Effects of rumen bypass melatonin feeding (RBMF) on milk quality and mastitis of Holstein cows.

ABSTRACT: Cow mastitis is a major problem frequently encountered by dairy farmers and it is manifested by the high number of somatic cells and the low quality of the milk. The conventional treatment for mastitis is use of antibiotics. In the current study, a new approach is applied to target this disorder: rumen bypass melatonin feeding (RBMF). The RBMF significantly reduced milk somatic cell count and improved milk nutritional values with the elevated protein, fat and dry matter levels. This approach also suppresses the stress and proinflammatory responses of the cows indicated by the reduced serum cortisol, TNF-α and IL-6 and increased IL-10 levels. Importantly, the beneficial effects of RBMF have lasted for several days after termination of the treatment. The effects of melatonin on the mastitis are probably attributed to the antioxidant and anti-inflammatory activities of melatonin. Considering the none or low toxicity of melatonin to organisms and the no invasive nature of this approach, we recommend that RBMF could be used in large scale in the dairy farming to target the cow mastitis.

Project description:BackgroundBovine mastitis significantly impacts the dairy industry, causing economic losses through reduced milk production, lower milk quality, and increased health risks, and early detection is critical for effective treatment. This study analyzed milk electrical conductivity data from 9,846 Chinese Holstein cows over a two-year period, collected during three daily milking sessions, alongside smart collar data and dairy herd improvement test results. The aim was to conduct a comprehensive genetic analysis and assess the potential of milk electrical conductivity as a biomarker for the early detection of bovine subclinical mastitis.ResultsThe results revealed significant phenotypic and strong genetic correlations (-0.286 to 0.457) between milk electrical conductivity, somatic cell score, milk yield, activity quantity, and milking speed. Logistic regression models yielded area under the curve values ranging from 0.636 to 0.697 and odds ratio values from 9.70 to 10.69, demonstrating a certain predictive capability of milk electrical conductivity for identifying subclinical mastitis. Various factors influencing milk electrical conductivity, including lactation stage, environmental conditions, age at first calving, parity, and body condition score, were identified. The random regression model demonstrated moderate to high heritability of milk electrical conductivity (0.458 to 0.487), particularly during the early to mid-lactation periods, with all estimates exceeding 0.35 However, after day 275 of lactation, the heritability decreased to below 0.2. Notably, shifts in genetic factors affecting milk components were observed around 60 and 270 days into lactation, with increased environmental sensitivity to milk electrical conductivity during these periods.ConclusionsThis study demonstrates that milk electrical conductivity is influenced by multiple factors, such as age at first calving, parity, and body condition score, and exhibits significant phenotypic associations with somatic cell score, milk yield, activity quantity, and milking speed. Although milk electrical conductivity showed moderate to high heritability and potential as a predictor for subclinical mastitis, its low genetic correlations with SCS limit its effectiveness as a standalone indicator. Future research should focus on combining EC with other indicators to improve the accuracy of mastitis detection.

Project description:Conjugated linoleic acid (CLA) can regulate fatty acid metabolism and increase the content of unsaturated fatty acids in milk during the nutritional regulation process in dairy cows. However, its effects on milk composition and milk energy output in dairy cows remain unclear. We aimed to investigate whether RP-CLA reduces milk energy output during early lactation while lowering milk fat, as well as whether it affects milk protein synthesis and the potential mechanisms. We examined the milk composition of Holstein fresh cows at different stages by administering RP-CLA for 7 days and analyzing related gene expression in the MAC-T cell line. RP-CLA did not significantly alter milk yield, lactose, or blood ketone levels in early lactation. Starting RP-CLA from day 21 postpartum reduced milk fat by about 26% (3.74% vs 2.78%) on day 28 postpartum, slightly increased milk protein, and improved blood glucose on day 24. No such effects were seen with RP-CLA beginning on day 7 postpartum. RP-CLA supplementation saved 6.45 and 11.43% milk energy output during days 7-14 and 21-28, respectively. We hypothesize that the limited RP-CLA regulation effect during days 7-21 may relate to intense lipid mobilization under negative energy balance. In MAC-T cells under low glucose, t-10, c-12 CLA decreased the expression of SREBP1 and AMPK, while increased CDK1, indicating its role in reducing fat synthesis, easing energy deficiency, promoting cell proliferation, and enhancing protein synthesis. Similarly, under normal glucose, t-10, c-12 CLA suppressed SREBP1 and elevated CDK1 expression, with AMPK reduction only at low doses. Inhibiting milk fat synthesis with an SREBP1 inhibitor slightly upregulated CDK 1 without significantly affecting AMPK. In conclusion, RP-CLA has the potential to alleviate negative energy balance in dairy cows during days 21-35 postpartum. t10, c12 CLA can inhibit milk fat synthesis by reducing the expression of SREBP1 and AMPK in mammary epithelial cells, while increasing the expression of CDK1, thereby improving the cellular energy status and promoting milk protein synthesis. The energy required for the increase in milk protein is not derived from the energy saved by the inhibition of milk fat synthesis by CLA.

Project description:Milk oligosaccharides (MOs) can be prebiotic and antiadhesive, while fatty acids (MFAs) can be antimicrobial. Both have been associated with milk microbes or mammary gland inflammation in humans. Relationships between these milk components and milk microbes or inflammation have not been determined for cows and could help elucidate a novel approach for the dairy industry to promote desired milk microbial composition for improvement of milk quality and reduction of milk waste. We aimed to determine relationships among milk microbiota, MFAs, MOs, lactose, and somatic cell counts (SCC) from Holstein cows, using our previously published data. Raw milk samples were collected at three time points, ranging from early to late lactation. Data were analyzed using linear mixed-effects modeling and repeated-measures correlation. Unsaturated MFA and short-chain MFA had mostly negative relationships with potentially pathogenic genera, including Corynebacterium, Pseudomonas, and an unknown Enterobacteriaceae genus but numerous positive relationships with symbionts Bifidobacterium and Bacteroides. Conversely, many MOs were positively correlated with potentially pathogenic genera (e.g., Corynebacterium, Enterococcus, and Pseudomonas), and numerous MOs were negatively correlated with the symbiont Bifidobacterium. The neutral, nonfucosylated MO composed of eight hexoses had a positive relationship with SCC, while lactose had a negative relationship with SCC. One interpretation of these trends might be that in milk, MFAs disrupt primarily pathogenic bacterial cells, causing a relative increase in abundance of beneficial microbial taxa, while MOs respond to and act on pathogenic taxa primarily through antiadhesive methods. Further research is needed to confirm the potential mechanisms driving these correlations. IMPORTANCE Bovine milk can harbor microbes that cause mastitis, milk spoilage, and foodborne illness. Fatty acids found in milk can be antimicrobial and milk oligosaccharides can have antiadhesive, prebiotic, and immune-modulatory effects. Relationships among milk microbes, fatty acids, oligosaccharides, and inflammation have been reported for humans. To our knowledge, associations among the milk microbial composition, fatty acids, oligosaccharides, and lactose have not been reported for healthy lactating cows. Identifying these potential relationships in bovine milk will inform future efforts to characterize direct and indirect interactions of the milk components with the milk microbiota. Since many milk components are associated with herd management practices, determining if these milk components impact milk microbes may provide valuable information for dairy cow management and breeding practices aimed at minimizing harmful and spoilage-causing microbes in raw milk.
