A Review of the Effect of Formic Acid and Its Salts on the Gastrointestinal Microbiota and Performance of Pigs.

ABSTRACT: Out of the alternatives to antibiotics and zinc oxide, organic acids, or simply acidifiers, play significant roles, especially in ensuring gut health and the growth performance of pigs. Regarding acidifiers, formic acid and its salts have shown very promising results in weaning, growing and finishing pigs. Although it is known that the main mechanisms by which acidifiers can improve livestock performance and health are related to the regulation of gastrointestinal pH, an improvement in intestinal digestibility and mineral utilization, and their antimicrobial properties against specific pathogens has been observed, while poor consensus remains in relation to the effect of acidifers on bacteria and the complex microbiome. Therefore, the aim of the present review was to critically evaluate the effects of formic acid and its salts on the performance and the gastrointestinal microbiota balance of pigs.

Project description:The choice of the calcium (Ca) source in pig diets and the addition of formic acid may affect the gastrointestinal inositol phosphate (InsP) degradation and thereby, phosphorus (P) digestibility in pigs. This study assessed the effects of different Ca sources (Ca carbonate, Ca formate), exogenous phytase, and chemical acidification on InsP degradation, nutrient digestion and retention, blood metabolites, and microbiota composition in growing pigs. In a randomized design, 8 ileal-cannulated barrows (24 kg initial BW) were fed 5 diets containing Ca formate or Ca carbonate as the only mineral Ca addition, with or without 1,500 FTU/kg of an exogenous hybrid 6-phytase. A fifth diet was composed of Ca carbonate with phytase but with 8 g formic acid/kg diet. No mineral P was added to the diets. Prececal InsP6 disappearance and P digestibility were lower (P ≤ 0.032) in pigs fed diets containing Ca formate. In the presence of exogenous phytase, InsP5 and InsP4 concentrations in the ileal digesta were lower (P ≤ 0.019) with Ca carbonate than Ca formate. The addition of formic acid to Ca carbonate with phytase diet resulted in greater (P = 0.027) prececal InsP6 disappearance (87% vs. 80%), lower (P = 0.001) InsP5 concentration, and greater (P ≤ 0.031) InsP2 and myo-inositol concentrations in the ileal digesta. Prececal P digestibility was greater (P = 0.004) with the addition of formic acid compared to Ca carbonate with phytase alone. Prececal amino acid (AA) digestibility of some AA was greater with Ca formate compared to Ca carbonate but only in diets with phytase (P ≤ 0.048). The addition of formic acid to the diet with Ca carbonate and phytase increased (P ≤ 0.006) the prececal AA digestibility of most indispensable AA. Exogenous phytase affected more microbial genera in the feces when Ca formate was used compared to Ca carbonate. In the ileal digesta, the Ca carbonate diet supplemented with formic acid and phytase led to a similar microbial community as the Ca formate diets. In conclusion, Ca formate reduced prececal InsP6 degradation and P digestibility, but might be of advantage in regard to prececal AA digestibility in pigs compared to Ca carbonate when exogenous phytase is added. The addition of formic acid to Ca carbonate with phytase, however, resulted in greater InsP6 disappearance, P and AA digestibility values, and changed ileal microbiota composition compared to Ca carbonate with phytase alone.

Project description:Grape pomace (GP), a by-product in wine production, is nutritious and can be used as a feed ingredient for ruminants; however, its role in shaping sheep gastrointestinal tract (GIT) microbiota is unclear. We conducted a controlled trial using a randomized block design with 10 Tan lambs fed a control diet (CD) and 10 Tan lambs fed a pelleted diet containing 8% GP (dry matter basis) for 46 days. Rumen, jejunum, cecum, and colon bacterial and archaeal composition were identified by 16S rRNA gene sequencing. Dry matter intake (DMI) was greater (p < 0.05) in the GP than CD group; however, there was no difference in average daily gain (ADG, p < 0.05) and feed conversion ratio (FCR, p < 0.05) between the two groups. The GP group had a greater abundance of Prevotella 1 and Prevotella 7 in the rumen; of Sharpe, Ruminococcaceae 2, and [Ruminococcus] gauvreauii group in the jejunum; of Ruminococcaceae UCG-014 and Romboutsia in the cecum, and Prevotella UCG-001 in the colon; but lesser Rikenellaceae RC9 gut group in the rumen and cecum, and Ruminococcaceae UCG-005 and Ruminococcaceae UCG-010 in the colon than the CD group. The pathways of carbohydrate metabolism, such as L-rhamnose degradation in the rumen, starch and glycogen degradation in the jejunum, galactose degradation in the cecum, and mixed acid fermentation and mannan degradation in the colon were up-graded; whereas, the pathways of tricarboxylic acid (TCA) cycle VIII, and pyruvate fermentation to acetone in the rumen and colon were down-graded with GP. The archaeal incomplete reductive TCA cycle was enriched in the rumen, jejunum, and colon; whereas, the methanogenesis from H2 and CO2, the cofactors of methanogenesis, including coenzyme M, coenzyme B, and factor 420 biosynthesis were decreased in the colon. The study concluded that a diet including GP at 8% DM did not affect ADG or FCR in Tan lambs. However, there were some potential benefits, such as enhancing propionate production by microbiota and pathways in the GIT, promoting B-vitamin production in the rumen, facilitating starch degradation and amino acid biosynthesis in the jejunum, and reducing methanogenesis in the colon.

Project description:Phenolic acid like with the 3-caffeoylquini acid (3-CQA) is formed by caffeic acid and qunic acid. This study was conducted to explore the effect of 3-CQA on growth performance and intestinal functions in weaned pigs. A total of 180 weaned pigs were randomly allocated into five treatments with 6 replicate pens per treatment (6 pigs per pen). Pigs in the control group (CON) were fed with basal diet (BD), and the others in the experimental groups were fed with BD and supplemented with 12.5, 25, 50, and 100 mg/kg 3-CQA. On day 43, the blood sample-collected pigs in the CON and optimal-dose group (only based on growth performance) were picked, and housed in metabolism cages (a total of 12 pigs, N = 6). 3-CQA increased the feed efficiency from days 21 to 42 of the trial and throughout the trial (P < 0.05). 3-CQA increased the serum concentrations of total protein, albumin, and total cholesterol (P < 0.05). Moreover, 3-CQA supplementation at 25 mg/kg increased the apparent digestibility of DM, energy, and ash (P < 0.05). Interestingly, 3-CQA decreased the crypt depth but increased the ratio of villus height to crypt depth in the jejunum and ileum (P < 0.05). Moreover, 3-CQA also increased the activities of sucrase, lactase, and catalase in the jejunal mucosa, and increased the activities of alkaline phosphatase and superoxide dismutase in the ileal mucosa (P < 0.05). 3-CQA also increased the abundance of secretory immunoglobulin A in the ileal mucosa (P < 0.05). Importantly, 3-CQA not only elevated the expression levels of critical functional genes such as the zonula occludens-1 , occludin, solute carrier family 7 , and nuclear factor erythroid 2-related factor 2 (Nrf2) in the duodenum but also elevated the expression levels of divalent metal transporter-1 and Nrf2 in the jejunum (P < 0.05). These results suggested a positive effect of 3-CQA supplementation on the growth and intestinal functions of weaned pigs. The mechanisms of action may be associated with elevated anti-oxidant capacity and improved intestinal barrier functions.
