Three new Scheffersomyces species associated with insects and rotting wood in China.

ABSTRACT: Three species of Scheffersomyces were identified during a diversity study of yeasts. All three are associated with insects and rotting wood in China. Phylogenetic analyses of a genomic dataset combining ITS and nrLSU revealed that these new collections are distinct from known species, thus three new species are introduced i.e. S. jinghongensis, S. paraergatensis, and S. anoplophorae. In our phylogenetic analyses, Scheffersomyces jinghongensis possesses a strong independent lineage and is closely related to S. titanus. S. paraergatensis is closely related to S. ergatensis, while S. anoplophorae is related to S. stambukii. Several differences in physiological traits and molecular data indicate that S. jinghongensis, S. paraergatensis, and S. anoplophorae are three newly identified species.

New species of Yamadazyma from rotting wood in China.

Project description:Yamadazyma is one of the largest genera in the family Debaryomycetaceae (Saccharomycetales, Saccharomycetes) with species mainly found in rotting wood, insects and their resulting frass, but also recovered from flowers, leaves, fruits, tree bark, mushrooms, sea water, minerals, and the atmosphere. In the present study, several strains obtained from rotting wood in Henan and Yunnan Provinces of China were isolated. Based on morphology and a molecular phylogeny of the rDNA internal transcribed spacer region (ITS) and the D1/D2 domain of the large subunit (LSU) rDNA, these strains were identified as three new species: Yamadazymaluoyangensis, Y.ovata and Y.paraaseri; and three previously described species, Y.insectorum, Y.akitaensis, and Y.olivae. The three new species are illustrated and their morphology and phylogenetic relationships with other Yamadazyma species are discussed. Our results indicate a high undiscovered diversity of Yamadazyma spp. inhabiting rotting wood in China.

Diversity of the genus Sugiyamaella and description of two new species from rotting wood in China.

Project description:Species of the genus Sugiyamaella (Trichomonascaceae, Saccharomycetales), found in rotting wood in China, were investigated using morphology and the molecular phylogeny of a combined ITS and nrLSU dataset. Nine taxa were collected in China: two were new species (viz. Sugiyamaella chuxiong sp. nov. and S. yunanensis sp. nov.) and seven were known species, S. americana, S. ayubii, S. novakii, S. paludigena, S. valenteae, S. valdiviana and S. xiaguanensis. The two new species are illustrated and their morphology and phylogenetic relationships with other Sugiyamaella species are discussed. Our results indicate a potentially great diversity of Sugiyamaella spp. inhabiting rotting wood in China just waiting to be discovered.

Description of Scheffersomyces henanensis sp. nov., a new D-xylose-fermenting yeast species isolated from rotten wood.

Project description:Two strains of a D-xylose-fermenting yeast species were isolated from rotten wood samples collected from the Baotianman Nature Reserve in Henan Province, central China. These strains formed hat-shaped ascospores in conjugated and deliquescent asci. Multilocus phylogenetic analysis that included the nearly complete small subunit (SSU), the internal transcribed spacer (ITS) region and the D1/D2 domain of the large subunit (LSU) rRNA genes, as well as RNA polymerase II largest subunit (RPB1) gene demonstrated that the two strains represent a novel yeast species closely related to Scheffersomyces segobiensis. A sequence comparison of xylose reductase (XYL1) gene, which was recently recommended for rapid identification of cryptic species in the Scheffersomyces clade, revealed a significant sequence divergence of 25 nucleotides between the novel strains and their closest relative S. segobiensis, supporting their classification as a distinct species. Furthermore, these new strains can be clearly distinguished from S. segobiensis by a number of morphological and physiological characteristics. Therefore, a novel yeast species, Scheffersomyces henanensis sp. nov., is proposed to accommodate these strains. The type strain is BY-41T ( = CICC 1974T = CBS 12475T).

Discovery of Three New Mucor Species Associated with Cricket Insects in Korea.

Project description:Species in the genus Mucor have a worldwide distribution and are isolated from various substrata and hosts, including soil, dung, freshwater, and fruits. However, their diversity from insects is still much too little explored. The aim of this study was to characterize three new species of Mucor: Mucor grylli sp. nov., M. hyangburmii sp. nov., and M. kunryangriensis sp. nov., discovered in Kunryang-ri, Cheongyang in the Chungnam Province of Korea, during an investigation of Mucorales from cricket insects. The new species are described using morphological characters and molecular data including ITS and LSU rDNA regions. Mucor grylli is characterized by the highly variable shape of its columellae, which are subglobose to oblong, obovoid, strawberry-shaped, and sometimes slightly or strongly constricted in the center. Mucor hyangburmii is characterized by the production of azygospores and growth at 40 °C. Mucor kunryangriensis is characterized by the variable shape of its columellae, which are elongated-conical, obovoid, cylindrical ellipsoid, cylindrical, and production of abundant yeast-like cells on PDA, MEA, and SMA media. Based on the sequence analysis of two genetic markers, our phylogenic assessment strongly supported M. grylli, M. hyangburmii, and M. kunryangriensis as new species. Detailed descriptions, illustrations, and phylogenetic trees are provided.

Molecular phylogeny and morphology reveal a new wood-rotting fungal species, Sistotrema yunnanense sp. nov. from the Yunnan-Guizhou Plateau.

Project description:Wood-rotting fungi are important components of woody plant ecosystems and play an active role in the decomposition and turnover of nutrients from wood, and are among the major groups of Basidiomycota. In this study, a new species of wood-rotting fungus, Sistotrema yunnanense, was proposed based on morphological characteristics and molecular evidence. It is characterized by resupinate basidiomata, a monomitic hyphal system having generative hyphae with clamp connections, suburniform to urniform basidia, and short-cylindrical to oblong ellipsoid basidiospores (4.5-6.5 × 3-4 µm). Phylogenetic analyses performed using the large subunit nuc rDNA indicated that S. yunnanense was nested within the genus Sistotrema s.l. of the family Hydnaceae, within the order Cantharellales.

Taxonomy and physiological characterisation of Scheffersomyces titanus sp. nov., a new D-xylose-fermenting yeast species from China.

Project description:Three strains of a d-xylose-fermenting yeast species were isolated from the host beetle Dorcus titanus collected from two different localities in Henan Province, Central China. These strains formed two hat-shaped ascospores in conjugated and deliquescent asci. Multilocus phylogenetic analysis that included the nearly complete small subunit (SSU), the internal transcribed spacer (ITS) region and the D1/D2 domains of the large subunit (LSU) rDNAs, as well as RNA polymerase II largest subunit (RPB1) gene demonstrated that these strains represent a novel yeast species belonging to the genus Scheffersomyces. The phylogenetic analysis based on the nucleotide sequences of the xylose reductase (XYL1) gene supported the view that the new strains could be grouped as a unique species. Although this new species is highly similar to Scheffersomyces stipitis-like yeasts in terms of nrDNA sequences and morphological and physiological characteristics, the species can be clearly differentiated from its close relatives on the basis of the sequences of XYL1 and RPB1. Therefore, a novel yeast species, Scheffersomyces titanus sp. nov., is proposed to accommodate these strains. The type strain is NYNU 14712(T) (CICC 33061(T) = CBS 13926(T)).

Three new microfungi (Ascomycota) species from southern China.

Project description:Apiospora, Microdochium and Pestalotiopsis have been reported as plant pathogens, endophytes or saprotrophes worldwide. Combining multiple molecular markers with morphological characteristics, this study proposes three new species, viz. Apiosporabambusigena, Microdochiumjianfenglingense and Pestalotiopsissolicola from southern China. Apiosporabambusigena and M.jianfenglingense were collected from bamboo in Hainan Province and P.solicola was collected from soil in Yunnan Province. The morphologically similar and phylogenetically closely-related species were compared.

Three new Diaporthe species from Shaanxi Province, China.

Project description:Diaporthe species (Sordariomycetes, Diaporthales) are often reported as important plant pathogens, saprobes and endophytes on a wide range of plant hosts. In this study, Diaporthe specimens were collected from symptomatic twigs and branches at the Huoditang Forest Farm in Shaanxi Province, China. Identification was done using a combination of morphology and comparison of DNA sequence data of the nuclear ribosomal internal transcribed spacer (ITS), calmodulin (cal), histone H3 (his3), partial translation elongation factor-1α (tef1) and β-tubulin (tub2) gene regions. Three new Diaporthe species are proposed: D. albosinensis, D. coryli and D. shaanxiensis. All species are illustrated and their morphology and phylogenetic relationships with other Diaporthe species are discussed.
