The first record on Giardia muris from mice in Turkey.

ABSTRACT: Giardia muris has been reported from both laboratory and wild rodents in worldwide. During routine care, soft consistency was observed in feces of Swiss albino mice. Motile Giardia spp. trophozoites were observed microscopically on all fecal preparations after stained with Lugol's iodine solution. The trophozoites were broadly ovoid in a form with a pointed end. They possess two large nuclei and an adhesive disk whose length overlapped one-half of the trophozoite body length. The caudal flagella are unequal. β giardin (bg) gene region of isolates was successfully amplified and sequenced. According to sequence analyses of the bg gene region, the mice isolate was identified as G. muris and deposited in GenBank (Accession Number: MK675656). The bg sequence of G. muris was shown 99.3-99.7% identity with the isolates of G. muris reported from different countries in GenBank. We have firstly demonstrated G. muris trophozoites in the fecal samples of naturally infected Swiss albino mice in Turkey.

First record of Aphelenchoides stammeri (Nematoda: Aphelenchoididae) from Turkey.

Project description:In previous surveys for the investigation of Bursaphelenchus xylophilus in Turkey, several Bursaphelenchus species, except B. xylophilus, were detected. To determine the insect vectors of these previously reported Bursaphelenchus species, trap trees of each pine species (Pinus brutia and P. nigra) were cut and kept in the field from April to September in 2011. Aphelenchoides stammeri was isolated from wood chip samples of P. nigra from Isparta (Sütçüler). A. stammeri was first identified by using morphological features and allometric criteria of females and males. In addition, molecular analysis using sequences of 18S and 28S ribosomal (rRNA) genes of A. stammeri was performed. Delayed development of the protective IL-17A response following a Giardia muris infection in neonatal mice.

Project description:Giardia is an intestinal protozoan parasite that has the ability to infect a wide range of hosts, which can result in the clinical condition 'giardiasis'. Over the years, experimental research has shown the crucial involvement of IL-17A to steer the protective immune response against Giardia. The development of the protective response, as reflected by a significant drop in cyst secretion, typically takes around 3 to 4 weeks. However, early-life infections often have a more chronic character lasting for several weeks or months. Therefore, the aim of the current study was to investigate the dynamics of a Giardia muris infection and the subsequent host immune response in neonatal mice infected 4 days after birth. The outcome of the study showed that a G. muris infection in pre-weaned mice failed to trigger a protective IL-17A response, which could explain the prolonged course of infection in comparison to older mice. Only after weaning, a protective intestinal immune response started to develop, characterized by an upregulation of IL-17A and Mbl2 and the secretion of parasite-specific IgA.

Identification of variant-specific surface proteins in Giardia muris trophozoites.

Project description:Giardia lamblia undergoes antigenic variation, a process that might allow the parasite to evade the host's immune response and adapt to different environments. Here we show that Giardia muris, a related species that naturally infects rodents, possesses multiple variant-specific surface proteins (VSPs) and expresses VSPs on its surface, suggesting that it undergoes antigenic variation similar to that of G. lamblia.

Gene expression in IL-17RA-deficient and wild-type mice following Giardia muris infection

Project description:To elucidate IL-17A-dependent immune mechanims involved in regulating host defense, we employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential of regulating protective immunity that failed to be upregulated in the absence of IL-17RA signaling. Whole small intestine tissue collected from IL-17RA-deficient and C57BL/6 (wild-type) mice was collected 2 weeks after Giardia muris infection and from uninfected controls. Expression levels of several genes that may have anti-parasitic potential (Defb1, Retnlb, Saa1, Saa2) and may be involved in parasite clearance were, on average, lower or unchanged in IL-17RA deficient compared to wild-type mice after infection.

First record of Graphosoma inexpectatum (Hemiptera, Pentatomidae, Podopinae) from Turkey with description of the female.

Project description:Graphosoma inexpectatum Carapezza & Jindra, 2008 is described from Syria, the southern neighbor of Turkey, and is known only from the type locality. The first observation of the species in Turkey dates back to 1995 with two females obtained from the provinces of Gaziantep (Şehitkamil-Aktoprak) and Adana (Pozantı-Bürücek Plateau). These two localities are situated inside the part of the Mediterranean region along the Syrian border. Females of the species, whose original description was based on males, are described here for the first time. A map showing the collecting localities and photographs of the female specimens are given.

Giardia muris infection in mice is associated with a protective interleukin 17A response and induction of peroxisome proliferator-activated receptor alpha.

Project description:The protozoan parasite Giardia duodenalis (Giardia lamblia) is one of the most commonly found intestinal pathogens in mammals, including humans. In the current study, a Giardia muris-mouse model was used to analyze cytokine transcription patterns and histological changes in intestinal tissue at different time points during infection in C57BL/6 mice. Since earlier work revealed the upregulation of peroxisome proliferator-activated receptors (PPARs) in Giardia-infected calves, a second aim was to investigate the potential activation of PPARs in the intestines of infected mice. The most important observation in all mice was a strong upregulation of il17a starting around 1 week postinfection. The significance of interleukin 17A (IL-17A) in orchestrating a protective immune response was further demonstrated in an infection trial or experiment using IL-17 receptor A (IL-17RA) knockout (KO) mice: whereas in wild-type (WT) mice, cyst secretion dropped significantly after 3 weeks of infection, the IL-17RA KO mice were unable to clear the infection. Analysis of the intestinal response further indicated peroxisome proliferator-activated receptor alpha (PPARα) induction soon after the initial contact with the parasite, as characterized by the transcriptional upregulation of ppara itself and several downstream target genes such as pltp and cpt1. Overall, PPARα did not seem to have any influence on the immune response against G. muris, since PPARα KO animals expressed il-17a and could clear the infection similar to WT controls. In conclusion, this study shows for the first time the importance of IL-17 production in the clearance of a G. muris infection together with an early induction of PPARα. The effect of the latter, however, is still unclear.

First Record of Steinernema kraussei (Rhabditida: Steinernematidae) from Turkey and Its Virulence against Agrotis segetum (Lepidoptera: Noctuidae).

Project description:During a survey of entomopathogenic nematodes (EPNs) in the eastern Black Sea region of Turkey in 2009-2012, a steinernematid species was recorded and isolated using the Galleria-baiting method. The isolate was identified as Steinernema kraussei based on its morphological and molecular properties. The analysis of the ITS rDNA sequence placed the Turkish population of S. kraussei in the "feltiae-kraussei" group in the clade that contains different isolates of the species. This is the first record of S. kraussei from Turkey. The efficacy of S. kraussei was tested on Agrotis segetum (Lepidoptera: Noctuidea) larvae at different densities (100, 300, and 500 infective juveniles (IJs) g(-1) dry sand ) in laboratory conditions at 25 °C. The highest mortality (98%) was obtained with 500 IJs g(-1) dry sand within 7 d after inoculation. Our results indicate that the new isolate is a highly promising biological control agent against A. segetum, one of the most serious soil pests of agricultural area and fruits worldwide.
