Decadal changes in blood δ¹³C values, at-sea distribution, and weaning mass of southern elephant seals from Kerguelen Islands.

ABSTRACT: Changes in the foraging environment and at-sea distribution of southern elephant seals from Kerguelen Islands were investigated over a decade (2004-2018) using tracking, weaning mass, and blood δ¹³C values. Females showed either a sub-Antarctic or an Antarctic foraging strategy, and no significant shift in their at-sea distribution was detected between 2004 and 2017. The proportion of females foraging in sub-Antarctic versus Antarctic habitats did not change over the 2006-2018 period. Pup weaning mass varied according to the foraging habitat of their mothers. The weaning mass of sub-Antarctic foraging mothers' pups decreased by 11.7 kg over the study period, but they were on average 5.8 kg heavier than pups from Antarctic foraging mothers. Pup blood δ¹³C values decreased by 1.1‰ over the study period regardless of their sex and the presumed foraging habitat of their mothers. Together, these results suggest an ecological change is occurring within the Indian sector of the Southern Ocean with possible consequences on the foraging performance of southern elephant seals. We hypothesize that this shift in δ¹³C is related to a change in primary production and/or in the composition of phytoplankton communities, but this requires further multidisciplinary investigations.

Project description:RationaleThe use of sulfur isotopes to study trophic ecology in marine ecosystems has increased in the past decade. Unlike other commonly used isotopes (e.g., carbon), sulfur can better discriminate benthic and pelagic productivity. However, how lipid extraction affects sulfur isotopic values has not been assessed, despite its frequent use to remove lipid effects on δ13 C values.MethodsWe used white muscle and liver samples from two species of sharks and skin samples from two species of pinnipeds (sea lion and fur seal) to assess the effects of lipid extraction on stable isotope values for δ34 S, δ13 C, and δ15 N. Isotopic values were determined using a continuous flow-isotope ratio mass spectrometer coupled to an elemental analyzer.ResultsLipid extraction significantly decreased δ34 S values in shark tissues, more so for liver than muscle (-4.6 ± 0.9‰ vs -0.8 ± 0.3‰, average change), with nearly no change in their standard deviations. Lipid extraction did not affect δ34 S values from pinniped skin samples (0.2 ± 0.8‰, average change). After lipid extraction, consistent increases in δ13 C values (0.2‰-7.3‰) were detected as expected, especially in tissue with high lipid content (C:N >4). After lipid extraction, significant increases in δ15 N values (0.5‰-1.4‰) were found in shark muscle and liver tissues. For pinniped skin samples, δ15 N values were not significantly lower after lipid extraction (-0.4‰ to -0.1‰).ConclusionsLipid extraction did not have a strong impact on δ34 S values of shark muscle and pinniped skin (≤1‰). However, our results suggest it is essential to consider the effects of lipid extraction when interpreting results from δ34 S values of shark liver tissue, as they significantly depleted values relative to bulk tissue (~5‰). This may reflect selective removal of sulfolipids and glutathione present in higher concentrations in the liver than in muscle and skin and requires further investigation.

Project description:Methods for identifying origin, movement, and foraging areas of animals are essential for understanding ecosystem connectivity, nutrient flows, and other ecological processes. Telemetric methods can provide detailed spatial coverage but are limited to a minimum body size of specimen for tagging. In recent years, stable isotopes have been increasingly used to track animal migration by linking landscape isotope patterns into movement (isoscapes). However, compared to telemetric methods, the spatial resolution of bulk stable isotopes is low. Here, we examined a novel approach by evaluating the use of compound-specific hydrogen and carbon stable isotopes of fatty acids (δ2HFA and δ13CFA) from fish liver, muscle, brain, and eye tissues for identifying site specificity in a 254 km2 sub-alpine river catchment. We analyzed 208 fish (European bullhead, rainbow trout, and brown trout) collected in 2016 and 2018 at 15 different sites. δ13CFA values of these fish tissues correlated more among each other than those of δ2HFA values. Both δ2HFA and δ13CFA values showed tissue-dependent isotopic fractionation, while fish taxa had only small effects. The highest site specificity was for δ13CDHA values, while the δ2H isotopic difference between linoleic acid and alpha-linolenic acid resulted in the highest site specificity. Using linear discrimination analysis of FA isotope values, over 90% of fish could be assigned to their location of origin; however, the accuracy dropped to about 56% when isotope data from 2016 were used to predict the sites for samples collected in 2018, suggesting temporal shifts in site specificity of δ2HFA and δ13CFA. However, the predictive power of δ2HFA and δ13CFA over this time interval was still higher than site specificity of bulk tissue isotopes for a single time point. In summary, compound-specific isotope analysis of fatty acids may become a highly effective tool for assessing fine and large-scale movement and foraging areas of animals.

Project description:With the increasing anthropogenic impacts on fish habitats, it has become more important to understand which primary resources sustain fish populations. This resource utilization can differ between fish life stages, and individuals can migrate between habitats in search of resources. Such lifetime information is difficult to obtain due to the large spatial and temporal scales of fish behavior. The otolith organic matrix has the potential to indicate this resource utilization and migration with δ13C values of essential amino acids (EAAs), which are a direct indication of the primary producers. In a proof-of-concept study, we selected the Acoupa weakfish, Cynoscion acoupa, as a model fish species with distinct ontogenetic migration patterns. While it inhabits the Brazilian mangrove estuaries during juvenile stages, it moves to the coastal shelf as an adult. Thus, we expected that lifetime resource utilization and migration would be reflected in δ13CEAA patterns and baseline values in C. acoupa otoliths. By analyzing the C. acoupa otolith edges across a size range of 12-119 cm, we found that baseline δ13CEAA values increased with size, which indicated an estuarine to coastal shelf distribution. This trend is highly correlated with inorganic δ13C values. The δ13CEAA patterns showed that estuarine algae rather than mangrove-derived resources supported the juvenile C. acoupa populations. Around the juvenile size of 40 cm, resource utilization overlapped with those of adults and mean baseline δ13CEAA values increased. This trend was confirmed by comparing otolith core and edges, although with some individuals potentially migrating over longer distances than others. Hence, δ13CEAA patterns and baseline values in otoliths have great potential to reconstruct ontogenetic shifts in resource use and habitats. The insight could aid in predictions on how environmental changes affect fish populations by identifying the controlling factors at the base of the food web.

Project description:RationaleThe carbon (δ13 C) and oxygen (δ18 O) isotope composition of siderite (FeCO3 ) is used widely to understand and quantify geochemical processes to reconstruct past climate and environmental change. However, few laboratories follow precisely the same protocol for the preparation and analysis of siderite-bearing materials, which combined with the absence of international reference materials and mineral-specific acid fractionation factors, leads potentially to significant differences in isotope data generated by individual laboratories. Here we examine procedures for the isotope analysis of siderite and discuss factors potentially contributing to inconsistencies in sample measurement data.MethodsIsotope analysis of siderite is first assessed using similar versions of the classical off-line, sealed vessel acid digestion method by comparing data sets obtained from intercomparison materials measured at two participating laboratories. We then compare data from the classical method against those generated using an automated preparation technique using data produced from an independent set of test materials.ResultsMeasurement of siderite δ13 C is generally both repeatable and reproducible, but measurement of δ18 O may be subject to large (~1‰), method-dependent bias for siderite reacted at differing temperatures (70°C and 100°C) under classical and automated CO2 preparation conditions. The potential for poor oxygen isotope measurement reproducibility is amplified by local differences in sample preparation protocols and procedures used to calibrate measurement data to international reference scales.ConclusionsWe offer suggestions for improving the repeatability and reproducibility of δ13 C and δ18 O analysis on siderite. The challenge of producing consistent isotope data from siderite can only be resolved by ensuring the availability of siderite reference materials to facilitate identical treatment as a basis for minimising method-dependent contributions to data inconsistency between laboratories.
