ABSTRACT: Kenaf is an annual crop that is widely cultivated as a source of bast (phloem) fibres, the phytoremediation of heavy metal-contaminated farmlands and textile-relevant compounds. Leaf shape played a unique role in kenaf improvement, due to the inheritance as a single locus and the association with fibre development in typical lobed-leaf varieties. Here we report a high-quality genome assembly and annotation for var. 'Fuhong 952' with 1078 Mbp genome and 66 004 protein-coding genes integrating single-molecule real-time sequencing, a high-density genetic map and high-throughput chromosome conformation capture techniques. Gene mapping assists the identification of a homeobox transcription factor LATE MERISTEM IDENTITY 1 (HcLMI1) gene controlling lobed-leaf. Virus-induced gene silencing (VIGS) of HcLMI1 in a lobed-leaf variety was critical to induce round (entire)-like leaf formation. Candidate genes involved in cell wall formation were found in quantitative trait loci (QTL) for fibre yield and quality-related traits. Comparative genomic and transcriptome analyses revealed key genes involved in bast fibre formation, among which there are twice as many cellulose synthase A (CesA) genes due to a recent whole-genome duplication after divergence from Gossypium. Population genomic analysis showed two recent population bottlenecks in kenaf, suggesting domestication and improvement process have led to an increase in fibre biogenesis and yield. This chromosome-scale genome provides an important framework and toolkit for sequence-directed genetic improvement of fibre crops.

Project description:Kenaf (Hibiscus cannabinus L.) is a natural fibre crop that can be used for a variety of purposes and has various applications in industry. Despite this, its potential has not been fully exploited because of low yields and a narrow genetic base, limiting hybrids' development. Based on this background, eight kenaf mutants and one commercial cultivar were selected and crossed in a half-diallel for general and specific combining abilities (GCA and SCA) to get the desired results done in this investigation. The 36 hybrid offspring and their parental lines were tested in the field over two environments. Diallel results based on Griffing B method 2 indicated significant differences for all characters studied except for GCA in top diameter and plant height and top diameter SCA, indicating the existence of both additive and nonadditive gene actions for the inheritance of the traits. The amplitude of GCA variation was much higher than that of SCA variation for all parameters except top diameter and node number, showing the additive gene's prevalence and the likelihood of genetic advancement through selection. In both conditions, Hayman and Jinks graphical studies demonstrated that partial dominance controlled various fibre yield component parameters such as plant height, middle diameter, stick weight, and fibre weight. On the other hand, fibre yield and the majority of physical features indicated either dominance or overdominance gene action. Plant height, base diameter, core diameter, middle diameter, fresh stem weight, and stick weight all strongly positively correlated with fibre yield. These traits also had a higher proportion of additive effects, a moderate narrow-sense heritability, and a higher baker ratio, indicating successful indirect selection for fibre yield. The parents P1, P3, and P4 had the most dominant alleles for most of the features, while the parents P2, P7, and P9 had the most recessive alleles. The hybrids P1 × P4, P1 × P9, P2 × P3, P2 × P5, P4 × P6, P4 × P7, P4 × P9, P5 × P8, and P7 × P9 outperformed the parents in terms of heterotic responses and showed that they have a lot of genetic potential for kenaf enhancement in tropical climates.

Project description:This study evaluates the coagulation performance of kenaf protein fractions (KPFs) comprising of albumin (AlbKP), globulin (GloKP), prolamin (ProKP), and glutenin (GluKP), in the treatment of high (500 NTU), medium (150 NTU), and low (30 NTU) turbidity water. Based on preliminary experimental results, the study focused on GloKP due to it outperforming the other kenaf coagulation products (KCPs) in all water types tested. The influence of GloKP, both as a primary coagulant and coagulant aid to aluminum sulfate (AS) on organic matter removal, was examined. Parametric analysis on turbidity, TSS, pH, dosages, retention time, and KPFs storage time was completed. Results indicated that GloKP could be used both as a primary coagulant and coagulant aid. GloKP had a higher turbidity and solids removal than the AlbKP and other KPFs (ProKP and GluKP). Solution pH greatly influenced the performance of the GloKP, and optimum dosage at pH 2 resulted in the highest organic matter removal. High dosages also resulted in negative mobility of particles and a more stable suspension. When used as a coagulant aid to AS, GloKP was more effective in removing dissolved organic carbon (DOC). Scanning electron microscopy elemental analysis (SEM-EDAX) and Fourier transform infrared (FT-IR) spectra showed the structure of the KPFs. SEM-EDAX indicated the presence of metal cations capable of forming complexes essential for flocs formation. The enhanced floc formation, detailed in this paper, is ascribed to the collective effect of charge neutralization of the AS species and the adsorption and bridging effect of the GloKP, which improves the bonds formed between flocs. The coagulation-flocculation process can be significantly improved using dual coagulants. GloKP was also an excellent alternative to its crude (CrKP) and solvent extract (HxKP) form for removing suspended and dissolved particles from all water types. PRACTITIONER POINTS: Kenaf protein fractionates can destabilize stable particles. The globulin protein fractionate (GloKP) aggregated the most particles and contained least dissolved organic material. GloKP is pH sensitive with pH 2 reported as best working pH. Coagulant dosage and coagulation mechanism were assessed.
