Characterization and phylogenetic analysis of the complete mitochondrial genome of Conidiobolus sp. (Entomophthorales: Ancylistaceae).

ABSTRACT: In the present study, we presented the complete mitochondrial genome of an entomophthoroid fungus Conidiobolus sp. The mitogenome of Conidiobolus sp. has a total length of 26,612 bp, with the base composition as follows: A (44.22%), T (27.10%), C (10.99%) and G (17.68%). The mitogenome contains 19 protein-coding genes, 2 ribosomal RNA genes (rRNA), and 23 transfer RNA (tRNA) genes. The taxonomic status of the Conidiobolus sp. mitogenome exhibited a close relationship with the mitogenome of Conidiobolus heterosporus.

Project description:Neoconidiobolus thromboides is a pandemic species in the genus Neoconidiobolus. In this article, we report the first complete sequence of mitochondrial genome from a common entomophthoroid fungus Neoconidiobolus thromboides under Illumina next-generation sequencing system. The total length of the mitogenome is 34,984 bp with a GC content of 26.99%. The gene annotation revealed 56 genes, including 30 protein-coding genes (PCGs), two ribosomal RNA genes (rDNAs), 24 transfer RNA (tRNA) genes. Phylogenetic analyses of 14 concatenated conserved PCGs indicated that N. thromboides was grouped with Capillidium heterosporum and Conidiobolus sp.

The complete mitochondrial genome of Microconidiobolus nodosus (Entomophthorales: Ancylistaceae).

Project description:In this study, the complete mitochondrial genome of Microconidiobolus nodosus was sequenced which is the first mitochondrial genome of the genus. The mitochondrial genome is 31,638 bp long and 27.18% in GC ratio, and it contains 14 conserved protein-coding genes, 2 ribosomal RNAs and 22 transfer RNAs. Phylogenetic analysis showed that M. nodosus was closely related to Conodiobolus sp. This study reported the whole mitochondrial genome and character of a basal fungus M .nodosus and provided a better understanding of the phylogeny of basal fungi.

A taxonomic revision of the genus Conidiobolus (Ancylistaceae, Entomophthorales): four clades including three new genera.

Project description:The genus Conidiobolus is an important group in entomophthoroid fungi and is considered to be polyphyletic in recent molecular phylogenies. To re-evaluate and delimit this genus, multi-locus phylogenetic analyses were performed using the large and small subunits of nuclear ribosomal DNA (nucLSU and nucSSU), the small subunit of the mitochondrial ribosomal DNA (mtSSU) and the translation elongation factor 1-alpha (EF-1α). The results indicated that the Conidiobolus is not monophyletic, being grouped into a paraphyletic grade with four clades. Consequently, the well-known Conidiobolus is revised and three new genera Capillidium, Microconidiobolus and Neoconidiobolus are proposed along with one new record and 22 new combinations. In addition, the genus Basidiobolus is found to be basal to the other entomophthoroid taxa and the genus Batkoa locates in the Entomophthoraceae clade.

Characterization and phylogenetic analysis of the complete mitochondrial genome of Rhinogobius sp. (Perciformes, Gobiidae).

Project description:The genus Rhinogobius was widely distributed in East Asia. In the present study, the complete mitochondrial genome of Rhinogobius sp., possible a new species of freshwater goby from Anhui province of China, was sequenced for the first time. Sequence analysis showed that it is 16,511 bp in length with A + T content of 52.3%, consisting of 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and a control region (CR). Phylogenetic analyses placed Rhinogobius sp. in a well-supported monophyletic cluster with other Rhinogobius fish and the phylogenetic position of Rhinogobius sp. was closer to Rhinogobius cliffordpopei.

Mitochondrial genome characterization and phylogenetic analysis of Blastocladiella sp. (Blastocladiales: Blastocladiaceae).

Project description:In the present study, the complete mitochondrial genome of an early diverging fungus Blastocladiella sp. was assembled by the next-generation sequencing. The complete mitochondrial genome of Blastocladiella sp. is 33, 800 bp in length and consists of 11,620 (34.38%) adenine, 5,047 (14.93%) cytosine, 6,025 (17.83%) guanosine and 11,108 (32.86%) thymine. The genome contains 19 protein-coding genes, 24 tRNA genes and 2 rRNA genes. Phylogenetic analysis based on the combined mitochondrial gene set showed that Blastocladiella sp. has a close relationship with Allomyces macrogynus and Blastocladiella emersonii.

The complete mitogenome of Cryptococcus sp. was composed of circular DNA molecules, with a total length of 30,029 bp. The mitogenome contains 20 protein-coding genes, 2 ribosomal RNA genes (rRNA), and 22 transfer RNA (tRNA) genes. Phylogenetic analysis showed that the mitogenome of the Cryptococcus sp. exhibited a closest relationship with Cryptococcus gattii.

The complete mitochondrial genome and phylogenetic analysis of Hiatella sp. J (Mollusca: Hiatellidae).

Project description:The genus Hiatella is one of most abundant and widespread marine bivalves. To date, its intra-generic phylogeny remains disputed and mitogenome information is therefore much needed. Here, we first report the complete circular mitogenome of Hiatella sp. J that is distributed in the coast of Asia Pacific. The total length of this mitochondrial genome is 21,233 base pairs. It consists of 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs, and a major noncoding region (MNR). Phylogenetic analysis of COI from 25 species (Hiatellidae) revealed that the Hiatella sp. J was closely related to Asian Hiatella in the family Hiatellidae. This Hiatella mitogenome provides new molecular data for the further taxonomic and phylogenetic studies of the genus Hiatella of marine bivalves.

Characterization and phylogenetic analysis of the complete mitochondrial genome sequence of Photinia serratifolia.

Project description:Plant mitochondrial genomes (mitogenomes) are a valuable source of genetic information for a better understanding of phylogenetic relationships. However, no mitogenome of any species in the genus of Photinia has been reported. In this study, using NGS sequencing, we reported the mitogenome assembly and annotation of Photinia serratifolia, which is 473,579 bp in length, contains 38 protein-coding genes, 23 tRNAs, and 6 rRNAs, with 61 genes have no introns. The rps2 and rps11 genes are missing in the P. serratifolia mitogenome. Although there are more editing sites (488) in the P. serratifolia mitogenome than in most angiosperms, fewer editing types were found in the P. serratifolia mitogenome, showing a clear bias in RNA-editing. Phylogenetic analysis based on the mitogenomes of P. serratifolia and 8 other taxa of the Rosaceae family reflected the exact evolutionary and taxonomic status of P. serratifolia. However, Ka/Ks analysis revealed that 72.69% of the protein-coding genes in the P. serratifolia mitogenome had undergone negative selections, reflecting the importance of those genes in the P. serratifolia mitogenome. Collectively, these results will provide valuable information for the evolution of P. serratifolia and provide insight into the evolutionary relationships within Photinia and the Rosaceae family.

Complete mitochondrial genome of Rhodeuscyanorostris (Teleostei, Cyprinidae): characterization and phylogenetic analysis.

Project description:Rhodeuscyanorostris Li, Liao & Arai, 2020 is a freshwater fish that is endemic to China and restricted to Chengdu City in Sichuan Province. This study is the first to sequence and characterize the complete mitochondrial genome of R.cyanorostris. The mitogenome of R.cyanorostris is 16580 bp in length, including 13 protein-coding genes, two rRNA genes, 22 tRNA genes, and a control region (D-loop). The base composition of the sequence is 28.5% A, 27.6% C, 26.4% T, and 17.5% G, with a bias toward A+T. The genome structure, nucleotide composition, and codon usage of the mitogenome of R.cyanorostris are consistent with those of other species of Rhodeus. To verify the molecular phylogeny of the genus Rhodeus, we provide new insights to better understand the taxonomic status of R.cyanorostris. The phylogenetic trees present four major clades based on 19 mitogenomic sequences from 16 Rhodeus species. Rhodeuscyanorostris exhibits the closest phylogenetic relationship with R.pseudosericeus, R.amarus, and R.sericeus. This study discloses the complete mitochondrial genome sequence of R.cyanorostris for the first time and provides the most comprehensive phylogenetic reconstruction of the genus Rhodeus based on whole mitochondrial genome sequences. The information obtained in this study will provide new insights for conservation, phylogenetic analysis, and evolutionary biology research.

Complete mitochondrial genome of Caranx equula (Perciformes, Carangidae): genome characterization and phylogenetic analysis.

Project description:Caranx equula is an important marine fish which due to its commercial values. However, some wild populations of C. equula are in danger because of the overfishing and environmental pollution. In this study, the complete mitochondrial genome of C. equula was firstly determined. The complete mitochondrial genome is 16,607 base pairs in length, and contained 13 protein-coding genes and 2 ribosomal RNA genes, 22 tRNA genes and 2 main non-coding regions. Overall A + T content was 52.9%. In addition, a phylogenetic tree was constructed using the complete mitochondrial genome and showed that C. equula clustered in a clade and formed a sister relationship with Caranx ignobilis belonged to the family of Carangidae.
