The mitochondrial genome of Conosia irrorata (Diptera: Limoniidae).

ABSTRACT: The crane fly Conosia irrorata belongs to the subfamily Limnophilinae of Limoniidae. The mitogenome (GenBank accession number: MK864103) of C. irrorata was sequenced, the new representative of the mitogenome of the subfamily. The nearly complete mitogenome is 14,634 bp totally, consisting of 13 protein-coding genes, two rRNAs, and 22 transfer RNAs. All genes have the similar locations and strands with that of other published species of Limoniidae. The nucleotide composition biases toward A and T, which together made up 77.5％ of the entirety. Bayesian inference analysis strongly supported the monophyly of Tipuloidea. It suggested that the monophyletic Tipulidae was assigned to the sister to the monophyletic Cylindrotomidae. The Limoniidae is the sister group to the clade of Tipulidae + Cylindrotomidae, and Pediciidae was assigned to the sister to the group that contain Limoniidae + (Tipulidae + Cylindrotomidae).

Project description:The crane fly Epiphragma (Epiphragma) mediale Mao and Yang, belongs to subfamily Limnophilinae of family Limoniidae. This mitogenome of E. (E.) mediale was sequenced used next-generation sequencing (NGS), the new representative of the mitogenome of the subfamily. The nearly complete mitogenome is 14,858 bp totally, consisting of 13 protein-coding genes, 2 rRNAs and 22 transfer RNAs. All genes have the similar locations and strands with that of other published species of Limoniidae. The nucleotide composition biases toward A and T, which together made up 75.2% of the entirety. Bayesian inference analysis strongly supported the monophyly of Tipuloidea. It suggested that Pediciidae is the basal clade of Tipuloidea and the monophyletic Tipulidae was assigned as the sister to the monophyletic Cylindrotomidae. The phylogenetic relationship within Tipuloidea was as follows: Pediciidae + (Limoniidae + (Tipulidae + Cylindrotomidae)).

The completed mitogenome is 15,985 bp totally, comprising of 13 protein-coding genes, 2 rRNAs, 22 transfer RNAs and control region. All genes have the similar locations and strands with other published species of Tipuloidea. The nucleotide composition biases toward A and T, which together made up 78.00％of the entirety. Bayesian inference analysis strongly supported the monophyly of Chioneinae + Limnophilinae. It suggested that the phylogenetic relationship within Tipuloidea is Pediciidae + (Limoniidae ((Chioneinae + Limnophilinae)+ Limoniinae) + (Tipulidae + Cylindrotomidae)).

Characterization of the complete mitochondrial genome of the snow crane-fly Chionea crassipes gracilistyla (Diptera, Tipuloidea, Limoniidae) with phylogenetic analysis.

Project description:The genus Chionea Dalman, 1816 is a peculiar group of crane flies with extremely short wings. In this study, we report the first complete mitochondrial (mt) genome sequence of the genus Chionea, which is a circular molecule of 15,775 bp with an AT content of 76.9% and contains 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and a long non-coding region. Phylogenetic analysis revealed that the Pediciidae was sister-group to the remaining Tipuloidea and strongly supported the sister-group relationship between Cylindrotomidae and Tipulidae.

Contribution to the Knowledge of Dicranoptychini (Diptera, Tipuloidea, Limoniidae) in China, with the First Mitochondrial Genome of the Tribe and Its Phylogenetic Implications.

Project description:Dicranoptychini is a tribe in the subfamily Limoniinae (Diptera, Tipuloidea, and Limoniidae) and includes only the genus Dicranoptycha Osten Sacken, 1860. However, the species diversity of the tribe in China was seriously underestimated, and the taxonomic status of Dicranoptycha has long been controversial. In this study, types of Chinese Dicranoptycha species and specimens collected from several localities in China were examined, and the first mitochondrial (mt) genome of the tribe Dicranoptychini is presented. Two Dicranoptycha species, D. jiufengshana sp. nov. and D. shandongensis sp. nov., from China, are described and illustrated as new to science. A Palaearctic species, D. prolongata Alexander, 1938, is recorded in China for the first time. In addition, the complete mt genome of D. shandongensis sp. nov. is sequenced and annotated, indicating that it is a typical circular DNA molecule with a length of 16,157 bp and shows a similar gene order, nucleotide composition, and codon usage to mt genomes of other Tipuloidea species. The two pairs of repeat elements are found in its control region. Phylogenetic results confirm the sister-group relationship between Cylindrotomidae and Tipulidae, question the position of the genus Epiphragma Osten Sacken, 1860 in Limoniidae, and indicate that Dicranoptychini may be a basal lineage within Limoniinae.

Hexatoma crane flies (Diptera, Limoniidae) of Korea.

Project description:Hexatoma Latreille, 1809 is a large genus of short-palped crane flies with a worldwide distribution. Accounting for more than 60 percent of global species, 362 extant species occur in Asia. Prior to our study, Hexatoma crane flies on the Korean Peninsula (both the Democratic People's Republic of Korea and the Republic of Korea) had been studied for 90 years starting in 1930, but only three species had been recorded, with one of them being a misidentification. This study adds six species to the fauna of the Korean Peninsula, all of which belong to the subgenus H. (Eriocera) Macquart, 1838. General information on genus and subgenus morphological characters is presented in this paper, with a redescription of each species based on Korean specimens, as well as illustrations of both sexes, the elevation range, the period of activity, habitat information, general distribution, and a distribution map for the Korean Peninsula. Three species H. (E.) ilwola Podenas, sp. nov., H. (E.) pianigra Podenas, sp. nov. and H. (E.) serenensis Podenas, sp. nov. are described as new to science. Hexatoma (E.) lygropis (Alexander, 1920) is deleted from the Korean species list as a misidentification. This publication is a continuation of our previous work on short-palped crane flies (Limoniidae) from Korea.

Hexatoma (Eriocera) Macquart (Diptera, Limoniidae) from Xizang, China.

Project description:One new species of the subgenus Eriocera Macquart,1838, Hexatoma (Eriocera) xizangensissp. nov. is described and illustrated from Xizang, China. The following four species are re-described and reported from Xizang for the first time: H. (E.) latigrisea Alexander, 1971, H. (E.) nepalensis (Westwood, 1836), H. (E.) paragnava Alexander, 1973 and H. (E.) perhirsuta Alexander, 1973. A key to the species of Eriocera from Xizang is presented.

Molecular phylogeny and biogeography of the Hawaiian craneflies Dicranomyia (Diptera: Limoniidae).

Project description:The Hawaiian Diptera offer an opportunity to compare patterns of diversification across large and small endemic radiations with varying species richness and levels of single island endemism. The craneflies (Limoniidae: Dicranomyia) represent a small radiation of 13 described species that have diversified within the Hawaiian Islands. We used Bayesian and maximum likelihood approaches to generate a molecular phylogeny of the Hawaiian Dicranomyia using a combination of nuclear and mitochondrial loci, estimated divergence times and reconstructed ancestral ranges. Divergence time estimation and ancestral range reconstruction suggest that the colonization that led to most of the diversity within the craneflies arrived prior to the formation of Kauai and demonstrates that the two major clades within that radiation contrast sharply in their patterns of diversification.

New species of the subgenus Epiphragma Osten Sacken from China (Diptera, Limoniidae).

Project description:Previously 12 species of the subgenus Epiphragma (Epiphragma) Osten Sacken, 1860 were known to occur in China. The following three species are described and illustrated as new to science: E. (E.) acuminatumsp. nov., E. (E.) henanensissp. nov., and E. (E.) longitubumsp. nov.. Epiphragma (E.) insigne van der Wulp, 1878 is reported from China for the first time and is illustrated based on additional morphological characters. An updated key to the species of the subgenus E. (Epiphragma) from China is presented.

A key to the four Japanese species of the genus is provided, with images of habitus and wings, and drawings of their male terminalia. Ulomorpha amamiana Kato & Kolcsár, sp. nov. is the first representative of the genus discovered from the Oriental region.

Two new species of Limonia Meigen, 1803 from Northwest China (Diptera, Limoniidae).

Project description:Two new species of Limonia Meigen, 1803, Limonia medexocha sp. nov. and Limonia subcosta sp. nov. are described and illustrated from Northwest China. The following five species are re-described and reported from China for the first time: L. macrostigma (Schummel, 1829), L. phragmitidis (Schrank, 1781), L. stigma (Meigen, 1818), L. sylvicola (Schummel, 1829) and L. taurica (Strobl, 1895). A key to adult males of Limonia from Northwest China is presented.
