percent of hydrophobic chain ends within the branched polymer emulsifiers was shown to maintain comparative emulsion stability. These redundant chain ends allowed for functionality to be incorporated into the polymers via a xanthate based initiator containing a masked thiol group; thiol groups are known to have mucoadhesive character, due to their ability to form disulfide bonds with the cysteine rich areas of mucus. The mucoadhesive nature of emulsions stabilised by thiol-containing branched copolymers was compared to non-functional emulsions in the presence of a biosimilar mucosal substrate and enhanced adherence to the mucosal surface was observed. Importantly, droplet rupture and mucus triggered release of dye-containing oil was seen from previously highly-stable thiol-functional emulsions; this observation was not mirrored by non-functional emulsions where droplet integrity was maintained even in the presence of mucus.
