Project description:In an ecosystem, environmental changes as a result of natural and human processes can cause some key parameters of the system to change with time. Depending on how fast such a parameter changes, a tipping point can occur. Existing works on rate-induced tipping, or R-tipping, offered a theoretical way to study this phenomenon but from a local dynamical point of view, revealing, e.g., the existence of a critical rate for some specific initial condition above which a tipping point will occur. As ecosystems are subject to constant disturbances and can drift away from their equilibrium point, it is necessary to study R-tipping from a global perspective in terms of the initial conditions in the entire relevant phase space region. In particular, we introduce the notion of the probability of R-tipping defined for initial conditions taken from the whole relevant phase space. Using a number of real-world, complex mutualistic networks as a paradigm, we find a scaling law between this probability and the rate of parameter change and provide a geometric theory to explain the law. The real-world implication is that even a slow parameter change can lead to a system collapse with catastrophic consequences. In fact, to mitigate the environmental changes by merely slowing down the parameter drift may not always be effective: Only when the rate of parameter change is reduced to practically zero would the tipping be avoided. Our global dynamics approach offers a more complete and physically meaningful way to understand the important phenomenon of R-tipping.

Project description:Complex and nonlinear ecological networks can exhibit a tipping point at which a transition to a global extinction state occurs. Using real-world mutualistic networks of pollinators and plants as prototypical systems and taking into account biological constraints, we develop an ecologically feasible strategy to manage/control the tipping point by maintaining the abundance of a particular pollinator species at a constant level, which essentially removes the hysteresis associated with a tipping point. If conditions are changing so as to approach a tipping point, the management strategy we describe can prevent sudden drastic changes. Additionally, if the system has already moved past a tipping point, we show that a full recovery can occur for reasonable parameter changes only if there is active management of abundance, again due essentially to removal of the hysteresis. This recovery point in the aftermath of a tipping point can be predicted by a universal, two-dimensional reduced model.

Project description:Transients are fundamental to ecological systems with significant implications to management, conservation and biological control. We uncover a type of transient synchronization behavior in spatial ecological networks whose local dynamics are of the chaotic, predator-prey type. In the parameter regime where there is phase synchronization among all the patches, complete synchronization (i.e. synchronization in both phase and amplitude) can arise in certain pairs of patches as determined by the network symmetry-henceforth the phenomenon of 'synchronization within synchronization.' Distinct patterns of complete synchronization coexist but, due to intrinsic instability or noise, each pattern is a transient and there is random, intermittent switching among the patterns in the course of time evolution. The probability distribution of the transient time is found to follow an algebraic scaling law with a divergent average transient lifetime. Based on symmetry considerations, we develop a stability analysis to understand these phenomena. The general principle of symmetry can also be exploited to explain previously discovered, counterintuitive synchronization behaviors in ecological networks.

Project description:Brain-state transitions are readily apparent from changes in brain rhythms,1 but are difficult to predict, suggestive that the underlying cause is latent to passive recording methods. Among the most important transitions, clinically, are the starts of seizures. We here show that an 'active probing' approach may have several important benefits for epileptic management, including by helping predict these transitions. We used mice expressing the optogenetic actuator, channelrhodopsin, in pyramidal cells, allowing this population to be stimulated in isolation. Intermittent stimulation at frequencies as low as 0.033 Hz (period = 30 s) delayed the onset of seizure-like events in an acute brain slice model of ictogenesis, but the effect was lost if stimulation was delivered at even lower frequencies (1/min). Notably, active probing additionally provides advance indication of when seizure-like activity is imminent, revealed by monitoring the postsynaptic response to stimulation. The postsynaptic response, recorded extracellularly, showed an all-or-nothing change in both amplitude and duration, a few hundred seconds before seizure-like activity began-a sufficient length of time to provide a helpful warning of an impending seizure. The change in the postsynaptic response then persisted for the remainder of the recording, indicative of a state change from a pre-epileptic to a pro-epileptic network. This occurred in parallel with a large increase in the stimulation-triggered Ca2+ entry into pyramidal dendrites, and a step increase in the number of evoked postsynaptic action potentials, both consistent with a reduction in the threshold for dendritic action potentials. In 0 Mg2+ bathing media, the reduced threshold was not associated with changes in glutamatergic synaptic function, nor of GABAergic release from either parvalbumin or somatostatin interneurons, but simulations indicate that the step change in the optogenetic response can instead arise from incremental increases in intracellular [Cl-]. The change in the response to stimulation was replicated by artificially raising intracellular [Cl-], using the optogenetic chloride pump, halorhodopsin. By contrast, increases in extracellular [K+] cannot account for the firing patterns in the response to stimulation, although this, and other cellular changes, may contribute to ictal initiation in other circumstances. We describe how these various cellular changes form a synergistic network of positive feedback mechanisms, which may explain the precipitous nature of seizure onset. This model of seizure initiation draws together several major lines of epilepsy research as well as providing an important proof-of-principle regarding the utility of open-loop brain stimulation for clinical management of the condition.

Project description:Determining mechanism-based biomarkers that distinguish adaptive and adverse cellular processes is critical to understanding the health effects of environmental exposures. Shifting from in vivo, low-throughput toxicity studies to high-throughput screening (HTS) paradigms and risk assessment based on in vitro and in silico testing requires utilizing toxicity pathway information to distinguish adverse outcomes from recoverable adaptive events. Little work has focused on oxidative stresses in human airway for the purposes of predicting adverse responses. We hypothesize that early gene expression-mediated molecular changes could be used to delineate adaptive and adverse responses to environmentally-based perturbations. Here, we examined cellular responses of the tracheobronchial airway to zinc (Zn) exposure, a model oxidant. Airway derived BEAS-2B cells exposed to 2–10 µM Zn2+ elicited concentration- and time-dependent cytotoxicity. Normal, adaptive, and cytotoxic Zn2+ exposure conditions were determined with traditional apical endpoints, and differences in global gene expression around the tipping point of the responses were used to delineate underlying molecular mechanisms. Bioinformatic analyses of differentially expressed genes indicate early enrichment of stress signaling pathways, including those mediated by the transcription factors p53 and NRF2. After 4 h, 154 genes were differentially expressed (p <0.01) between the adaptive and cytotoxic Zn2+ concentrations. Nearly 40% of the biomarker genes were related to the p53 signaling pathway with 30 genes identified as likely direct targets using a database of p53 ChIP-seq studies. Despite similar p53 activation profiles, these data revealed widespread dampening of p53 and NRF2-related genes as early as 4 h after exposure at higher, unrecoverable Zn2+ exposures. Thus, in our model early increased activation of stress response pathways indicated a recoverable adaptive event. Overall, this study highlights the importance of characterizing molecular mechanisms around the tipping point of adverse responses to better inform HTS paradigms.

Project description:The Amazon rainforest has been proposed as a tipping element of the earth system, with the possibility of a dieback of the entire ecosystem due to deforestation only of parts of the rainforest. Possible physical mechanisms behind such a transition are still subject to ongoing debates. Here, we use a specifically designed model to analyse the nonlinear couplings between the Amazon rainforest and the atmospheric moisture transport from the Atlantic to the South American continent. These couplings are associated with a westward cascade of precipitation and evapotranspiration across the Amazon. We investigate impacts of deforestation on the South American monsoonal circulation with particular focus on a previously neglected positive feedback related to condensational latent heating over the rainforest, which strongly enhances atmospheric moisture inflow from the Atlantic. Our results indicate the existence of a tipping point. In our model setup, crossing the tipping point causes precipitation reductions of up to 40% in non-deforested parts of the western Amazon and regions further downstream. The responsible mechanism is the breakdown of the aforementioned feedback, which occurs when deforestation reduces transpiration to a point where the available atmospheric moisture does not suffice anymore to release the latent heat needed to maintain the feedback.

Project description:Understanding tipping point dynamics in harvested ecosystems is of crucial importance for sustainable resource management because ignoring their existence imperils social-ecological systems that depend on them. Fisheries collapses provide the best known examples for realizing tipping points with catastrophic ecological, economic and social consequences. However, present-day fisheries management systems still largely ignore the potential of their resources to exhibit such abrupt changes towards irreversible low productive states. Using a combination of statistical changepoint analysis and stochastic cusp modelling, here we show that Western Baltic cod is beyond such a tipping point caused by unsustainable exploitation levels that failed to account for changing environmental conditions. Furthermore, climate change stabilizes a novel and likely irreversible low productivity state of this fish stock that is not adapted to a fast warming environment. We hence argue that ignorance of non-linear resource dynamics has caused the demise of an economically and culturally important social-ecological system which calls for better adaptation of fisheries systems to climate change.

Project description:Detection in high fidelity of tipping points, the emergence of which is often induced by invisible changes in internal structures or/and external interferences, is paramountly beneficial to understanding and predicting complex dynamical systems (CDSs). Detection approaches, which have been fruitfully developed from several perspectives (e.g., statistics, dynamics, and machine learning), have their own advantages but still encounter difficulties in the face of high-dimensional, fluctuating datasets. Here, using the reservoir computing (RC), a recently notable, resource-conserving machine learning method for reconstructing and predicting CDSs, we articulate a model-free framework to accomplish the detection only using the time series observationally recorded from the underlying unknown CDSs. Specifically, we encode the information of the CDS in consecutive time durations of finite length into the weights of the readout layer in an RC, and then we use the learned weights as the dynamical features and establish a mapping from these features to the system's changes. Our designed framework can not only efficiently detect the changing positions of the system but also accurately predict the intensity change as the intensity information is available in the training data. We demonstrate the efficacy of our supervised framework using the dataset produced by representative physical, biological, and real-world systems, showing that our framework outperforms those traditional methods on the short-term data produced by the time-varying or/and noise-perturbed systems. We believe that our framework, on one hand, complements the major functions of the notable RC intelligent machine and, on the other hand, becomes one of the indispensable methods for deciphering complex systems.

Project description:BackgroundAlthough risk factors for poor outcomes and complications have been studied, there remain limited objective criteria to guide surgeons about the timing of arthroplasty. The purpose of this study was to further characterize the tipping-point scores for a group of patient-reported outcome measures (PROMs) in patients undergoing primary shoulder arthroplasty.MethodsWe retrospectively reviewed 5670 primary shoulder arthroplasties (1833 anatomic total shoulder arthroplasties and 3837 reverse shoulder arthroplasties [RSAs]) performed over a 10-year period. Preoperative range of motion, PROMs (American Shoulder and Elbow Surgeons, Simple Shoulder Test, and Shoulder Pain and Disability Index scores), and Constant scores were evaluated. The tipping point for each PROM was evaluated. Univariate and multivariate analyses were performed to assess risk factors for lower tipping points.ResultsPatients undergoing RSA demonstrated lower tipping points for all range-of-motion parameters as well as American Shoulder and Elbow Surgeons, Shoulder Pain and Disability Index, and Simple Shoulder Test scores. Female sex was predictive of a lower tipping point prior to shoulder arthroplasty, regardless of implant type. When the total shoulder arthroplasty subgroup was evaluated, both female sex and a higher body mass index were shown to be associated with a lower tipping point.DiscussionThe choice to undergo shoulder arthroplasty is a multifactorial decision that encompasses both physical and social factors. Female patients and patients undergoing RSA are more likely to accept slightly worse shoulder function prior to making the decision to undergo shoulder arthroplasty.

Project description:Background Chronic kidney disease (CKD) is the progressive loss of renal function. Although advances have been made in understanding the progression of CKD, key molecular events in complex pathophysiological mechanisms that mark each stage of renal failure remain largely unknown. Changes in plasma protein profiles in different disease stages are important for identification of early diagnostic markers and potential therapeutic targets. The goal of this study was to determine the molecular profile of each CKD stage (from 1 to 5), aiming to specifically point out markedly expressed or downregulated proteins. Methods We performed a cross-sectional shotgun-proteomic study of plasma proteome across CKD stages and compared them to healthy controls. After sample pooling and heparin-column purification we analysed proteomes from healthy to CKD stage 1 through 5 participants’ plasma by liquid-chromatography/mass-spectrometry. Results We identified 298 proteins in all study groups, as well as several outliers. Our results indicate that key events, that may later affect the course of disease progression and the overall pathophysiological background, are most pronounced in CKD stage 2, with an emphasis on inflammation, lipoprotein metabolism, angiogenesis and tissue regeneration. Conclusions Our data is consistent with multiple previous studies in CKD research, but also uncovers previously unknown CKD mediators and brings forth several novel potential protein-candidates which might have a significant impact on the development of diagnostic and therapeutic strategies for CKD. We hypothesize that CKD stage 2 is the tipping point in disease progression and a suitable point in disease course for the development of therapeutic solutions.
