The complete mitochondrial genome of brown algae: Sargassum fusiforme (Harvey) Setchell.

ABSTRACT: We describe the complete mitochondrial genome sequence of Sargassum fusiforme. This mitogenome is a circular molecule of 34,695 bp in length and had an overall GC content of 37.54%%. Gene annotation showed that 35 protein-coding genes, 2 open reading frames, 25 transfer RNA genes, and 3 ribosomal RNA genes. The phylogenetic tree based on Bayesian shows that S. fusiforme belongs to genus Sargassum, support current taxonomic systems.

Circular mapping revealed that the complete chloroplast DNA sequence of S. fusiforme was 124,298 bp in length and had an overall AT content of 69.57%, including 137 protein-coding genes, 2 open reading frames, 28 transfer RNA genes, and 6 ribosomal RNA genes. The phylogenetic tree based on Bayesian shows that all kinds of Phaeophyceae were clustered into two monophyletic groups.

Optimization and kinetic modeling of ultrasonic-assisted extraction of fucoxanthin from edible brown algae Sargassum fusiforme using green solvents.

Project description:The development of green and sustainable extraction technologies for various naturally active biomaterials is gaining increasing attention due to their environmentally friendly advantages. In this work, the ultrasonic-assisted extraction of fucoxanthin from edible brown algae Sargassum fusiforme using different green solvents was presented. Ethyl lactate, limonene, soybean oil, and sunflower oil were used in place of traditional organic solvents. Ethyl lactate showed similar performance to organic solvents, whereas limonene and vegetable oil exhibited higher selectivity for fucoxanthin. Moreover, the effects of various extraction factors, including liquid/solid ratio, extraction time, extraction temperature, as well as amplitude were studied. The optimal conditions were optimized as follows: liquid/solid ratio, 40 mL/g; extraction time, 27 min; extraction temperature, 75 ℃; amplitude, 53%; and solvent, ethyl lactate. Optimal model of second-order kinetic parameters (rate constant, equilibrium concentration, and initial extraction rate) was successfully developed for describing the dynamic ultrasonic extraction process under different operating conditions.

Characterization of the complete chloroplast genome of Sargassum fusiforme and its phylogenomic position within phaeophyceae.

Project description:Sargassum fusiforme is an important economic seaweed in East Asia. In this study, we characterized the complete chloroplast genome sequence of S. fusiforme using PacBio long-read sequencing technology. It had a circular mapping molecular with the length of 124,286 bp, with a large single-copy region (LSC, 73,437 bp) and a small single copy region (SSC, 40,131 bp) separated by a pair of inverted repeats (IRs, 5,359 bp). The cp genome contained 173 genes including 139 protein-coding, 6 rRNA, and 28 tRNA genes. The phylogenomic analysis indicated that S. fusiforme is closely related to S. thunbergii.

Invasive brown algae (Sargassum spp.) as a potential source of biocontrol against Aedes aegypti.

Project description:Influxes of sargassos are responsible for economic and environmental disasters in areas where they bloom, especially in regions whose main income relies on tourism and with limited capacity for sanitation and public health response. A promising way of valorization would be to convert this incredible biomass into tools to fight the deadly vector mosquito Aedes aegypti. In the present study, we generated hydrolates and aqueous extracts from three main Sargassum morphotypes identified in Guadeloupe (French West Indies): Sargassum natans VIII, Sargassum natans I and Sargassum fluitans. We conducted a chemical characterization and a holistic evaluation of their potential to induce toxic and behavioral effects in Ae. aegypti. Despite the low insecticidal potential observed for all the extracts, we found that S. natans VIII and S. fluitans hydrolates deterred oviposition, induced contact irritancy and stimulated blood feeding behavior in host seeking Ae. aegypti females, while aqueous extracts from S. natans I and S. fluitans deterred both blood feeding behavior and oviposition. Chemical characterization evidenced the presence of phenylpropanoid, polyphenols, amino acids and esters. Thus, Sargassum spp. aqueous extracts and hydrolates could be used to manipulate Ae. aegypti behavior and be valorized as control tools against this mosquito.

Phlorotannins of the Brown Algae Sargassum vulgare from the Mediterranean Sea Coast.

Project description:Brown seaweeds are a good source of bioactive compounds, particularly of phlorotannins, which may exert a wide spectrum of pharmacological properties. In the present study, phlorotannins of S. vulgare were extracted using a 70% acetone solution and the crude extract was further purified through liquid-liquid partition, giving rise to n-hexane, ethyl acetate and aqueous residue fractions. The crude extract and the purified fractions were evaluated for potential antioxidant abilities as well as for inhibitory potential towards the digestive enzymes α-amylase and pancreatic lipase, and anti-inflammatory potential through the hindering of albumin denaturation. Overall, the ethyl acetate fraction was the richest in phlorotannins (9.4 ± 0.03 mg PGE/g) and was also the most promising regarding the tested bioactive properties. Of note, its inhibitory potential towards α-amylase was about nine times that of the commercial drug acarbose and its inhibitory activity against high temperature-induced protein denaturation was superior to that of the non-steroidal drug ketoprofen. According to UHPLC-DAD-ESI-MS/MS analysis, this fraction contained a range of phlorotannins with at least six units of phloroglucinol, including dibenzodioxine-1,3,6,8-tetraol, fuhalol, pentaphlorethol, fucopentaphlorethol and dihydroxypentafuhalol, in addition to several less common phlorotannin sulfate derivatives.
