The complete chloroplast genome sequence of Magnolia mexicana DC. (Magnoliaceae) from Central America.

ABSTRACT: The chloroplast genome of the Magnolia species from Central America has never been reported. With its local use for food flavoring, medicine, and wood, M. mexicana has been of good economic importance. In the present study, the complete chloroplast genome of M. mexicana was assembled via the genome skimming data. As a typical quadripartite structure, the plastome of M. mexicana with 159,906 bp in length includes two inverted repeats (26,554 bp) separated by a small single copy region (18,761 bp) as well as a large single copy region (88,037 bp). This chloroplast genome consists of 131 different genes, including 86 protein coding genes (CDS), eight rRNA genes, and 37 tRNA genes. The maximum likelihood phylogenetic analysis showed that M. mexicana from Central America was closely related to an evergreen species, M. odoratissima from East Asia.

Project description:As an endangered species, Magnolia kobus is distributed in Jeju island in Korea with only about 500-1000 individuals. In this study, we presented a complete chloroplast genome of M. kobus which is 159,443 bp and has four sub-regions: 87,484 bp of large single copy and 18,783 bp of small single copy regions are separated by 26,588 bp of inverted repeat regions including 113 genes (79 unique genes, four rRNAs and 30 tRNAs). Phylogenetic analysis using chloroplast genomes showed that M. kobus is a sister of M. insignis and M. laevifolia clade.

Characterization of the complete chloroplast genome of Magnolia wilsonii (Magnoliaceae).

Project description:The complete chloroplast (cp) genome of Magnolia wilsonii, an endangered and endemic species in China, was first reported in this study. The M. wilsonii cp genome was 157,434 bp in length, including two inverted repeat (IR) regions of 26,868 bp, a large single copy (LSC) region of 85,387 bp, and a small single copy (SSC) region of 18,311 bp. A total of 113 unique genes, including 79 protein-coding genes, 4 ribosomal RNA genes, and 30 transfer RNA genes, were annotated. The overall GC content was 38.8%. Phylogenetic analysis of 34 representative cp genomes within the genus Magnolia suggests that M. wilsonii was sister to M. sieboldii.

Project description:Magnolia coriacea, Chang et B. L. (Magnoliaceae) is a critically endangered tree, endemic to Yunnan province, China. In this study, the complete chloroplast genome of M. coriacea was sequenced and analyzed. The total chloroplast genome size of M. coriacea is 160,113 bp, including a pair of inverted repeat regions (IRs, 26,576 bp) separated by a large single-copy (LSC, 88,175 bp) region and a small single-copy region (SSC, 18,786 bp). The complete chloroplast genome contains 86 protein-coding (PCGs), 37 transfer RNA (tRNAs), and 8 ribosomal RNA (rRNAs) genes. The phylogenetic analysis showed that M. coriacea is closely related to M. cathcartii. This study contributes to the bioinformatics on the evolution, genetic, conservation, and molecular biology for future studies of Magnoliaceae.

Determination of chloroplast genome sequence of Magnolia ofeliae A. Vazquez & Cuevas (Magnoliaceae), an endangered Mexican Magnolia.

Project description:Magnolia ofeliae A. Vázquez & Cuevas, a plant species endemic to south Jalisco, Mexico, is a critically endangered (CR) species based on the IUCN Red List. In this study, we assembled its complete chloroplast (cp) genome. The total genome size of M. ofeliae was 159,839 bp including four subregions: a large single-copy (LSC) region of 88,027 bp and a small single-copy (SSC) region of 18,752 bp separated by a pair of identical inverted repeat regions (IRs) of 26,530 bp each. The GC content of the cp genome of M. ofeliae is 39.3%. The cp genome encoded a set of 113 genes, containing 79 protein-coding genes, 30 tRNA genes, and four rRNA genes. Phylogenetic analysis results that M. ofeliae is a sister to all other magnolias in the subfamily Magnolioideae.

Project description:Magnolia delavayi is a rare, famous ornamental and important medical tree endemic to China. Here, we assembled the complete chloroplast (cp) genome sequence of M. delavayi. Its length is 159,715 bp with four sub-regions: 87,906 bp of large single-copy region and 18,761 bp of small single-copy region are separated by two inverted repeats regions, each 26,524 bp. The genome contains 77 protein-coding genes, 6 rRNAs, and 29 tRNAs genes. Phylogenetic analysis of cp genome of M. delavayi with previously reported chloroplast genomes in Magnolia shows that M. delavayi is close to M. odoratissima with high bootstrap value.

The complete chloroplast genome of a widespread ornamental shrub in China, Magnolia figo (Magnoliaceae).

Project description:Plants in Michelia, presented by Magnolia figo DC, are wonderful resources in Magnoliaceae, covering a series of aromatic plants. Despite extensive studies in this family, the M. figo complete chloroplast genome and the taxonomical status based on the whole chloroplast sequences remain unclear. Herein, we report the complete chloroplast genome of M. figo. The chloroplast genome was 160,113 bp in length, with a large single-copy (LSC) region of 88,113 bp and a small single-copy (SSC) region of 18,797 bp, separated by two inverted repeat (protein-coding) regions of 26,602 bp. A total of 135 CDSs were found, including 129 genes, 85 protein-coding mRNAs, 36 tRNA genes, and eight rRNA genes. The overall GC content was 39.3%, and GC percentages range from 34.3% to 43.2% throughout LSC, IRs, and SSC regions. Phylogenetic analysis showed that M. figo is most closely to Michelia odora and displayed a relationship that three Michelia were nested inside Magnolia. This announcement of the complete M. figo cp genome sequence could provide valuable information for further breeding, cp genetic modification, and phylogenetic study in Magnoliaceae.

Project description:The first complete chloroplast genome (cpDNA) sequence of Magnolia maudiae was determined from Illumina HiSeq pair-end sequencing data in this study. The cpDNA is 160,205 bp in length, contains a large single-copy region (LSC) of 88,249 bp and a small single-copy region (SSC) of 18,806 bp, which were separated by a pair of inverted repeats (IR) regions of 26,575 bp. The genome contains 132 genes, including 87 protein-coding genes, 8 ribosomal RNA genes, and 37 transfer RNA genes. Further phylogenomic analysis showed that M. maudiae was close to Magnolia odora and Magnolia laevifolia in Magnolia genus.

Project description:Manglietia yuyuanensis is an important afforestation and excellent broad-leaved tree species in southern China. In this study, we assembled the complete chloroplast genome of M. yuyuanensis based on the Illumina sequences, sequence analysis showed the genome was 160,078 bp in length presenting a typical quadripartite structure and contains an inverted repeat region (IR, 26,467 b), a small single-copy (SSC) region, and a large single-copy (LSC) region (18,785 and 88,359 bp, respectively). The overall GC content was 39.27%. The sequence contained 128 unique genes, including 81 protein-coding genes, 38 tRNA genes, and 8 rRNA genes. The maximum-likelihood (ML) phylogenetic analysis revealed that M. maudiae was closely related to Manglietia insignis. A phylogenetic analysis revealed that M. yuyuanensis is closely related to Manglietia glaucifolia, with the genus Manglietia.

Complete plastome sequence of Magnolia omeiensis (W.C. Cheng) Dandy (Magnoliaceae).

Project description:Magnolia omeiensis is an evergreen tree with 25 meters heights. It is distributed in tropical 1200-1300 m. Mount Emei. And it has been ranked as a CR (Critically Endangered) species in China. Here, we report and characterize the complete plastome of M. omeiensis in an effort to provide genomic resources useful for promoting its systematics research. The plastome of M. omeiensis is found to possess a total length 160,021 bp with the typical quadripartite structure of angiosperms, contains two Inverted Repeats (IRs) of 26,336 bp, a Large Single-Copy (LSC) region of 88,061 bp and a Small Single-Copy (SSC) region of 19,288 bp. The plastome contains 114 genes, consisting of 80 unique protein-coding genes, 30 unique tRNA genes and 4 unique rRNA genes. The overall A/T content in the plastome of M. omeiensis is 60.70%. The phylogenetic analysis indicated that M. omeiensis is close to Magnolia yunnanensis within Magnoliaceae in this study. The complete plastome sequence of M. omeiensis will provide a useful resource for the conservation genetics of this species as well as for the phylogenetic studies of Magnoliaceae.
