Characterization of the complete mitochondrial genome of Oryzaephilus surinamensis Linne (Insecta: Coleoptera: Silvanidae) from Xichuan.

ABSTRACT: The saw-toothed grain beetle, Oryzaephilus surinamensis Linné, is a well-known stored-product insect. Beetles were obtained from Xichuan County and the mitochondrial genome was characterized (GenBank accession number MN535903). The mitogenome consists of a circular DNA molecule of 15,941 bp, with only 27.36% GC content. It comprises 13 protein-coding, 22 tRNA, and 2 rDNA genes. The protein-coding genes have typical ATN (Met) initiation codons and are terminated by typical TAN stop codons.

The mitogenome comprises 13 protein-coding, 22 tRNA, and 2 rDNA genes. The protein-coding genes have typical ATN (Met) initiation codons, and are terminated by typical TAN stop codons. A phylogenetic tree generated by the neighbour-joining method showed that B. rynchopetera is closely related to Platydema sp. PLA01, which has the potential for medicinal development.

Project description:The flightless intertidal beetle genus Aegialites (family Salpingidae) is distributed along the Northern Pacific coasts, from California to Alaska and from Northern Japan to Kamchatka. Systematics of Aegialites and its phylogenetic relationships to other members of Salpingidae are unclear, and little genetic information is available. We here present the first complete mitochondrial genome of this genus, represented by Aegialites californicus (Motchoulsky, 1845) from Sonoma County, California, U.S.A. The complete mitochondrial genome of A. californicus is 15,899 bp long and comprises 13 protein-coding (PCG), two ribosomal RNA (rRNA) and 22 transfer RNA (tRNA) genes. The phylogenetic analysis places A.californicus as sister to other members of family Salpingidae. The mitochondrial genome sequence of A. californicus will contribute to future phylogenetic and taxonomic studies of genus Aegialites, family Salpingidae and superfamily Tenebrionoidea.

The mitochondrial genome of T. obscurus is a circular molecule of 15,771 bp (GenBank accession number MG739327), with A + T bias of 72.69%. It comprises 13 protein-coding, 22 tRNA and two rDNA genes. The protein-coding genes had typical ATN (Met) as the initiation codon and were terminated by the typical stop codon TAN, except for COX2, COX3, nadh5 and nadh4. The phylogenetic tree based on maximum-likelihood method showed that the phylogenetic position of T. obscurus is closely related to T. molitor.

Complete mitochondrial genome and phylogenetic analysis of Mastax latefasciata Liebke 1931 (Insecta: Coleoptera: Carabidae).

Project description:The genus Mastax Fischer von Waldheim 1827 belongs to the family Carabidae. Specimens of adult Mastax latefasciata Liebke, 1931 were collected from Yājì Hill, Huáihuà City, Húnán Province, China. The complete mitochondrial genome (GenBank accession number ON674050.1) of M. latefasciata was sequenced, annotated, and characterized. The results showed that it was a circular DNA molecule of 16,735 bp with 81.07% AT content and comprised 13 protein-coding genes (PCG), 22 tRNA genes, 2 rRNA genes, and 1 control region. The PCGs were initiated using typical ATN (Met) and TTG (Met) start codons and terminated using typical TAN stop codons. The phylogenetic position of Mastax within the Carabidae was first evaluated using complete mitogenomes, and the results showed that it was close to Cicindela anchoralis and Manticora tibialis.

Complete mitochondrial genome of Morphostenophanes yunnanus (Zhou, 2020) (Insecta: Coleoptera: Tenebrionidae) and phylogenetic analysis.

Project description:Morphostenophanes yunnanus (Zhou, 2020) is widely distributed from central to eastern Yunnan with distinct geographical variations in morphology. Beetles were collected in Manwan Town, and a mitochondrial genome sequence (GenBank accession number MZ298928) of this species was sequenced using the MGI-SEQ 2000 platform, assembled using NOVOPlasty v4.3.1, and characterized. The mitogenome was a circular DNA molecule of 15,690 bp with 64.710% AT content, which comprised 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one control region. The protein-coding genes showed the typical ATN (Met) and TTG (Met) start codons, except nad1 and cox1 (TTG as start codon), and were terminated by typical TAN stop codons. The maximum-likelihood polygenetic tree was generated using protein sequences of thirteen protein-coding regions of seventeen mitogenomes with mtREV + G + F + I with 1000 replicates under the Bayesian information criterion using MEGA 11, which showed that M. yunnanus was the most closely related to M. sinicus. This study provides essential genetic and molecular data for phylogenetic analyses of the genus Morphostenophanes.
