The complete mitochondrial genome of Amesia sanguiflua (Lepidoptera, Zygaenidae).

ABSTRACT: Amesia sanguiflua (Lepidoptera, Zygaenidae) is found in northern India, Myanmar, Indochina, Malay Peninsula, Sumatra, Java, and China. In the present study, we sequenced the complete mitochondrial genome of A. sanguiflua. The mitochondrial genome was 15,203 bp in length, containing a typical set of 37 genes (13 protein-coding genes, 2 rRNA genes, 22 tRNA genes) and a 346bp non-coding A+T-rich region. Phylogenetic analysis using mitochondrial genomes of 40 species showed that A. sanguiflua formed a well-supported monophyletic group with other Zygaenidae species.

Project description:Histia rhodope Cramer, found in India, Chinese, Burma, Indonesia and other Southeast Asian regions, belongs to Lepidoptera family Zygaenidae. In this study, we describe the genomic features of the mitogenome sequences of these insects. The mitogenome of Histia rhodope Cramer is 15,205 bp long consisting a typical set of genes (13 protein-coding genes, 22 tRNA genes and two rRNA genes) and one major 376 bp non-coding A + T-rich region. All PCGs of Histia rhodope Cramer start with ATN codons except cox1 which start with CGA codon and all PCGs stop at TAA codons. Phylogenetic analysis demonstrates that Histia rhodope Cramer and Rhodopsona rbiginosa are clustered together into a monophyletic group Zygaenidae. Zygaenidae is phylogenetically closer to Limacodiae than Tortricidea.

The complete mitochondrial genome of Pidorus atratus (Lepidoptera: Zygaenoidea: Zygaenidae).

Project description:The complete mitochondrial genome (mitogenome) of Pidorus atratus (Lepidoptera: Zygaenoidea: Zygaenidae) is described in this study. The circular molecule is 15,383 bp in length and contains 37 typical mitochondrial genes and one non-coding AT-rich region. All protein-coding genes (PCGs) start with ATN, except for cox1 gene with CGA; 10 of the 13 PCGs harbour the typical stop codon TAA, whereas cox1, cox2, and nad4 end with a single T. Two ribosomal RNA genes (rRNAs) are 1366 bp and 544 bp in length, respectively. The AT-rich region is 658 bp in size and harbours several features characteristic of the lepidopterans, including the motif ATAGA followed by a 20 bp poly-T stretch and a microsatellite-like (TA)9 element. The complete mitogenome data would be useful for further understanding the taxonomy and phylogeny of Zygaenoidea.

Characteristics and phylogenetic analysis of the complete mitochondrial genome of Erasmia pulchella Hope, 1840 (lepidoptera: zygaenidae).

Project description:Erasmia pulchella has brightly colored wings and releases toxic cyanide as a defense against predation. At present, the molecular phylogenetic status of this species is still unclear. Here, we presented the first complete mitochondrial genome of the genus Erasmia, which was assembled from data generated using a genome skimming method. The assembled mitogenome was 15,197 bp in length and consists of 37 genes, including 13 protein-coding genes, two rRNAs, 22 tRNAs, and a control region. Phylogenetic analysis based on both maximum likelihood (ML) and Bayesian inference (BI) revealed that E. pulchella was most closely related to Amesia sanguiflua.

Chromosome-level genome assembly of the forest pest Achelura yunnanensis (Lepidoptera: Zygaenidae).

Project description:Achelura yunnanensis is a destructive pest of forests, causing substantial damage on tree growth and severe economic losses. Additionally, as a daytime-active moth, this species also holds important scientific value for investigating the genetic mechanisms governing day-night activity patterns of Lepidoptera. To facilitate effective pest control and deepen our understanding of the diurnal behavior's genetic basis of moths, genomic data for this species are crucial. In this study, we present a chromosome-level reference genome of A. yunnanensis (368.15 Mb in 32 chromosomes; scaffold N50 = 12.61 Mb; BUSCO completeness = 98.0%). Genome annotation shows that the new assembly comprises 37.10% (136.55 Mb) repetitive elements, 1,828 non-coding RNAs, and 15,523 protein-coding genes. Genes involved in lipid metabolism and xenobiotics biodegradation and metabolism, such as cytochrome P450 families, experienced significant expansion in the A. yunnanensis genome. The chromosome-level genome of A. yunnanensis provides a valuable genomic resource for devising novel pest control strategies, and will also help to study the genetic mechanism of the shift of diurnal behavior in Lepidoptera.

The complete mitochondrial genome of Rondotia menciana (Lepidoptera: Bombycidae).

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.

Complete mitochondrial genome of Cnidocampa flavescens (Lepidoptera: Limacodidae).

Project description:Cnidocampa flavescens, lives in Nepal, Bhutan, China, Far East of Russia, Korea, and Japan, belongs to the Lepidoptera family Limacodidae. In this study, we describe the genomic features of the mitogenome sequences of the insects. The mitogenome of C. flavescens is 15,406 bp long consisting a typical set of genes (13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes) and one major 415 bp non-coding A + T-rich region. All PCGs of C. flavescens start with ATN codons and end with TAA codons. The gene arrangement of C. flavescens mitogenome is same to Monema flavescens while the intergenic spacers and overlaps are different. The 415 bp A + T-rich region contains a conserved ATAGA motif followed a poly-T stretch.

Complete mitochondrial genome of Parnassius glacialis (Lepidoptera: Papilionidae).

Project description:Parnassius glacialis is a butterfly species distributed in China, Korea, Japan. The complete P. glacialis mitochondrial genome was assembled using Illumina sequencing data. The mitogenome is 15,353 bp long and contains 13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes. A phylogenetic analysis of P. glacialis and 14 related Papilionidae species indicated that P. glacialis is clustered with other Parnassius species. This study generated useful genetic information for future studies on the taxonomy, phylogeny, and evolution of Papilionidae species.

Complete mitochondrial genome of Acanthopsyche nigraplaga (Lepidoptera: Psychidae).

Project description:We report the mitochondrial genome (mitogenome) of a case-making moth Acanthopsyche nigraplaga Wileman, 1911 (Lepidoptera: Psychidae). The 15,704 bp long complete mitogenome comprises a typical set of genes [13 protein-coding genes (PCGs), 2 rRNA genes, and 22 tRNA genes] and one major non-coding, A + T-rich region, with an arrangement identical to that observed in most lepidopteran mitogenomes. Twelve of the 13 PCGs of the A. nigraplaga mitogenome initiate with a typical ATN start codon, however COI contains the atypical CGA start codon that is common for lepidopteran COI genes. A phylogenetic analysis using concatenated nucleotide sequences of the 13 PCGs and 2 rRNA genes using the Bayesian inference method fully resolved A. nigraplaga in a monophyletic clade within the Psychidae. Acanthopsyche nigraplaga was situated in a sister position to Eumeta variegata and Mahasena oolona with high nodal support. As more mitogenome sequences are available further scrutinized analysis for the superfamily Tineoidea including Psychidae will be possible.

Complete mitochondrial genome of Mycalesis intermedia (Lepidoptera: Nymphalidae).

Project description:The Mycalesis intermedia belongs to Nymphalidae in Lepidoptera. We described the complete mitogenome of M. intermedia, which is typical circular duplex molecules and 15,386 bp in length, containing the standard metazoan set of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an A + T-rich region with macro-repeat sequences. All the inferred tRNA secondary structures show the common cloverleaf pattern, with the exception of trnS1(AGN) which lacks the DHU arm. The M. intermedia mitochondrial genome has the same gene order with other lepidopterans.

All PCGs initiate with typical start codon of ATN and share the complete stop codon of TAA, whereas cox1 starts with CGA. The ML analysis was performed using a dataset matrix containing 13 PCGs concatenated from the mitogenomes of Geometridae species. Our study presented the phylogenetic relationship of (Larentiinae + ((Sterrhinae + (Ennominae + Geometrinae))). Within the genera Biston, B. thoracicaria grouped with other species as the sister group.
