The mitochondrial genome of the gold-ringed cowry Monetaria annulus (Mollusca: Gastropoda: Cypraeidae) determined by whole-genome sequencing.

ABSTRACT: The mitochondrial genome of the cowry snail Monetaria annulus (Caenogastropoda: Cypraeoidea: Cypraeidae) was determined by whole-genome next-generation sequencing. The mitogenome is composed of 13 protein-coding genes (PCGs), 2 ribosomal RNA (12S and 16S) genes, and 22 transfer RNA genes (tRNAs). This gene order is consistent with the previously published mitochondrial genomes of other species belonging to the order Littorinimorpha. The superfamily Cypraeoidea was recovered as a sister clade to the group of Tonnoidea and Neogastropoda.

Project description:A new morphologically distinct species of cowry (family Cypraeidae Rafinesque, 1815) is described from the Pliocene of Flinders Island, Tasmania. Austrocypraeajimgracei sp. nov. differs morphologically from other members of the genus and is particularly characterised by the development of a heavily callused labral margin, with a distinct marginal edge that bends up towards the dorsum centrally. This feature is unique within the genus. The new taxon is only the second known Austrocypraea from the Pliocene. A revised key to the known Austrocypraea fossil species is presented.

Complete mitochondrial genome analysis of Sakuraeolis japonica (Baba, 1937) (Mollusca, Gastropoda, Nudibranchia).

Project description:In this study, a complete mitochondrial genome of a nudibranch species, Sakuraeolis japonica was sequenced and analyzed. The mitochondrial genome size is 15,059 bp with 28.1% A, 14.9% C, 19.3% G, and 37.7% T nucleotide distributions. This is the eighth record for complete mitochondrial genome of the Nudibranchia and first record for the genus. Furthermore, phylogenetic relationship of S. Japonica in the Nudibranchia was investigated by using protein-coding genes of complete mitochondrial genome. The present study suggests that S. japonica belongs to the family Facelinidae and it is placed in monophyletic Nudibranchia. The closest species to S. japonica are Chromodoris magnifica and Chromodoris quadricolor that belong to the family Chromodorididae.

Complete sequence analysis of the mitochondrial genome of Auriculastra duplicata (Mollusca, Gastropoda, Ellobiidae).

Project description:The mitochondrial genome of the gastropod Auriculastra duplicata was completely sequenced. It was 13,920 bp in length and comprised 37 genes; two rrn genes and 22 trn genes. Phylogenetic analyses based on the concatenated protein-coding genes depicted the polyphyly of all species belonging to the family Ellobiidae; however, monophyly was observed among all species belonging to the subfamily Ellobiinae, in which A. duplicata clustered consistently with Auriculinella bidentata.

The complete mitochondrial genome of Galba pervia (Gastropoda: Mollusca), an intermediate host snail of Fasciola spp.

Project description:Complete mitochondrial (mt) genomes and the gene rearrangements are increasingly used as molecular markers for investigating phylogenetic relationships. Contributing to the complete mt genomes of Gastropoda, especially Pulmonata, we determined the mt genome of the freshwater snail Galba pervia, which is an important intermediate host for Fasciola spp. in China. The complete mt genome of G. pervia is 13,768 bp in length. Its genome is circular, and consists of 37 genes, including 13 genes for proteins, 2 genes for rRNA, 22 genes for tRNA. The mt gene order of G. pervia showed novel arrangement (tRNA-His, tRNA-Gly and tRNA-Tyr change positions and directions) when compared with mt genomes of Pulmonata species sequenced to date, indicating divergence among different species within the Pulmonata. A total of 3655 amino acids were deduced to encode 13 protein genes. The most frequently used amino acid is Leu (15.05%), followed by Phe (11.24%), Ser (10.76%) and IIe (8.346%). Phylogenetic analyses using the concatenated amino acid sequences of the 13 protein-coding genes, with three different computational algorithms (maximum parsimony, maximum likelihood and bayesian analysis), all revealed that the families Lymnaeidae and Planorbidae are closely related two snail families, consistent with previous classifications based on morphological and molecular studies. The complete mt genome sequence of G. pervia showed a novel gene arrangement and it represents the first sequenced high quality mt genome of the family Lymnaeidae. These novel mtDNA data provide additional genetic markers for studying the epidemiology, population genetics and phylogeographics of freshwater snails, as well as for understanding interplay between the intermediate snail hosts and the intra-mollusca stages of Fasciola spp..

The Complete Mitochondrial Genome of Entemnotrochus rumphii, a Living Fossil for Vetigastropoda (Mollusca: Gastropoda).

Project description:Pleurotomarioidea represents a truly isolated and basally diverging lineage in Vetigastropoda (Mollusca: Gastropoda) whose fossil record can date back to the late Cambrian, thus providing rare insights into the evolutionary history of molluscs. Here, we sequenced and assembled the complete mitochondrial genome of one representative species from Pleurotomarioidea-Entemnotrochus rumphii (Schepman, 1879)-of which the mitogenome is 15,795 bp in length, including 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNA genes. The nucleotide composition was biased toward AT, and A + T content reached 65.2%. E. rumphii was recovered as sister to all other living vetigastropods according to mitogenome-based phylogenetic analysis. The mitochondrial gene order was consistent with major vetigastropods and the hypothetical ancestral gastropoda, suggesting the deep conservation of mitogenome arrangement in Vetigastropoda.

Project description:In this study, complete nucleotide sequences of the mitochondrial genome of a nudibranch species, Hypselodoris festiva (A. Adams, 1861) were determined and characterized. The mitogenome size is 14 880 bp. This is the longest among the known nudibranch mitochondrial genomes. Furthermore, phylogenetic relationship of H. festiva in the Nudibranchia reconstructed due to amino acid sequences of mitochondrial protein coding genes. This is the sixth record for complete mitochondrial genome of the Nudibranchia.

Project description:The file ramshorn snail Planorbella pilsbryi Baker, 1926 (Gastropoda: Hygrophila: Planorbidae) is a widespread herbivorous North American freshwater snail found in diverse habitats, including standing and moving water bodies. Genome skimming by Illumina sequencing allowed the assembly of a complete nuclear rRNA repeat sequence and a complete circular mitogenome of 13,720 bp from P. pilsbryi consisting of 75.3% AT nucleotides, 22 tRNAs, 13 protein-coding genes, 2 rRNAs and a control region in the typical order found in panpulmonate snails. Planorbella pilsbryi COXI features a rare TTG start codon while COXII, CYTB, ND2, ND3, and ND5 exhibit incomplete stop codons completed by the addition of 3' A residues to the mRNA. Phylogenetic reconstruction of mitochondrial protein-coding gene and rRNA sequences places P. pilsbryi as sister taxon to Planorbella duryi (Planorbidae) within family Planorbidae, which is consistent with previous phylogenetic hypotheses.

The mitochondrial genome of Erronea caurica (Cypraeidae).

Project description:The mitochondrial genome of Erronea caurica from the South China Sea has been determined (GenBank Accession No. MT522622), which was the second report of mitochondrial genome in the superfamily Cypraeoidea. It is 16,053 bp long and consists of 21 tRNA genes, 2 rRNA genes, 13 protein-coding genes, and 1 control region. As previously reported mitochondrial genome in Cypraeoidea, all protein-coding genes of E. caurica use a typical start codon (ATN) and a complete stop codon (TAA or TAG). Phylogenetic tree demonstrated that E. caurica belongs to the family Cypraeoidea and closer to the superfamily Tonnoidea.

Complete mitochondrial genome sequences and phylogenetic relationship of Elysia ornata (Swainson, 1840) (Mollusca, Gastropoda, Heterobranchia, Sacoglossa).

Project description:The sacoglossans are small sea slugs in the Heterobranchia. In this study, mitochondrial genome of a sacoglossan sea slug, Elysia ornata (Swainson, 1840) is analyzed and phylogenetic tree of the species in the Heterobranchia is reconstructed based on amino acid sequences of mitochondrial protein coding genes. The phylogenetic relationship analysis shows that E. ornata belongs to the monophyletic Sacoglossa in the Heterobranchia and the Sacoglossa shows sister group relationship with a lineage including Anaspidea and Siphonarioidea. This is the fourth record for the mitochondrial genome of sacoglossan sea slugs.

De novo genome assembly of the land snail Candidula unifasciata (Mollusca: Gastropoda).

Project description:Among all molluscs, land snails are a scientifically and economically interesting group comprising edible species, alien species and agricultural pests. Yet, despite their high diversity, the number of genome drafts publicly available is still scarce. Here, we present the draft genome assembly of the land snail Candidula unifasciata, a widely distributed species along central Europe, belonging to the Geomitridae family, a highly diversified taxon in the Western-Palearctic region. We performed whole genome sequencing, assembly and annotation of an adult specimen based on PacBio and Oxford Nanopore long read sequences as well as Illumina data. A genome draft of about 1.29 Gb was generated with a N50 length of 246 kb. More than 60% of the assembled genome was identified as repetitive elements. In total, 22,464 protein-coding genes were identified in the genome, of which 62.27% were functionally annotated. This is the first assembled and annotated genome for a geometrid snail and will serve as reference for further evolutionary, genomic and population genetic studies of this important and interesting group.
