Mapping O₂ concentration in ex-vivo tissue samples on a fast PLIM macro-imager.

ABSTRACT: O₂ PLIM microscopy was employed in various studies, however current platforms have limitations in sensitivity, image acquisition speed, accuracy and general usability. We describe a new PLIM imager based on the Timepix3 camera (Tpx3cam) and its application for imaging of O₂ concentration in various tissue samples stained with a nanoparticle based probe, NanO2-IR. Upon passive staining of mouse brain, lung or intestinal tissue surface with minute quantities of NanO2-IR or by microinjecting the probe into the lumen of small or large intestine fragments, robust phosphorescence intensity and lifetime signals were produced, which allow mapping of O₂ in the tissue within 20 s. Inhibition of tissue respiration or limitation of O₂ diffusion to tissue produced the anticipated increases or decreases in O₂ levels, respectively. The difference in O₂ concentration between the colonic lumen and air-exposed serosal surface was around 140 µM. Furthermore, subcutaneous injection of 5 µg of the probe in intact organs (a paw or tail of sacrificed mice) enabled efficient O₂ imaging at tissue depths of up to 0.5 mm. Overall, the PLIM imager holds promise for metabolic imaging studies with various ex vivo models of animal tissue, and also for use in live animals.

Project description:Electrical properties (EP), namely conductivity and permittivity, can provide endogenous contrast for tissue characterization. Using electrical property tomography (EPT), maps of EP can be generated from conventional MRI data. This report investigates the feasibility and accuracy of EPT at 21.1 T for multiple RF coils and modes of operation using phantoms. Additionally, it demonstrates the EP of the in vivo rat brain with and without ischemia. Helmholtz-based EPT was implemented in its Full-form, which demands the complex [Formula: see text] field, and a simplified form requiring either just the [Formula: see text] field phase for conductivity or the [Formula: see text] field magnitude for permittivity. Experiments were conducted at 21.1 T using birdcage and saddle coils operated in linear or quadrature transceive mode, respectively. EPT approaches were evaluated using a phantom, ex and in vivo Sprague-Dawley rats under naïve conditions and ischemic stroke via transient middle cerebral artery occlusion. Different conductivity reconstruction approaches applied to the phantom displayed average errors of 12%-73% to the target acquired from dielectric probe measurements. Permittivity reconstructions showed higher agreement and an average 3%-8% error to the target depending on reconstruction approach. Conductivity and permittivity of ex and in vivo rodent brain were measured. Elevated EP in the ischemia region correlated with the increased sodium content and the influx of water intracellularly following ischemia in the lesion were detected. The Full-form technique generated from the linear birdcage provided the best accuracy for EP of the phantom. Phase-based conductivity and magnitude-based permittivity mapping provided reasonable estimates but also demonstrated the limitations of Helmholtz-based EPT at 21.1 T. Permittivity reconstruction was improved significantly over lower fields, suggesting a novel metric for in vivo brain studies. EPT applied to ischemic rat brain proved sensitivity to physiological changes, motivating the future application of more advanced reconstruction approaches.

Project description:BackgroundIn vitro models of prostate cancer (PCa) are not always reliable to evaluate anticancer treatment efficacy. This limitation may be overcome by using viable tumor slice material. Here we report on the establishment of an optimized ex vivo method to culture tissue slices from patient-derived xenografts (PDX) of prostate cancer (PCa), to assess responses to PCa treatments.MethodsThree PDX models were used that are characterized by different androgen receptor (AR) expression and different homology directed DNA repair capacities, due to a breast cancer associated two (BRCA2) wild-type or mutated status. Tumors were removed from mice, sliced using a vibratome and cultured for a maximum of 6 days. To test the sensitivity to androgen antagonist, tumor slices from the AR-expressing and AR-negative PDX tumors were treated with the anti-androgen enzalutamide. For sensitivity to DNA repair intervention, tumors slices from BRCA2 wild-type and mutated PDXs were treated with the poly (ADP-ribose) polymerase-1 inhibitor olaparib. Treatment response in these tumor slices was determined by measuring slice morphology, cell proliferation, apoptosis, AR expression level, and secretion of prostate specific antigen (PSA).ResultsWe compared various culture conditions (support materials, growth media, and use of a 3D smooth rocking platform) to define the optimal condition to maintain tissue viability and proliferative capacity up to least 6 days. Under optimized conditions, enzalutamide treatment significantly decreased proliferation, increased apoptosis, and reduced AR-expression and PSA secretion of AR-expressing tumor slices compared to AR-negative slices, that did not respond to the intervention. Olaparib treatment significantly increased cell death in BRCA2 mutated tumors slices as compared to slices from BRCA2 wild type tumors.ConclusionsEx vivo treatment of PCa PDX tumor slices with enzalutamide and olaparib recapitulates responses previously observed in vivo. The faithful retention of tissue structure and function in this ex vivo model offers an ideal opportunity for treatment efficacy screening, thereby reducing costs and numbers of experimental animals.
