Biogenic Silver Nanoparticles: Evaluation of Their Biological and Catalytic Potential.

ABSTRACT: The biogenic tailoring of silver nanoparticles using plant extract is becoming an attractive approach in the current scenario. Manilkara zapota (MZ) is well known for its antibacterial, hepato-protective, anti-inflammatory, anti-tussive, anti-fungal, anti-tumour, and free radical scavenging potential. Its plants extract is a rich source of secondary metabolites. Nowadays, silver nanoparticles (AgNPs) have been advocated for a variety of biomedical applications. In present work, silver nanoparticles have been synthesized using an aqueous extract of MZ, physicochemically characterized and finally evaluated for antimicrobial effects, catalytic reduction/degradation of organic dyes and cytotoxicity. The nanosized AgNPs (~ 84 nm) were found to possess prominent antibacterial potential against gram positive and gram negative pathogens (MIC 50 μg/ml) in comparison to native plant extract. Moreover, these particles were found to be non-toxic and efficient eradicators of environmental toxicants via rapid catalytic reduction of toxic chemicals and dyes. Altogether, these results suggest promising potential of these nanoparticles that can be used as multifunctional agents for future biomedical applications.

Project description:Salvia (Lamiaceae family) is used as a brain tonic to improve cognitive function. The species including S. plebeia and S. moorcroftiana are locally used to cure hepatitis, cough, tumours, hemorrhoids, diarrhoea, common cold, flu, and asthma. To the best of authors' knowledge, no previous study has been conducted on synthesis of S. plebeia and S. moorcroftiana silver nanoparticles (SPAgNPs and SMAgNPs). The study was aimed to synthesize AgNPs from the subject species aqueous and ethanol extracts, and assess catalytic potential in degradation of standard and extracted (from yums, candies, and snacks) dyes, nitrophenols, and antibiotics. The study also aimed at AgNPs as probe in sensing metalloids and heavy metal ions including Pb2+, Cu2+, Fe3+, Ni2+, and Zn2+. From the results, it was found that Salvia aqueous extract afforded stable AgNPs in 1:9 and 1:15 (quantity of aqueous extract and silver nitrate solution concentration) whereas ethanol extract yielded AgNPs in 1:10 (quantity of ethanol extract and silver nitrate solution concentration) reacted in sunlight. The size of SPAgNPs and SMAgNPs determined by scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were 21.7 nm and 19.9 nm, with spherical, cylindrical, and deep hollow morphology. The synthesized AgNPs demonstrated significant potential as catalyst in dyes; Congo red (85 %), methylene blue (75 %), Rhodamine B (<50 %), nitrophenols; ortho-nitrophenol (95-98 %) and para-nitrophenol (95-98 %), dyes extracted from food samples including yums, candies, and snacks. The antibiotics (amoxicillin, doxycycline, levofloxacin) degraded up to 80 %-95 % degradation. Furthermore, the synthesized AgNPs as probe in sensing of Pb2+, Cu2+, and Fe3+ in Kabul river water, due to agglomeration, caused a significant decrease and bathochromic shift of SPR band (430 nm) when analyzed after 30 min. The Pb2+ ions was comparatively more agglomerated and chelated. Thus, the practical applicability of AgNPs in Pb2+ sensing was significant. Based on the results of this research study, the synthesized AgNPs could provide promising efficiency in wastewater treatment containing organic dyes, antibiotics, and heavy metals.

Project description:Developing sustainable and eco-friendly methods for nanoparticle (NP) production in an era of environmental consciousness is crucial. This study introduces a novel approach to synthesizing silver (Ag) NPs using Citrus paradisi leaves extract (CPLE) as a green precursor at optimum conditions of the AgNO3 (2 mM) with CPLE in 1:3 ratio, at pH 14 and 80 °C temperature for reaction time of 4 h. The CP@AgNPs were formed and stabilized by Naringen, a major Citrus paradisi component. CP@AgNPs were thoroughly characterized through ultraviolet-visible (UV-vis) and Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD) analysis, and field emission scanning electron microscopy (FE-SEM) imaging techniques. CP@AgNPs demonstrated SPR peak at 450 nm, face cubic crystal structure, the average size of 8 nm, rod-shaped particle adsorbed on quasi-spherical shaped agglomerated NPs, significantly impacting both environmental and biomedical fields. In the catalytic degradation experiment, an application for environment pollutant reducer, CP@AgNPs, achieved an impressive 85% degradation efficiency of the methyl orange (MO) dye, showcasing their potential as a sustainable solution for wastewater treatment. Additionally, CP@AgNPs exhibited potent anti-biofilm properties, with half maximal inhibitory concentration (IC50) values of 0.13 and 0.12 mg/ml against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), respectively, indicating their promise in addressing biofilm-related issues in healthcare and industrial settings. CP@AgNPs also displayed exceptional antioxidant potential with IC50 values of 2.02, 0.07, and 0.035 mg/ml for CPLE, CP@AgNPs, and ascorbic acid, respectively, in scavenging DPPH radical, suggesting their utility in biomedical applications for mitigating oxidative stress. Notably, the bio-activity results of CP@AgNPs surpassed those of CP leaf extract, highlighting the enhanced properties achieved through this green synthesis approach. This study provides a sustainable and environmental remediation to biomedical science.

Project description:BackgroundBiological synthesis of nanomaterials possesses unprecedented potential in the production of nanomaterials due to their ability to produce nanomaterials with improved biocompatibility in addition to eco-friendly synthetic procedures.MethodsThis article reports the isolation of an air-borne fungus from the campus of Integral University, Lucknow, with an exceptional ability to withstand very high concentrations of silver salt. The fungus was found to produce pentagonal silver nanoparticles (AgPgNps) when silver ions were reduced from silver nitrate. Molecular analysis and biochemical characterization techniques based on 18-seconds rRNA identified the fungus to belong to the Aspergillus sp. with the NCBI accession no KF913249. Material characterization techniques including ultraviolet (UV)-visible spectroscopy, transmission electron microscopy, and zeta potential analysis were used to satisfactorily characterize the as-synthesized AgPgNps.ResultsThe AgPgNps synthesized by the fungus Aspergillus sp. exhibit an absorption that is maximum centered at about 416 nm, with a standard particle size of 23.22±2 nm. These AgPgNps exhibited broad-spectrum antimicrobial activities against an array of bacterial pathogens with remarkable minimum inhibitory concentration (MIC50) values: Staphylococcus aureus (ATCC 25923) - 9.230 µg/mL, Bacillus sp. (ATCC 14593) - 12.781 µg/mL, Escherichia coli (ATCC 25922) - 5.063 µg/mL, and Klebsiella pneumoniae (ATCC 13883) - 5.426 µg/mL. In vitro cytotoxicity analysis of biosynthesized AgPgNps showed a dose-response activity against human cervical cancer cell line (HeLa) and adenocarcinoma cells (A549) with MIC50 values of 0.038 µg/mL and 0.044 µg/mL, respectively.ConclusionThese findings are very crucial to evaluate the biosynthetic process for the synthesis of nanoparticles (NPs) with unique properties. These NPs may find potential applications in sensing, medicine, and antimicrobial and anticancer therapies.
