Delta-Frequency Augmentation and Synchronization in Seizure Discharges and Telencephalic Transmission.

ABSTRACT: Epileptic seizures constitute a common neurological disease primarily diagnosed by characteristic rhythms or waves in the local field potentials (LFPs) of cerebral cortices or electroencephalograms. With a basolateral amygdala (BLA) kindling model, we found that the dominant frequency of BLA oscillations is in the delta range (1-5 Hz) in both normal and seizure conditions. Multi-unit discharges are increased with higher seizure staging but remain phase-locked to the delta waves in LFPs. Also, the change in synchrony precedes and outlasts the changes in discharging units as well as behavioral seizures. One short train of stimuli readily drives the pyramidal-inhibitory neuronal networks in BLA slices into prolonged reverberating activities, where the burst and interburst intervals may concurrently set a "natural wavelength" for delta frequencies. Seizures thus could be viewed as erroneous temporospatial continuums to normal oscillations in a system with a built-in synchronizing and resonating nature for information relay.

Project description:ObjectiveMarkers of seizure recurrence are needed to personalize antiseizure medication (ASM) therapy. In the clinical practice, EEG features are considered to be related to the risk of seizure recurrence for genetic generalized epilepsies (GGE). However, to our knowledge, there are no studies analyzing systematically specific EEG features as indices of ASM efficacy in GGE. In this study, we aimed at identifying EEG indicators of ASM responsiveness in Juvenile Myoclonic Epilepsy (JME), which, among GGE, is characterized by specific electroclinical features.MethodsWe compared the features of prolonged ambulatory EEG (paEEG, 22 h of recording) of JME patients experiencing seizure recurrence within a year ("cases") after EEG recording, with those of patients with sustained seizure freedom for at least 1 year after EEG ("controls"). We included only EEG recordings of patients who had maintained the same ASM regimen (dosage and type) throughout the whole time period from the EEG recording up to the outcome events (which was seizure recurrence for the "cases", or 1-year seizure freedom for "controls"). As predictors, we evaluated the total number, frequency, mean and maximum duration of epileptiform discharges (EDs) and spike density (i.e. total EDs duration/artifact-free EEG duration) recorded during the paEEG. The same indexes were assessed also in standard EEG (stEEG), including activation methods.ResultsBoth the maximum length and the mean duration of EDs recorded during paEEG significantly differed between cases and controls; when combined in a binary logistic regression model, the maximum length of EDs emerged as the only valid predictor. A cut-off of EDs duration of 2.68 seconds discriminated between cases and controls with a 100% specificity and a 93% sensitivity. The same indexes collected during stEEG lacked both specificity and sensitivity.SignificanceThe occurrence of prolonged EDs in EEG recording might represent an indicator of antiepileptic drug failure in JME patients.

Project description:The self-synchronization of rotors mounted on different vibrating bodies can be easily controlled by adjusting the coupling parameters. To reveal the synchronization characteristics of a weakly damped system with two rotors mounted on different vibrating bodies, we propose a simplified physical model. The topics described in this paper are related to coupling dynamic problems between two vibrating systems. Both synchronization and vibratory synchronization transmission of the system are studied. The coupling mechanism between the two rotors is analyzed to derive the synchronization condition and the stability criterion of the system. The vibration of the system is described by an averaging method that can separate fast motion (high frequency) from slow motion (low frequency). Theoretical research shows that vibration torque is the key factor in balancing the energy distribution between the rotors. Taking the maximum vibration torque (MVT) as a critical parameter, we investigate the synchronization characteristics of the vibrating system in different cases. The curve of the maximum vibration torque (MVT) versus coupling frequency is divided into several parts by the coupling characteristic frequency and the input torque difference between the rotors. Simulations of the system with coupling frequencies from different parts are carried out. For the system with rotational frequencies larger than the natural frequencies, the coupling characteristic frequency or characteristic frequency curve should be considered. When the coupling frequency is close to the characteristic frequency or the vibration state is close to the characteristic frequency curve, self-synchronization of the two rotors can be obtained easily. Under certain conditions when the coupling effect between the rotors is strong enough, the rotors can maintain synchronous rotation even when one of the two motors is shut off after synchronization is achieved, which is called vibratory synchronization transmission. Vibratory synchronization transmission of the system occurs in a new synchronous condition, and the phase difference between the rotors takes on a new value, that is, the system approaches a new synchronization state.
