Project description:Electronic-cigarette, or vaping, product use-associated lung injury (EVALI) is a syndrome of acute respiratory failure characterized by monocytic and neutrophilic alveolar inflammation. Epidemiological and clinical evidence suggests a role of vitamin E acetate (VEA) in the development of EVALI, yet it remains unclear whether VEA has direct pulmonary toxicity. To test the hypotheses that aerosolized VEA causes lung injury in mice and directly injures human alveolar epithelial cells, we exposed adult mice and primary human alveolar epithelial type II (AT II) cells to an aerosol of VEA generated by a device designed for vaping oils. Outcome measures in mice included lung edema, BAL analysis, histology, and inflammatory cytokines; in vitro outcomes included cell death, cytokine release, cellular uptake of VEA, and gene-expression analysis. Comparison exposures in both models included the popular nicotine-containing JUUL aerosol. We discovered that VEA caused dose-dependent increases in lung water and BAL protein compared with control and JUUL-exposed mice in association with increased BAL neutrophils, oil-laden macrophages, multinucleated giant cells, and inflammatory cytokines. VEA aerosol was also toxic to AT II cells, causing increased cell death and the release of monocyte and neutrophil chemokines. VEA was directly absorbed by AT II cells, resulting in the differential gene expression of several inflammatory biological pathways. Given the epidemiological and clinical characteristics of the EVALI outbreak, these results suggest that VEA plays an important causal role.

Project description:Cloud condensation nuclei (CCN) number concentrations alongside with submicrometer particle number size distributions and particle chemical composition have been measured at atmospheric observatories of the Aerosols, Clouds, and Trace gases Research InfraStructure (ACTRIS) as well as other international sites over multiple years. Here, harmonized data records from 11 observatories are summarized, spanning 98,677 instrument hours for CCN data, 157,880 for particle number size distributions, and 70,817 for chemical composition data. The observatories represent nine different environments, e.g., Arctic, Atlantic, Pacific and Mediterranean maritime, boreal forest, or high alpine atmospheric conditions. This is a unique collection of aerosol particle properties most relevant for studying aerosol-cloud interactions which constitute the largest uncertainty in anthropogenic radiative forcing of the climate. The dataset is appropriate for comprehensive aerosol characterization (e.g., closure studies of CCN), model-measurement intercomparison and satellite retrieval method evaluation, among others. Data have been acquired and processed following international recommendations for quality assurance and have undergone multiple stages of quality assessment.

Project description:Particulate phosphorus (PP) is often the largest component of the total phosphorus (P) load in stormwater. Fine-resolution measurement of particle sizes allows us to investigate the mechanisms behind the removal of PP in stormwater wetlands, since the diameter of particles influences the settling velocity and the amount of sorbed P on a particle. In this paper, we present a novel method to estimate PP, where we measure and count individual particles in stormwater and use the total surface area as a proxy for PP. Our results show a strong relationship between total particle surface area and PP, which we use to put forth a simple mechanistic model of PP removal via gravitational settling of individual mineral particles, based on a continuous particle size distribution. This information can help improve the design of stormwater Best management practices to reduce PP loading in both urban and agricultural watersheds.

Project description:The growing use of silver nanoparticles (AgNPs) in consumer products raises concerns about potential health effects. This study investigated the persistence and clearance of 2 different size AgNPs (20 and 110 nm) delivered to rats by single nose-only aerosol exposures (6 h) of 7.2 and 5.4 mg/m(3), respectively. Rat lung tissue was assessed for silver accumulations using inductively-coupled plasma mass spectrometry (ICP-MS), autometallography, and enhanced dark field microscopy. Involvement of tissue macrophages was assessed by scoring of silver staining in bronchoalveolar lavage fluid (BALF). Silver was abundant in most macrophages at 1 day post-exposure. The group exposed to 20 nm AgNP had the greatest number of silver positive BALF macrophages at 56 days post-exposure. While there was a significant decrease in the amount of silver in lung tissue at 56 days post-exposure compared with 1 day following exposure, at least 33% of the initial delivered dose was still present for both AgNPs. Regardless of particle size, silver was predominantly localized within the terminal bronchial/alveolar duct junction region of the lung associated with extracellular matrix and within epithelial cells. Inhalation of both 20 and 110 nm AgNPs resulted in a persistence of silver in the lung at 56 days post-exposure and local deposition as well as accumulation of silver at the terminal bronchiole alveolar duct junction. Further the smaller particles, 20 nm AgNP, produced a greater silver burden in BALF macrophages as well as greater persistence of silver positive macrophages at later timepoints (21 and 56 days).

Project description:Here, we present a novel protocol concept for quantifying the cooling performance of particle-based radiative cooling (PBRC). PBRC, known for its high flexibility and scalability, emerges as a promising method for practical applications. The cooling power, one of the cooling performance indexes, is the typical quantitative performance index, representing its cooling capability at the surface. One of the primary obstacles to predicting cooling power is the difficulty of simulating the non-uniform size and shape of micro-nanoparticles in the PBRC film. The present work aims to develop an accurate protocol for predicting the cooling power of PBRC film using image processing and regression analysis techniques. Specifically, the protocol considers the particle size distribution through circle object detection on SEM images and determines the probability density function based on a chi-square test. To validate the proposed protocol, a PBRC structure with PDMS/Al2O3 micro-nanoparticles is fabricated, and the proposed protocol precisely predicts the measured cooling power with a 7.8% error. Through this validation, the proposed protocol proves its potential and reliability for the design of PBRC.

Project description:Comminution is important in the processing of biological materials, such as cereal grains, wood biomass, and food waste. The most popular biomaterial grinders are hammer and roller mills. However, the grinders with great potential in the processing of biomass are mills that use cutting, e.g., disc mills. When it comes to single-disc and multi-disc grinders, there are not many studies describing the relationships between energy, motion, material, and processing or describing the effect of grinding, meaning the size distribution of a product. The relationship between the energy and size reduction ratio of disc-type grinder designs has also not been sufficiently explored. The purpose of this paper was to develop models for the particle size distribution of the ground product in multi-disc mills depending on the variable process parameters, i.e., disc rotational velocity and, consequently, power consumption, and the relationship between the grinding energy and the shape of graining curves, which would help predict the product size reduction ratio for these machines. The experiment was performed using a five-disc mill, assuming the angular velocity of the grinder discs was variable. Power consumption, product particle size, and specific comminution energy were recorded during the tests. The Rosin-Rammler-Sperling-Bennet (RRSB) distribution curves were established for the ground samples, and the relationships between distribution coefficients and the average angular velocity of grinder discs, power consumption, and specific comminution energy were determined. The tests showed that the specific comminution energy increases as the size reduction ratio increases. It was also demonstrated that the RRSB distribution coefficients could be represented by the functions of angular velocities, power consumption, and specific comminution energy. The developed models will be a source of information for numerical modelling of comminution processes.

Project description:RationaleThere are unexplained geographical and seasonal differences in the short-term effects of fine particulate matter (PM(2.5)) on human health. The hypothesis has been advanced to include the possibility that such differences might be due to variations in the PM(2.5) chemical composition, but evidence supporting this hypothesis is lacking.ObjectivesTo examine whether variation in the relative risks (RR) of hospitalization associated with ambient exposure to PM(2.5) total mass reflects differences in PM(2.5) chemical composition.MethodsWe linked two national datasets by county and by season: (1) long-term average concentrations of PM(2.5) chemical components for 2000-2005 and (2) RRs of cardiovascular and respiratory hospitalizations for persons 65 years or older associated with a 10-microg/m(3) increase in PM(2.5) total mass on the same day for 106 U.S. counties for 1999 through 2005.Measurements and main resultsWe found a positive and statistically significant association between county-specific estimates of the short-term effects of PM(2.5) on cardiovascular and respiratory hospitalizations and county-specific levels of vanadium, elemental carbon, or nickel PM(2.5) content.ConclusionsCommunities with higher PM(2.5) content of nickel, vanadium, and elemental carbon and/or their related sources were found to have higher risk of hospitalizations associated with short-term exposure to PM(2.5).

Project description:Nearly two years after vitamin E acetate (VEA) was identified as the potential cause of the 2019-2020 outbreak of e-cigarette, or vaping product-associated lung injuries (EVALI), the toxicity mechanisms of VEA vaping are still yet to be fully understood. Studies since the outbreak have found that e-liquids such as VEA undergo thermal degradation during the vaping process to produce various degradation products, which may pose a greater risk of toxicity than exposure to unvaped VEA. Additionally, a wide range of customizable parameters-including the model of e-cigarette used, puffing topography, or the applied power/temperature used to generate aerosols-have been found to influence the physical properties and chemical compositions of vaping emissions. However, the impact of heating coil temperature on the chemical composition of VEA vaping emissions has not been fully assessed. In this study, we investigated the emission product distribution of VEA vaping emissions produced at temperatures ranging from 176 to 356°C, corresponding to a variable voltage vape pen set at 3.3 to 4.8V. VEA degradation was found to be greatly enhanced with increasing temperature, resulting in a shift towards the production of lower molecular weight compounds, such as the redox active duroquinone (DQ) and short-chain alkenes. Low temperature vaping of VEA resulted in the production of long-chain molecules, such as phytol, exposure to which has been suggested to induce lung damage in previous studies. Furthermore, differential product distribution was observed in VEA degradation products generated from vaping and from pyrolysis using a tube furnace in the absence of the heating coil at equivalent temperatures, suggesting the presence of external factors such as metals or oxidation that may enhance VEA degradation during vaping. Overall, our findings indicate that vaping behavior may significantly impact the risk of exposure to toxic vaping products and potential for vaping-related health concerns.

Project description:True thermodynamic stability of a solid colloidal dispersion is generally unexpected, so much that thorough experimental validation of proposed stable systems remains incomplete. Such dispersions are under investigated and would be of interest due to their long-term stability and insensitivity to preparation pathway. We apply classical nucleation theory (CNT) to such colloidal systems, providing a relationship which links the size-dependent interfacial free energy density of the particles to their size distribution, and use this expression in the fitting of previously reported size distributions for putatively thermodynamically stable nanoparticles. Experimental data from a gold-thiol system exhibiting inverse coarsening or "digestive ripening" can be well-described in terms of a power-law dependence of the interfacial free energy γ on radius based on capacitive charging of the nanoparticles, going as r-3 , as suggested by prior authors. Data from magnetite nanoparticles in highly basic solutions also can be well-fit using the CNT relation, but with γ going as r-2 . Slightly better fits are possible if the power of the radius is non-integral, but we stress that more complex models of γ will require richer data sets to avoid the problem of overfitting. Some parameters of the fits are still robustly at odds with earlier models that implicitly assumed absolute thermodynamic stability: first, the extrapolated free energy density of the flat surface in these systems is small and positive, rather than strongly negative; second, the shape of the distributions indicates the solution phase to be supersaturated in monomer relative to the bulk, and thus that these two systems may only be metastable. For future work, we derive expressions for the important statistical thermodynamic and chemical parameters of the interface energy in terms of 1) the surfactant concentration, 2) the temperature dependence, and 3) the concentrations of particles in the tail of the distribution.
