Non-Invasive Skin Cancer Diagnosis Using Hyperspectral Imaging for In-Situ Clinical Support.

ABSTRACT: Skin cancer is one of the most common forms of cancer worldwide and its early detection its key to achieve an effective treatment of the lesion. Commonly, skin cancer diagnosis is based on dermatologist expertise and pathological assessment of biopsies. Although there are diagnosis aid systems based on morphological processing algorithms using conventional imaging, currently, these systems have reached their limit and are not able to outperform dermatologists. In this sense, hyperspectral (HS) imaging (HSI) arises as a new non-invasive technology able to facilitate the detection and classification of pigmented skin lesions (PSLs), employing the spectral properties of the captured sample within and beyond the human eye capabilities. This paper presents a research carried out to develop a dermatological acquisition system based on HSI, employing 125 spectral bands captured between 450 and 950 nm. A database composed of 76 HS PSL images from 61 patients was obtained and labeled and classified into benign and malignant classes. A processing framework is proposed for the automatic identification and classification of the PSL based on a combination of unsupervised and supervised algorithms. Sensitivity and specificity results of 87.5% and 100%, respectively, were obtained in the discrimination of malignant and benign PSLs. This preliminary study demonstrates, as a proof-of-concept, the potential of HSI technology to assist dermatologists in the discrimination of benign and malignant PSLs during clinical routine practice using a real-time and non-invasive hand-held device.

Project description:Wheat stripe rust (WSR), a fungal disease capable of inflicting severe crop loss, threatens most of global wheat production. Breeding for genetic resistance is the primary defense against stripe rust infection. Further development of rust-resistant wheat varieties depends on the ability to accurately and rapidly quantify rust resilience. In this study we demonstrate the ability of visible through shortwave infrared reflectance spectroscopy to effectively provide high-throughput classification of wheat stripe rust severity and identify important spectral regions for classification accuracy. Random forest models were developed using both leaf-level and canopy-level hyperspectral reflectance observations collected across a breeding population that was scored for WSR severity using 10 and 5 severity classes, respectively. The models were able to accurately diagnose scored disease severity class across these fine scoring scales between 45-52% of the time, which improved to 79-96% accuracy when allowing scores to be off-by-one. The canopy-level model demonstrated higher accuracy and distinct spectral characteristics relative to the leaf-level models, pointing to the use of this technology for field-scale monitoring. Leaf-level model performance was strong despite clear variation in scoring conducted between wheat growth stages. Two approaches to reduce predictor and model complexity, principal component dimensionality reduction and backward feature elimination, were applied here. Both approaches demonstrated that model classification skill could remain high while simplifying high-dimensional hyperspectral reflectance predictors, with parsimonious models having approximately 10 unique components or wavebands. Through the use of a high-resolution infection severity scoring methodology this study provides one of the most rigorous tests of the use of hyperspectral reflectance observations for WSR classification. We demonstrate that machine learning in combination with a few carefully-selected wavebands can be leveraged for precision remote monitoring and management of WSR to limit crop damage and to aid in the selection of resilient germplasm in breeding programs.

Project description:BackgroundOral microbiome dysbiosis is an important risk factor affecting the occurrence and progression of coronary artery disease (CAD). However, the dysbiosis on the tongue in patients with CAD is still unclear, and whether the oral alteration caused by these disorders can be identified by other tools for CAD diagnosis needs to be further explored. Hyperspectral imaging (HSI) is characterized as high spectral resolution, broad spectral range, and superior spatial resolution. Hyperspectral images contain high-dimensional data that generally require machine learning algorithms for feature identification and model construction. Therefore, this study aims to investigate the variation of tongue microbiota and the effectiveness of HSI models in CAD diagnosis.MethodsBetween 2023 and 2024, we prospectively approached 276 patients with chest pain and exhibiting risk for CAD who underwent coronary artery angiography (CAG). And 190 patients were enrolled in this study. Tongue dorsum swabs were collected for subsequent 16sRNA sequencing and microbiome analysis. Tongue dorsum features were extracted from hyperspectral images. The HSI analysis incorporated a total of 4750 hyperspectral images from all patients. All images are divided into training set (N = 2555), internal test set (N = 1095) and external test set (N = 1095). A total of 31 models were constructed. 30 single machine learning algorithms were used to construct and test the CAD prediction models. Furthermore, the best performing fusion model was established. The efficacy of the model was evaluated employing several metrics, including area under the curve (AUC), decision curve analysis (DCA), calibration curve, accuracy (ACC), sensitivity (SE), specificity (SP), positive predictive value (PPV), negative predictive value (NPV) and F1 score.ResultsThe 16sRNA sequencing results indicated significant dysbiosis in the oral microbiota of patients with CAD, with decreased microbial abundance, network complexity and stability. The fusion model (GP-GB-SVM) demonstrated the highest performance, achieving an AUC of 0.92, ACC of 0.82, SE of 0.70, SP of 0.92, PPV of 0.88 and NPV of 0.79 in the internal test set and AUC of 0.86, ACC of 0.70, SE of 0.90, SP of 0.46, PPV of 0.60 and NPV of 0.90 in the external test set.ConclusionThese findings not only emphasize the significant alteration of microbiome colonized on the tongue dorsum in CAD patients but also demonstrate the tongue features associated with microbiome dysbiosis can be identified in hyperspectral images. Thereby the integration of HSI and machine learning provides novel insights into non-invasive diagnosis of CAD.

Project description:IntroductionMelanoma diagnosis traditionally relies on microscopic examination of hematoxylin and eosin (H&E) slides by dermatopathologists to search for specific architectural and cytological features. Unfortunately, no single molecular marker exists to reliably differentiate melanoma from benign lesions such as nevi. This study explored the potential of autofluorescent molecules within tissues to provide molecular fingerprints indicative of degenerated melanocytes in melanoma.MethodsUsing hyperspectral imaging (HSI) and spectral phasor analysis, we investigated autofluorescence patterns in melanoma compared to intradermal nevi. Using UV excitation and a commercial spectral confocal microscope, we acquired label-free HSI data from the whole-slice samples.ResultsOur findings revealed distinct spectral phasor distributions between melanoma and intradermal nevi, with melanoma displaying a broader phasor phase distribution, signifying a more heterogeneous autofluorescence pattern. Notably, longer wavelengths associated with larger phases correlated with regions identified as melanoma by expert dermatopathologists using H&E staining. Quantitative analysis of phase and modulation histograms within the phasor clusters of five melanomas (with Breslow thicknesses ranging from 0.5 mm to 6 mm) and five intradermal nevi consistently highlighted differences between the two groups. We further demonstrated the potential for the discrimination of several melanocytic lesions using center-of-mass comparisons of phase and modulation variables. Remarkably, modulation versus phase center of mass comparisons revealed strong statistical significance among the groups. Additionally, we identified the molecular endogenous markers responsible for tissue autofluorescence, including collagen, elastin, NADH, FAD, and melanin. In melanoma, autofluorescence is characterized by a higher phase contribution, indicating an increase in FAD and melanin in melanocyte nests. In contrast, NADH, elastin, and collagen dominate the autofluorescence of the nevus.DiscussionThis work underscores the potential of autofluorescence and HSI-phasor analysis as valuable tools for quantifying tissue molecular fingerprints, thereby supporting more effective and quantitative melanoma diagnosis.
