The complete chloroplast genome sequence of a traditional Chinese medicine: Achyranthes bidentata (Amaranthaceae).

ABSTRACT: Achyranthes bidentata (Amarathaceae) has been commonly used as a traditional Chinese medicine in the treatment of osteoporosis and bone nonunion. Here, the complete chloroplast genome of A. bidentata was assembled and characterized. The cp genome is 151,451 bp in length, composed of a pair of 25,150 bp inverted repeat (IR) regions separated by a large single-copy (LSC) region of 83,899 bp and a small single-copy (SSC) region of 17,252 bp. The whole cp genome of A. bidentata contains 130 genes(85 protein-coding genes, 37 tRNAs and eight rRNAs) and the overall GC content is 36.5%. Phylogenetic analysis based on the cp genome data showed that A. bidentata was close to Cyathula capitata.

Project description:Achyranthes bidentata Blume is an important herbal medicine. However, little genetic information about Achyranthes is available. To facilitate species identification, we sequenced its complete chloroplast genome using the Illumina MiSeq platform. The data show that the chloroplast genome of A. bidentata is 151,441 bp in size and comprises an 83,890 bp large single-copy region, a 17,261 bp small single-copy region, and two inverted repeat (IR) regions, IRa and IRb (each 25,145 bp). The genome contains 112 unique genes, including 79 protein-coding genes, 4 ribosomal RNAs genes, and 30 transfer RNAs genes. Phylogenetic analysis revealed that A. bidentata is most closely related to Alternanthera philoxeroides.

The complete cp genome is 151,543 bp in length and includes a large single-copy (LSC) region of 83,922 bp, a small single-copy (SSC) region of 17,251 bp, and a pair of inverted repeat regions (IRs) of 25,185 bp. It encodes 131 genes, including 86 protein-coding genes, eight rRNAs, and 37 tRNAs. Phylogenetic analysis indicates that four samples of A. bidentata formed a clade with a 100% bootstrap value.

Project description:Uncaria macrophylla (Rubiaceae) is a medicinal vine plant of the Rubiaceae family that was distributed in East Asia and Southeast Asia. The first complete chloroplast genome of Uncaria macrophylla was sequenced and assembled in this study. The genome is 155,138 bp in length and contained 129 encoded genes in total, including 79 protein-coding genes, eight ribosomal RNA genes, and 37 transfer RNA genes. The phylogenomic analysis showed that U. macrophylla was closely related to Uncaria rhynchophylla according to the current sampling extent.

Complete chloroplast genome sequence of Pueraria thomsonii, an important traditional Chinese medicine plant.

Project description:Pueraria thomsonii is a leguminous plant with high root yield and starch content. It is also a medicinal material in the Chinese pharmacopeia. However, the raw materials of P. thomsonii are often confused with some non-medicinal Pueraria plants. To enrich the genetic resources of P. thomsonii and guide its molecular identification, the complete chloroplast genome was sequenced and reported. The total genome of P. thomsonii is 153,434 bp in length. consisting of two inverted repeat regions (IRS, 25,640 bp each) separated by a large single-copy (LSC, 84,155 bp) and a small single-copy region (SSC, 17,999 bp). The overall GC content is 35.41%. It contains 130 genes, including 85 protein coding genes, 8 rRNA genes and 37 tRNA genes. Phylogenetic analysis showed that P. thomsonii could be distinguished from other plants and closely related to the legume Pachyrhizus erosus. This study enriches the genetic information of P. thomsonii and contributes to the screening of excellent germplasm.

Project description:Tribulus terrestris is an important traditional medicine in China, which is widely distributed in north China. Here, the chloroplast genome sequences were detected. The chloroplast genome of T. terrestris is circular-mapping molecule of 158,184 bp in size, which consisted of a pair of inverted repeat regions of 25,842 bp each, a large single copy region of 88,878 bp, and a small single copy region of 17,622 bp. A total of 129 genes were annotated, including 37 tRNA, 8 rRNA, and 84 protein-coding genes. Phylogenetic analysis showed T. terrestris clustered with Krameria lanceolate and Krameria bicolor.

The complete chloroplast genome sequence of Mentha canadensis (Labiatae), a traditional Chinese herbal medicine.

Project description:Mentha canadensis is a well-known resource of traditional Chinese herbal medicine, belonging to the Mentha of the Labiatae family. In this study, the whole chloroplast genome of the M. canadensis chloroplast genome was sequenced, assembled and annotated, which contains 134 unique genes, including 89 protein-coding genes, 37 tRNA genes and 8 rRNA genes. A maximum likelihood phylogenetic tree based on 11 complete chloroplast genomes revealed that M. canadensis is closely related to M. longifolia and M. spicata. The chloroplast genome could be used for variety identification, genetic engineering and effective protection of germplasm resources.

The traditional Chinese medicine Achyranthes bidentata and our de novo conception of its metastatic chemoprevention: from phytochemistry to pharmacology.

Project description:Our recent biosystems analysis revealed similarities between embryonic implantation and cancer cell adhesion, which suggests that abortifacients may be good for safe and effective metastatic chemoprevention targeting circulating tumor cells (CTC). Here we test the hypothesis by using the well-known abortion herb Achyranthes bidentata Blume (A. bidentata). Five compounds were separated from the herb root. Among them, ginsenoside Ro was the most potent in inhibiting embryonic implantation within non-cytotoxic concentrations. It specifically inhibited the metastatic dissemination capability of colon cancer cells HT29, including the migration and invasion ability, and their adhesion to human endothelium through inhibiting integrin αvβ6, MMP-2, MMP-9, and ERK phosphorylation by HT29. Pretreatment of nude mice with oral ginsenoside Ro followed by HT29 intravenous inoculation and 40-day oral ginsenoside Ro significantly prevented lung metastasis with downregulation of integrin αvβ6 and no toxicity. The present study firstly introduces the new conception of utilizing safe and effective abortion botanic medicines for CTC-based metastatic chemoprevention.

The genome of B. disciflorum is 148,554 bp in length, including a large single-copy (LSC) of 79,001 bp, a small single-copy (SSC) of 16,797 bp, and a pair of inverted repeat regions (IRa and IRb) of 26,378bp. It contains 108 unique genes consisting of 74 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. The phylogenetic analysis revealed that Bulbophyllum is sister to the genus of Dendrobium.

The chloroplast genome of S. angustata was 155,869 bp in length, containing a large single-copy region (84,343 bp), a small single-copy region (18,820 bp), and two inverted repeat regions (26,353 bp). The overall GC content of S. angustata chloroplast genome was 36.80%. The chloroplast genome of S. angustata contained 127 unique genes, including 83 protein-coding genes, 36 tRNA genes and eight rRNA genes. Phylogenetic analysis revealed that S. angustata was related to Malus ioensis, Malus florentina and Malus trilobata.
