Upscale Design, Process Development, and Economic Analysis of Industrial Plants for Nanomagnetic Particle Production for Environmental and Biomedical Use.

ABSTRACT: Very few economical and process engineering studies have been made concerning the scale-up and implementation of nanomagnetic particle manufacturing into a full-scale plant, and determination of its viability. In this work we describe such a study for two types of industrial plants, one for manufacturing magnetic particles for applications in the environmental area, and the other for manufacturing nanomagnetic particles for applications in the biotechnology area; the two different applications are compared. The following methodology was followed: establish the manufacturing process for each application; determine the market demand of the product (magnetic nanoparticles) for both applications; determine the production capacity of each plant; engineer all the manufacturing process, determining all the process units and performing all the mass and energy balances for both plants; scale-up the main equipment; and determine the global economic impact and profitability. At the end both plants are found to be technologically and economically viable, the characteristics of the final products being, however, quite different, as well as the process engineering, economic analysis, and scale-up.

Project description:The objective of this work was to identify industrial scenarios for the most promising microalgal biorefinery value chains on the basis of product selection, yields, and techno-economic performance, using biological characteristics of algae species. The development, value creation, and validation of several new processing routes with applications in food, aquafeeds and non-food products were particularly considered in this work. The techno-economic performance of various single product value chains (SP) and multiproduct value chains (MP) was evaluated for four industrial microalgal strains. Cost-revenue optimization was done for a 10 kton microalgal dry weight y-1 simulated biorefinery plant, using flow sheeting software for equipment sizing, mass and energy flow modeling, and subsequent techno-economic evaluation. Data on yield, material and energy consumption were based on pre- and pilot size production plants (TRL 5-6). Revenue optimization was accomplished by first analyzing the performance of single product value chains of the microalgal strains. Subsequently, a strategy was developed to exploit almost all biomass based on the most promising microalgal strains. The cultivation costs are most of the time the major costs of the value chains. For the single product value chains common process bottlenecks are low product yields, especially for soluble proteins where only a small fraction of the biomass is leading to economic value. The biorefinery costs (excluding cultivation) vary significantly for various species, due to the species-specific operating conditions as well as differences in product yields. For the evaluated single product value chain scenarios the costs for utilities and other inputs were in general the highest contributing expenses. A biorefinery approach significantly increases the biomass utilization potential to marketable products from 7-28% to more than 97%. Although the cascading approach increases the total production costs of the multiproduct value chains significantly, this is more than compensated by the increased overall biomass revenue. For all selected multiproduct chains there is a significant potential to become profitable at a relevant industrial scale of 10 kton per year. Additional insights in the product functionality, quality, and their market size are needed to narrow down the wide range of foreseen product revenues and resulting profits.

Project description:BackgroundVirus-like particles (VLP) have an increasing range of applications including vaccination, drug delivery, diagnostics, gene therapy and nanotechnology. These developments require large quantities of particles that need to be obtained in efficient and economic processes. Production of VLP in yeast is attractive, as it is a low-cost protein producer able to assemble viral structural proteins into VLP. However, to date only single-layered VLP with simple architecture have been produced in this system. In this work, the first steps required for the production of rotavirus-like particles (RLP) in S. cerevisiae were implemented and improved, in order to obtain the recombinant protein concentrations required for VLP assembly.ResultsThe genes of the rotavirus structural proteins VP2, VP6 and VP7 were cloned in four Saccharomyces cerevisiae strains using different plasmid and promoter combinations to express one or three proteins in the same cell. Performance of the best constructs was evaluated in batch and fed-batch cultures using a complete synthetic media supplemented with leucine, glutamate and succinate. The strain used had an important effect on recombinant protein concentration, while the type of plasmid, centromeric (YCp) or episomal (YEp), did not affect protein yields. Fed-batch culture of the PD.U-267 strain resulted in the highest concentration of rotavirus proteins. Volumetric and specific productivities increased 28.5- and 11-fold, respectively, in comparison with batch cultures. Expression of the three rotavirus proteins was confirmed by immunoblotting and RLP were detected using transmission electron microscopy.ConclusionsWe present for the first time the use of yeast as a platform to express multilayered rotavirus-like particles. The present study shows that the combined use of molecular and bioprocess tools allowed the production of triple-layered rotavirus RLP. Production of VLP with complex architecture in yeasts could lead to the development of new vaccine candidates with reduced restrictions by regulatory agencies, using the successful experience with other yeast-based VLP vaccines commercialized worldwide.

Project description:Heavy metal-organic pollutants compound pollution at industrial legacy sites and have caused damage to the ecological environment and human health during recent decades. In view of the difficulty and high cost of post-contamination remediation, it is worth studying, and practically applying, cutoff walls to reduce the spread of pollution in advance. In this study, field-scale studies were carried out at e-waste dismantling legacy sites in Taizhou, Zhejiang Province of China, through the process of site investigation, numerical simulation, and cutoff wall practical application. Firstly, the concentrations and spatial distributions of Pb, Cd and polychlorinated biphenyls (PCBs) and poly brominated diphenyl ethers (PBDEs) were identified in both soil and groundwater. Then, potential dispersal routes of key combined contaminants (Pb and PCBs) at the soil-groundwater interface were systematically studied through numerical simulation applying Visual MODFLOW-MT3DMS. One site was chosen to predict the barrier effect of differently sized cutoff walls based on the migration path of compound pollutants. A protocol for a cutoff wall (50 m length × 2 m width × 3 m height) was finally verified and applied at the real contaminated site for the blocking of compound pollutant diffusion. Further, the groundwater quality of the contaminated site was monitored consecutively for six months to ensure the durability and stability of barrier measures. All pollutant indicators, including for Pb and PCB complex pollutants, were reduced to below the national Grade IV groundwater standard value, achieving environmental standards at these polluted sites and providing possibilities for land reuse. In summary, this field-scale test provided new ideas for designing cutoff walls to block the diffusion of complex pollutants; it also laid a basis for the practical application of cutoff walls in pollution prevention and control of complex contaminated sites and for soil-groundwater environmental protection at industrial heritage sites.
