Constructing and Forgetting Temporal Context in the Human Cerebral Cortex.

ABSTRACT: How does information from seconds earlier affect neocortical responses to new input? We found that when two groups of participants heard the same sentence in a narrative, preceded by different contexts, the neural responses of each group were initially different but gradually fell into alignment. We observed a hierarchical gradient: sensory cortices aligned most quickly, followed by mid-level regions, while some higher-order cortical regions took more than 10 seconds to align. What computations explain this hierarchical temporal organization? Linear integration models predict that regions that are slower to integrate new information should also be slower to forget old information. However, we found that higher-order regions could rapidly forget prior context. The data from the cortical hierarchy were instead captured by a model in which each region maintains a temporal context representation that is nonlinearly integrated with input at each moment, and this integration is gated by local prediction error.

Constructing temporal regulatory cascades in the context of development and cell differentiation.

Project description:Cell differentiation is a complex process orchestrated by sets of regulators precisely appearing at certain time points, resulting in regulatory cascades that affect the expression of broader sets of genes, ending up in the formation of different tissues and organ parts. The identification of stage-specific master regulators and the mechanism by which they activate each other is a key to understanding and controlling differentiation, particularly in the fields of tissue regeneration and organoid engineering. Here we present a workflow that combines a comprehensive general regulatory network based on binding site predictions with user-provided temporal gene expression data, to generate a a temporally connected series of stage-specific regulatory networks, which we call a temporal regulatory cascade (TRC). A TRC identifies those regulators that are unique for each time point, resulting in a cascade that shows the emergence of these regulators and regulatory interactions across time. The model was implemented in the form of a user-friendly, visual web-tool, that requires no expert knowledge in programming or statistics, making it directly usable for life scientists. In addition to generating TRCs the tool links multiple interactive visual workflows, in which a user can track and investigate further different regulators, target genes, and interactions, directing the tool along the way into biologically sensible results based on the given dataset. We applied the TRC model on two different expression datasets, one based on experiments conducted on human induced pluripotent stem cells (hiPSCs) undergoing differentiation into mature cardiomyocytes and the other based on the differentiation of H1-derived human neuronal precursor cells. The model was successful in identifying previously known and new potential key regulators, in addition to the particular time points with which these regulators are associated, in cardiac and neural development.

Constructing scenes from objects in human occipitotemporal cortex.

Project description:We used functional magnetic resonance imaging (fMRI) to demonstrate the existence of a mechanism in the human lateral occipital (LO) cortex that supports recognition of real-world visual scenes through parallel analysis of within-scene objects. Neural activity was recorded while subjects viewed four categories of scenes and eight categories of 'signature' objects strongly associated with the scenes in three experiments. Multivoxel patterns evoked by scenes in the LO cortex were well predicted by the average of the patterns elicited by their signature objects. By contrast, there was no relationship between scene and object patterns in the parahippocampal place area (PPA), even though this region responds strongly to scenes and is believed to be crucial for scene identification. By combining information about multiple objects within a scene, the LO cortex may support an object-based channel for scene recognition that complements the processing of global scene properties in the PPA.

Temporal stability of the hemodynamic response function across the majority of human cerebral cortex.

Project description:The hemodynamic response function (HRF) measured with functional magnetic resonance imaging is generated by vascular and metabolic responses evoked by brief (<4 s) stimuli. It is known that the human HRF varies across cortex, between subjects, with stimulus paradigms, and even between different measurements in the same cortical location. However, our results demonstrate that strong HRFs are remarkably repeatable across sessions separated by time intervals up to 3 months. In this study, a multisensory stimulus was used to activate and measure the HRF across the majority of cortex (>70%, with lesser reliability observed in some areas of prefrontal cortex). HRFs were measured with high spatial resolution (2-mm voxels) in central gray matter to minimize variations caused by partial-volume effects. HRF amplitudes and temporal dynamics were highly repeatable across four sessions in 20 subjects. Positive and negative HRFs were consistently observed across sessions and subjects. Negative HRFs were generally weaker and, thus, more variable than positive HRFs. Statistical measurements showed that across-session variability is highly correlated to the variability across events within a session; these measurements also indicated a normal distribution of variability across cortex. The overall repeatability of the HRFs over long time scales generally supports the long-term use of event-related functional magnetic resonance imaging protocols.

Orbito-frontal cortex is necessary for temporal context memory.

Project description:Lesion and neuroimaging studies suggest that orbito-frontal cortex (OFC) supports temporal aspects of episodic memory. However, it is unclear whether OFC contributes to the encoding and/or retrieval of temporal context and whether it is selective for temporal relative to nontemporal (spatial) context memory. We addressed this issue with two complimentary studies: functional magnetic resonance imaging to measure OFC activity associated with successful temporal and spatial context memory during encoding and retrieval in healthy young participants, and a neuropsychological investigation to measure changes in spatial and temporal context memory in OFC lesion patients. Imaging results revealed that OFC contributed to encoding and retrieval of associations between objects and their temporal but not their spatial contexts. Consistent with this, OFC patients exhibited impairments in temporal but not spatial source memory accuracy. These results suggest that OFC plays a critical role in the formation and subsequent retrieval of temporal context.

Mapping the orbitofrontal cortex using temporal fluctuations in cerebral blood flow.

Project description:IntroductionThe orbitofrontal cortex (OFC) is involved in diverse cognitive and behavioral processes including incentive valuation, decision-making, and reinforcement learning. Anatomic and cytoarchitectonic studies divide the OFC along both medial-lateral and rostral-caudal axes. OFC regions diverge in structure and function, assessed in vivo using white matter tractography and blood oxygenation level-dependent (BOLD) MRI, respectively. However, interpretation of T2 *-weighted BOLD is limited by susceptibility artifacts in the inferior frontal lobes, with the spatial pattern of these artifacts frequently assuming the geometry of OFC organization. Here, we utilize a novel perfusion-weighted arterial spin labeling (ASL) functional connectivity approach, which is minimally susceptibility-weighted, to test the hypothesis that OFC topology reflects correlated temporal hemodynamic activity.MethodsIn healthy participants (n = 20; age = 29.5 ± 7.3), 3D ASL scans were acquired (TR/TE = 3,900/13 ms; spatial resolution = 3.8 mm isotropic). To evaluate reproducibility, follow-up scanning on a separate day was performed on a participant subset (n = 8). ASL-based connectivity was modeled for gray matter OFC voxels, and k-means clustering (k = 2-8) applied to correlation statistics.ResultsThese approaches revealed both medial-lateral and rostral-caudal OFC divisions, confirming our hypothesis. Longitudinal reproducibility testing revealed 84% voxel clustering agreement between sessions for the k = 2 solution.ConclusionTo our knowledge, this constitutes the first in vivo cortical parcellation based on perfusion fluctuations. Our approach confirms functional OFC subdivisions predicted from anatomy using a less susceptibility-sensitive method than the conventional approach.

Constructing autobiographical events within a spatial or temporal context: a comparison of two targeted episodic induction techniques.

Project description:Recalling and imagining autobiographical experiences involves constructing event representations within spatiotemporal contexts. We tested whether generating autobiographical events within a primarily spatial (where the event occurred) or temporal (the sequence of actions that occurred) context affected how the associated mental representation was constructed. We leveraged the well-validated episodic specificity induction (ESI) technique, known to influence the use of episodic processes on subsequent tasks, to develop variants that selectively enhance spatial or temporal processing. We tested the effects of these inductions on the details used to describe past and future autobiographical events. We first replicated the standard ESI effect, showing that ESI enhances generating episodic details, particularly those that are perception-based, when describing autobiographical events (Experiment 1). We then directly compared the effects of the spatial and temporal inductions (Experiment 2 and 3). When describing autobiographical events, spatial induction enhanced generating episodic details, specifically perception-based details, compared to the control or temporal inductions. A greater proportion of the episodic details generated after the temporal induction were gist-based than after the spatial induction, but this proportion did not differ from a control induction. Thus, using a spatial or temporal framework for autobiographical event generation alters the associated details that are accessed.

Stimulus context modulates competition in human extrastriate cortex.

Project description:When multiple stimuli appear simultaneously in the visual field, they are not processed independently, but rather interact in a mutually suppressive way, suggesting that they compete for neural representation in visual cortex. The biased competition model of selective attention predicts that the competition can be influenced by both top-down and bottom-up mechanisms. Directed attention has been shown to bias competition in favor of the attended stimulus in extrastriate cortex. Here, we show that suppressive interactions among multiple stimuli are eliminated in extrastriate cortex when they are presented in the context of pop-out displays, in which a single item differs from the others, but not in heterogeneous displays, in which all items differ from each other. The pop-out effects seemed to originate in early visual cortex and were independent of attentional top-down control, suggesting that stimulus context may provide a powerful influence on neural competition in human visual cortex.

Alleviating catastrophic forgetting using context-dependent gating and synaptic stabilization.

Project description:Humans and most animals can learn new tasks without forgetting old ones. However, training artificial neural networks (ANNs) on new tasks typically causes them to forget previously learned tasks. This phenomenon is the result of "catastrophic forgetting," in which training an ANN disrupts connection weights that were important for solving previous tasks, degrading task performance. Several recent studies have proposed methods to stabilize connection weights of ANNs that are deemed most important for solving a task, which helps alleviate catastrophic forgetting. Here, drawing inspiration from algorithms that are believed to be implemented in vivo, we propose a complementary method: adding a context-dependent gating signal, such that only sparse, mostly nonoverlapping patterns of units are active for any one task. This method is easy to implement, requires little computational overhead, and allows ANNs to maintain high performance across large numbers of sequentially presented tasks, particularly when combined with weight stabilization. We show that this method works for both feedforward and recurrent network architectures, trained using either supervised or reinforcement-based learning. This suggests that using multiple, complementary methods, akin to what is believed to occur in the brain, can be a highly effective strategy to support continual learning.

Separation of item and context in item-method directed forgetting.

Project description:Contextual information plays a critical role in directed forgetting (DF) of lists of items, whereas DF of individual items has been primarily associated with item-level processing. This study was designed to investigate whether context processing also contributes to the forgetting of individual items. Participants first viewed a series of words, with task-irrelevant scene images (used as "context tags") interspersed between them. Later, these words reappeared without the scenes and were followed by an instruction to remember or forget that word. Multivariate pattern analyses of fMRI data revealed that the reactivation of context information associated with the studied words (i.e., scene-related activity) was greater whereas the item-related information diminished after a forget instruction compared to a remember instruction. Critically, we found the magnitude of the separation between item information and context information predicted successful forgetting. These results suggest that the unbinding of an item from its context may support the intention to forget, and more generally they establish that contextual processing indeed contributes to item-method DF.
