This work reports the measured and derived quantities of an experimental campaign conducted in an existing laundry to record the conditions of the exhaust air of a batch industrial natural gas-fired dryer for cotton flat fabrics, like bed linens or tablecloths. The load of fabrics is 85 kg, referred to the dry fabric. Project description:Today׳s engineering systems and machine are so sophisticated that mere energy analysis cannot accurately and reliably describe the thermodynamic behaviour, viz-a-viz the energy changes occurring in these complex systems. Hence, a more efficient and realistic parameter that provides us with useful information about thermodynamic losses and energy efficiency improvement potential is the exergy analyses. Fresh samples of onion fruits were washed with distilled water to remove particles and contaminants that can adversely affect the experimental results. Hence, 36.50 g of the sample at different thicknesses of 0.50 cm, 1.00 cm and 1.50 cm were taken into the cabinet dryer for drying at different temperatures of 65 °C, 75 °C,85 °C and 95 °C and the weight loss at each temperature and thickness was determined with the aid of a digital weighing balance. Hence, it was on this premise that the exergy analyses in terms of exergy loss, exergetic improvement potential and exergetic sustainability index of drying process of onion at different drying air temperatures, drying periods and thicknesses in a cabinet dryer was performed.

Project description:The dataset presented in this article represent the drying characteristics of a semi-automated gas-fired fish dryer. A cabinet dryer was constructed mainly from mild steel sheet metal and stainless steel, and was used for drying prepared fish samples of Clarias gariepinus species. Major operating parameters which included mass of fish, mass of Liquefied Petroleum Gas used, inside temperature of the dryer, and drying time were monitored. Also, output parameters such as the moisture content and energy utilization amongst others were observed and recorded at varying time intervals and specified drying temperatures. The drying temperature was sustained via an incorporated PID temperature controller that allowed drying to proceed until a relatively constant mass of the dried fish samples was attained. The information contained in this data article include a schematic drawing of drying kinetics analysis of the semi-automated gas-fired fish dryer and a pictorial view of the gas-fired cabinet dryer. Also included are pictorial representations of the washed and neatly folded degutted fish samples and dried fish samples. Data provided in this article are those relating to process parameters of the semi-automated fish dryer, data of output parameters of the fish dryer and comparisons of moisture content and energy utilization at different drying temperatures with time.

Project description:The construction of industrial projects involves fabricating and installing massive quantities of pipelines. The design data of pipelines is complex, usually unique for each project and not easily available to researchers and the public for confidentiality reasons. As a result, an industrial pipelines data generator able to realistically simulate pipelines structures will lessen the dependence on real-life data. This article describes an industrial pipelines data generator that was developed using topological and physical properties of pipelines from real industrial projects. This generator can simulate the properties (the type of component, length, diameter, running direction, and the connectivity relationships between components) of real industrial pipelines. Its application in construction engineering and management research, more specifically in the experimental analysis of an optimization algorithm, was described by the authors in previous work [1]. The data generator can be used to develop benchmark problem instances for optimization problems and for simulation studies of construction operations in industrial projects.

Project description:Microbial communities are highly dynamic and sensitive to changes in the environment. Thus, microbiome data are highly susceptible to batch effects, defined as sources of unwanted variation that are not related to and obscure any factors of interest. Existing batch effect correction methods have been primarily developed for gene expression data. As such, they do not consider the inherent characteristics of microbiome data, including zero inflation, overdispersion and correlation between variables. We introduce new multivariate and non-parametric batch effect correction methods based on Partial Least Squares Discriminant Analysis (PLSDA). PLSDA-batch first estimates treatment and batch variation with latent components, then subtracts batch-associated components from the data. The resulting batch-effect-corrected data can then be input in any downstream statistical analysis. Two variants are proposed to handle unbalanced batch x treatment designs and to avoid overfitting when estimating the components via variable selection. We compare our approaches with popular methods managing batch effects, namely, removeBatchEffect, ComBat and Surrogate Variable Analysis, in simulated and three case studies using various visual and numerical assessments. We show that our three methods lead to competitive performance in removing batch variation while preserving treatment variation, especially for unbalanced batch $\times $ treatment designs. Our downstream analyses show selections of biologically relevant taxa. This work demonstrates that batch effect correction methods can improve microbiome research outputs. Project description:Targeted mutagenesis by the CRISPR/Cas9 system is currently revolutionizing genetics. The ease of this technique has enabled genome engineering in-vitro and in a range of model organisms and has pushed experimental dimensions to unprecedented proportions. Due to its tremendous progress in terms of speed, read length, throughput and cost, Next-Generation Sequencing (NGS) has been increasingly used for the analysis of CRISPR/Cas9 genome editing experiments. However, the current tools for genome editing assessment lack flexibility and fall short in the analysis of large amounts of NGS data. Therefore, we designed BATCH-GE, an easy-to-use bioinformatics tool for batch analysis of NGS-generated genome editing data, available from https://github.com/WouterSteyaert/BATCH-GE.git. BATCH-GE detects and reports indel mutations and other precise genome editing events and calculates the corresponding mutagenesis efficiencies for a large number of samples in parallel. Furthermore, this new tool provides flexibility by allowing the user to adapt a number of input variables. The performance of BATCH-GE was evaluated in two genome editing experiments, aiming to generate knock-out and knock-in zebrafish mutants. This tool will not only contribute to the evaluation of CRISPR/Cas9-based experiments, but will be of use in any genome editing experiment and has the ability to analyze data from every organism with a sequenced genome.

Project description:The synthesis of active pharmaceutical ingredients (APIs) is commonly perceived as more efficient when performed using continuous-flow methods, whereas batch processes are often seen as less favorable due to their limitations in yield, heat and mass transfer, and safety. This perception largely stems from existing studies that focus on green metrics such as the E-factor and yield. However, a comprehensive comparison of batch and flow processes through full techno-economic analyses (TEA) and life-cycle assessments (LCA) remains underexplored, leaving key aspects of their environmental and economic impacts inadequately assessed. This work addresses this gap by presenting a detailed comparison of batch and flow syntheses of seven industrially relevant APIs, including amitriptyline hydrochloride, tamoxifen, zolpidem, rufinamide, artesunate, ibuprofen, and phenibut. Eleven environmental impact categories within the framework of nine planetary boundaries were assessed, and the study also included an evaluation of capital and operating costs for both production methods. The results demonstrated that, on average, continuous-flow processes are significantly more sustainable with improvements in energy efficiency, water consumption, and waste reduction. Flow processes also show a marked reduction in carbon emissions and up to a 97% reduction in energy consumption, highlighting their potential for greener API manufacturing. Despite these advantages, the study identified areas where the continuous-flow technology requires further development. Specifically, manufacturing certain APIs in flow show lower-than-average improvements in operating expenditure and land system changes, the latter being directly correlated with the consumption of organic solvents, that can be comparable to or even higher than in batch. These challenges highlight the need for further optimization of flow processes to fully realize their potential in API production.

Project description:Cytometry by Time-Of-Flight (CyTOF) uses antibodies conjugated to isotopically pure metals to identify and quantify a large number of cellular features with single-cell resolution. A barcoding approach allows for 20 unique samples to be pooled and processed together in one tube, reducing the intra-barcode technical variability. However, with only 20 samples per barcode, multiple barcode sets (batches) are required to address questions in robustly powered study designs. A batch adjustment procedure is required to reduce variability across batches and to facilitate direct comparison of runs performed across multiple barcodes run over weeks, months, or years. We describe a method using technical replicates that are included in each run to determine and apply an appropriate adjustment per batch without manual intervention. The use of technical replicate samples (i.e., anchors or reference samples) avoids assumptions of sample homogeneity among batches, and allows direct estimation of batch effects and appropriate adjustment parameters applicable to all samples within a batch. Quantification of cell subpopulations and mean signal intensity pre- and post-adjustment using both manual gating and unsupervised clustering demonstrate substantial mitigation of batch effects in the anchor samples used for this adjustment calculation, and in a second validation set of technical replicates.

Project description:Batch marking is common and useful for many capture-recapture studies where individual marks cannot be applied due to various constraints such as timing, cost, or marking difficulty. When batch marks are used, observed data are not individual capture histories but a set of counts including the numbers of individuals first marked, marked individuals that are recaptured, and individuals captured but released without being marked (applicable to some studies) on each capture occasion. Fitting traditional capture-recapture models to such data requires one to identify all possible sets of capture-recapture histories that may lead to the observed data, which is computationally infeasible even for a small number of capture occasions. In this paper, we propose a latent multinomial model to deal with such data, where the observed vector of counts is a non-invertible linear transformation of a latent vector that follows a multinomial distribution depending on model parameters. The latent multinomial model can be fitted efficiently through a saddlepoint approximation based maximum likelihood approach. The model framework is very flexible and can be applied to data collected with different study designs. Simulation studies indicate that reliable estimation results are obtained for all parameters of the proposed model. We apply the model to analysis of golden mantella data collected using batch marks in Central Madagascar.

Project description:The expression microarray is a frequently used approach to study gene expression on a genome-wide scale. However, the data produced by the thousands of microarray studies published annually are confounded by "batch effects," the systematic error introduced when samples are processed in multiple batches. Although batch effects can be reduced by careful experimental design, they cannot be eliminated unless the whole study is done in a single batch. A number of programs are now available to adjust microarray data for batch effects prior to analysis. We systematically evaluated six of these programs using multiple measures of precision, accuracy and overall performance. ComBat, an Empirical Bayes method, outperformed the other five programs by most metrics. We also showed that it is essential to standardize expression data at the probe level when testing for correlation of expression profiles, due to a sizeable probe effect in microarray data that can inflate the correlation among replicates and unrelated samples.

Project description:BackgroundAir pollution is recognized as an emerging environmental risk factor for neurological diseases. Large-scale epidemiological studies associate traffic-related particulate matter (PM) with impaired cognitive functions and increased incidence of neurodegenerative diseases such as Alzheimer's disease. Inhaled components of PM may directly invade the brain via the olfactory route, or act through peripheral system responses resulting in inflammation and oxidative stress in the brain. Microglia are the immune cells of the brain implicated in the progression of neurodegenerative diseases. However, it remains unknown how PM affects live human microglia.ResultsHere we show that two different PMs derived from exhausts of cars running on EN590 diesel or compressed natural gas (CNG) alter the function of human microglia-like cells in vitro. We exposed human induced pluripotent stem cell (iPSC)-derived microglia-like cells (iMGLs) to traffic related PMs and explored their functional responses. Lower concentrations of PMs ranging between 10 and 100 µg ml-1 increased microglial survival whereas higher concentrations became toxic over time. Both tested pollutants impaired microglial phagocytosis and increased secretion of a few proinflammatory cytokines with distinct patterns, compared to lipopolysaccharide induced responses. iMGLs showed pollutant dependent responses to production of reactive oxygen species (ROS) with CNG inducing and EN590 reducing ROS production.ConclusionsOur study indicates that traffic-related air pollutants alter the function of human microglia and warrant further studies to determine whether these changes contribute to adverse effects in the brain and on cognition over time. This study demonstrates human iPSC-microglia as a valuable tool to study functional microglial responses to environmental agents.
