The complete mitogenome of Saccharina cultivar 'Haiyi No.1' (Saccharina japonica × latissima) and the phylogenetic analysis.

ABSTRACT: Here, we reported the complete mitogenome of Saccharina cultivar 'Haiyi No.1' (Saccharina japonica × latissima). This mitogenome had a circular mapping organization with the length of 37,657 bp and contained 66 genes including 35 protein-encoding genes, 3 rRNAs, 25 tRNAs, and 3 open reading frames (ORFs). The AT content accounted for 64.7% of the entire mitogenome. The gene content and gene order were consistent with those reported Saccharina species and cultivars. Phylogenetic analysis showed the phylogenetic relationship among Chinese Saccharina cultivars and demonstrated that 'Haiyi No.1' had a closer relationship with Pingbancai which strongly supported its genetic source.

Here, we reported the complete mitogenome of Saccharina cultivar 'Haiyi No.1' (Saccharina japonica × latissima). This mitogenome had a circular mapping organization with the length of 37,657 bp and contained 66 genes including 35 protein-encoding genes, 3 rRNAs, 25 tRNAs, and 3 open reading frames (ORFs). Project description:Complete mitogenome of Saccharina variety 'Ailunwan' (Saccharina japonica × latissima) was obtained in this work. Circular-mapping mitogenome was 37,657 bp with overall A + T content of 64.71%, encoding 3 rRNAs (23S, 16S and 5S), 25 tRNAs, 35 protein-coding genes and 3 open reading frames (ORFs). Gene content and genome organization of 'Ailunwan' mitogenome were identical to those Saccharina species. From the total alignment of 'Ailunwan' and another reported variety 'Rongfu' (S. japonica × latissima), 33 nucleotide substitutions were detected, and one intergenic region of 19 nucleotides insertion/deletion was found which located between rps3 and rps9. The phylogenetic analysis based on mitogenomes showed that 'Ailunwan' had a closer evolutionary relationship with Saccharina than Laminaria and validated the genus Saccharina and Laminaria were polyphyletic.

Complete mitochondrial genome of Saccharina cultivar 'Hainong No.1' (Saccharina japonica × latissima).

Project description:Here, we sequenced the complete mitogenome of 'Hainong No.1', a hybrid Saccharina cultivar produced by crossing the cultivars 'Zaohoucheng' (♂) and 'Pingbancai' (♀). Circular mapping revealed that the mitogenome was 37,657 bp in length and had an overall AT content of 64.66%, including 35 protein-encoding genes, three ribosomal RNA genes (rRNA), 25 transfer RNA genes (tRNA) and three open reading frames (ORF). A phylogenetic tree constructed from the amino acids separated the six cultivars into two groups; 'Hainong No.1' had a closer evolutionary relationship with the cultivars 'Zaohoucheng', 'Pingbancai' and 'Ailunwan', whereas 'Rongfu' and 'Fujian' formed a distinct cluster. Further comparison between the 'Hainong No.1' and the parental mitogenomes displayed that 'Pingbancai' and 'Zaohoucheng' were the female and male parents, respectively.

Project description:Saccharina japonica is one of the most important economic seaweeds. Here, complete mitogenome of S. japonica cultivar 'Zaohoucheng' were reported. Its circular mapping organization with the length of 37,657 bp had an overall A + T content of 64.70% and encoded three rRNAs, 25 tRNAs, 35 known mitochondrial proteins and three ORFs. From the total alignment of S. japonica and its cultivar 'Zaohoucheng', only 12 nucleotide substitutions were detected. Gene arrangement and component of 'Zaohoucheng' mitogenome were identical to those of reported Saccharina species, indicating highly conservative evolution. The phylogenetic analysis showed that 'Zaohoucheng' had a closer relationship with S. japonica which strongly supported the parental origin of 'Zaohoucheng'.

It had circular mapping organization with the length of 37,638 bp and encoded 66 genes including three rRNAs, 25 tRNAs, 35 known proteins, and three ORFs. Gene arrangement and component were conserved in those reported Saccharina species and cultivars. Only 33 nucleotide substitutions were found according to the total alignment of S. japonica and 'Fujian'. The intergenic region of 19 nucleotides which is located between rps3 and rps9 in S. japonica mitogenome was not detected in 'Fujian'. Phylogenetic analysis showed that 'Fujian' had a closer relationship with S. japonica and cultivar 'Rongfu' which strongly supported its genetic origin and phylogenetic relationship of Chinese Saccharina cultivars.

This mitogenome has a circular mapping organization with the length of 37,657 bp and contains 66 genes, including 35 protein-coding genes, three rRNAs, 25 tRNAs, and three open reading frames (orfs). The overall AT content is 64.73%, showing a higher AT content. The gene content and gene sequence are consistent with those reported varieties and cultivars of Saccharina. Chinese main Saccharina cultivars are analyzed by phylogenetic analysis. It indicates that 'Dongfang No.3' has a close relationship with Saccharina japonica, which strongly supports its genetic origin. The complete mitogenome analysis in this work would help in understanding the genetic background of Chinese Saccharina cultivars.

Further phylogenetic analysis of mitogenomes confirmed that 'Shichang' had closer relationship with cultivar 'Pingbancai', which suggested its parental origin and genetic relationship.

Patterns of Saccharina latissima recruitment.

Project description:The lack of recovery in Norwegian populations of the kelp Saccharina latissima (Linnaeus) C. E. Lane, C. Mayes, Druehl & G. W. Saunders after a large-scale disturbance that occurred sometime between the late 1990s and early 2000s has raised considerable concerns. Kelp forests are areas of high production that serve as habitats for numerous species, and their continued absence may represent the loss of an entire ecosystem. Some S. latissima populations remain as scattered patches within the affected areas, but today, most of the areas are completely devoid of kelp. The question is if natural recolonization by kelp and the reestablishment of the associated ecosystem is possible. Previous studies indicate that a high degree of reproductive synchrony in macrophytes has a positive effect on their potential for dispersal and on the connectivity between populations, but little is known about the patterns of recruitment in Norwegian S. latissima. More is, however, known about the development of fertile tissue (sori) on adult individuals, which is easily observed. The present study investigated the degree of coupling between the appearance of sori and the recruitment on clean artificial substrate beneath adult specimens. The pattern of recruitment was linked to the retreat of visible sori (i.e. spore release) and a seasonal component unrelated to the fertility of the adults. The formation and the retreat of visible sori are processes that seem synchronized along the south coast of Norway, and the link between sori development and recruitment may therefore suggest that the potential for S. latissima dispersal is relatively large. These results support the notion that the production and dispersal of viable spores is unlikely to be the bottleneck preventing recolonization in the south of Norway, but studies over larger temporal and spatial scales are still needed to confirm this hypothesis.
