In vivo and in vitro inflammatory responses to fine particulate matter (PM_2.5) from China and California.

ABSTRACT: Ambient PM_2.5 was collected during the winter season from Taiyuan, Shanxi, China; Jinan, Shandong, China; and Sacramento, California, USA, and used to create PM_SX, PM_SD, and PM_CA extracts, respectively. Time-lag experiments were performed to explore the in vivo and in vitro toxicity of the PM extracts. In vivo inflammatory lung responses were assessed in BALB/c mice using a single oropharyngeal aspiration (OPA) of PM extract or vehicle (CTRL) on Day 0. Necropsies were performed on Days 1, 2, and 4 post-OPA, and pulmonary effects were determined using bronchoalveolar lavage (BAL) and histopathology. On Day 1, BAL neutrophils were significantly elevated in all PM- versus CTRL-exposed mice, with PM_CA producing the strongest response. However, histopathological scoring showed greater alveolar and perivascular effects in PM_SX-exposed mice compared to all three other groups. By Day 4, BAL neutrophilia and tissue inflammation were resolved, similar across all groups. In vitro effects were examined in human HepG2 hepatocytes, and U937 cells following 6, 24, or 48 h of exposure to PM extract or DMSO (control). Luciferase reporter and quantitative polymerase chain reaction assays were used to determine in vitro effects on aryl hydrocarbon receptor (AhR) activation and gene transcription, respectively. Though all three PM extracts activated AhR, PM_SX produced the greatest increases in AhR activation, and mRNA levels of cyclooxygenase-2, cytochrome P450, interleukin (IL)-8, and interleukin (IL)-1β. These effects were assumed to result from a greater abundance of polycyclic aromatic hydrocarbons (PAHs) in PM_SX compared to PM_SD and PM_CA.

Project description:Condensable particulate matter (CPM) emitted from stationary combustion and mobile sources exhibits high emissions and a large proportion of organic components. However, CPM is not generally measured when conducting emission surveys of PM in most countries, including China. Consequently, previous emission inventories have not included emission rates for CPM. Here, we construct an emission inventory of CPM in China with a focus on organic aerosols (OAs) based on collected CPM emission information. Results show that OA emissions are enhanced twofold after the inclusion of CPM in a new inventory for China for the years 2014 and 2017. Considering organic CPM emissions and model representations of secondary OA (SOA) formation from CPM, a series of sensitivity cases have been simulated here using the three-dimensional Community Multiscale Air Quality (CMAQ) model to estimate the contributions of CPM emissions to atmospheric OA and fine PM (PM2.5, particulate matter with aerodynamic diameter not exceeding 2.5 μm) concentrations in China. Compared with observations at a Beijing site during a haze episode from 14 October to 14 November 2014, estimates of the temporal average primary OA (POA) and SOA concentrations were greatly improved after including the CPM effects. These scenarios demonstrated the significant contributions of CPM emissions from stationary combustion and mobile sources to the POA (51 %-85 %), SOA (42 %-58 %), and total OA concentrations (45 %-75 %). Furthermore, the contributions of CPM emissions to total OA concentrations were demonstrated over the 2 major cities and 26 other cities of the Beijing-Tianjin-Hebei region (hereafter referred to as the "BTH2 + 26 cities") in December 2018, with average contributions of up to 49 %, 53 %, 54 %, and 50 % for Handan, Shijiazhuang, Xingtai, and Dezhou, respectively. Correspondingly, the inclusion of CPM emissions also narrowed the gap between simulated and observed PM2.5 concentrations over the BTH2 + 26 cities. These results improve the simulation performance of atmospheric OA and PM2.5 and may also provide important implications for the sources of OA.

Project description:The main objective of this study was chemical characterization and source apportionment of the oxidative potential of ambient PM2.5 samples collected in an urban background area in Athens, Greece. Ambient PM2.5 samples were collected during the summer (June-September) of 2017 and winter (February-March) of 2018 at a residential, urban background site in the outlying neighborhood of the Demokritos National Laboratory in Athens, Greece. The collected PM samples were analyzed for their chemical constituents including metals and trace elements, water-soluble organic carbon (WSOC), elemental and organic carbon (EC/OC), and marker of biomass burning (i.e., levoglucosan). In addition, the DCFH in vitro assay was performed to determine the oxidative potential of the PM2.5 samples. We performed a series of statistical analyses, including Spearman rank-order correlation analysis, principal component analysis (PCA), and multi linear regression (MLR) to determine the most significant species (as source tracers) contributing to the oxidative potential of PM2.5. Our findings revealed that the intrinsic (per PM mass) and extrinsic (per m3 of air volume) oxidative potentials of the collected ambient PM2.5 samples were significantly higher than those measured in many urban areas around the world. The results of the MLR analyses indicated that the major pollution sources contributing to the oxidative potential of ambient PM2.5 were vehicular emissions (characterized by EC) (44%), followed by secondary organic aerosol (SOA) formation (characterized by WSOC) (16%), and biomass burning (characterized by levoglucosan) (9%). The oxidative potential of the collected ambient PM2.5 samples was also higher in summer compared to the winter, mainly due to higher concentrations of EC and WSOC during this season. Results from this study corroborate the impact of traffic and SOA on the oxidative potential of ambient PM2.5 in greater Athens area, and can be helpful in adopting appropriate public health policies regarding detrimental outcomes of exposure to PM2.5.
