Sequencing and analysis of the complete mitochondrial genome of Habrobracon hebetor (Hymenoptera: Braconidae).

ABSTRACT: We determined the complete mitochondrial genome sequence of Habrobracon hebetor (Say). The complete mitogenome sequence of H. hebetor was observed to be a circular molecule 15,708 bp long and consists of 13 protein-coding genes (PCG), 2 ribosomal RNA (rRNA) genes, and 22 transfer RNA (tRNA) genes (GenBank accession no. MN842279). This nucleotide composition is biased toward adenine and thymine (85.2% A + T). The A + T-rich region is found between trnM and trnQ, and this entire region was 864 bp long.

Project description:In this study, we sequenced the mitogenome of Habrobracon hebetor, and obtained almost complete mitogenome of it. The mitogenome contains 14,629 bp which consists of 13 protein-coding genes (PCGs), 20 transfer RNA genes (trnI and trnM are absent), and 2 ribosomal RNA genes (GenBank accession no. MT558946). Gene rearrangement events occurred in this species, five tRNA genes with changes in positions or/and directions are found. All of 13 PCGs started with ATN. Eight PCGs used the typical stop codon 'TAA', five PCGs terminated with incomplete stop codons (T). Phylogenetic analyses within the Cyclostomes were performed based on mitochondrial PCGs.

Transcriptome Analysis Reveals the Venom Genes of the Ectoparasitoid Habrobracon hebetor (Hymenoptera: Braconidae).

Project description:The ectoparasitoid Habrobracon hebetor (Hymenoptera: Braconidae) exhibits a broad parasitic capability towards various lepidopteran pests, with venom serving as a crucial virulent factor ensuring successful parasitization and subsequent host mortality. Analyzing the constituents of its venom is essential for elucidating the mechanisms underlying efficient host killing by this parasitoid and for exploring potentially functional venom proteins. Through a transcriptomic analysis, a total of 34 venom proteins were identified within the venom of H. hebetor, encompassing known components such as serine protease, metalloproteinase, esterase, and serine protease inhibitors commonly present in parasitoid venoms. Unique components like paralytic protein and ion transport peptide-like were identified, possibly specific to certain parasitoids, along with novel proteins with uncharacterized functions. Spatial gene expression profiling of the identified venom proteins using transcriptomic data, corroborated by quantitative PCR validation for 13 randomly selected proteins, revealed abundant expression levels in the venom apparatus, affirming them as genuine venom components. Notably, the paralytic protein exhibited prominent expression, with the highest FPKM (fragments per kilobase of transcript per million fragments mapped) value of 24,704.87 in the venom apparatus, indicative of its significant role in successful parasitism by H. hebetor. The identification of these venom proteins establishes a foundation for the further exploration of bioactive agents for pest management strategies.

The complete mitochondrial genome and phylogenetic analysis of Cyanopterus ninghais (Hymenoptera: Braconidae).

Project description:Cyanopterus ninghais is an important gregarious ectoparasitoid during the larval stage of Monochamus alternatus, a key vector for pine wilt disease in Asia. In this study, the complete mitochondrial genome of C. ninghais was sequenced and analyzed. The mitochondrial genome of C. ninghais is 15,386 bp in length, comprising 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes (rRNAs). The nucleotide composition is 41.32% A, 8.29% G, 6.06% C, and 44.33% T. Phylogenetic trees of Braconidae were constructed using 13 PCG sequences via Bayesian inference (BI) and maximum likelihood (ML) analyses to determine their phylogenetic position. Both ML and BI analyses revealed that C. ninghais is closely related to Euurobracon yokahamae, Virgulibracon endoxylaphagus, and Habrobracon hebetor.

Project description:The genome-level features are crucial genetic resources for species identification and phylogenetic analysis. Here, the complete mitochondrial genome of Aphidius colemani Viereck 1912 (Hymenoptera: Braconidae: Aphidiinae) was sequenced, determined and analyzed. The circular genome is 16,372 bp in length with an overall base composition of 38.9% for A, 46.2% for T, 6.7% for C, and 8.2% for G. The mitochondrial genome of A. colemani contained 13 protein-coding genes that initiated by the ATN codon, 22 transfer RNA genes, two ribosomal RNA genes (rRNAs), and a control region (CR). It shared the same gene arrangement patterns that occurred in two tRNA clusters of trnI-trnQ-trnM and trnW-trnC-trnY with Aphidius gifuensis. Phylogenetic analyses using Bayesian inference and Maximum-likelihood methods supported that the two species of Aphidiinae formed a clade and sister to other subfamilies of Braconidae.

Complete mitochondrial genome of a parasitic wasp Microplitis pallidipes (Hymenoptera: Braconidae: Microgastrinae).

Project description:The complete mitochondrial genome of Microplitis pallidipes contains 15931 bp, with an A + T content of 86.5% and consists of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a control region (GenBank accession no. MN892396). All of the 22 tRNA genes displayed an usual clover-leaf structure. Gene rearrangement events occurred in this species, there are eight tRNA genes changed their positions or/and directions. 13 PCGs started with ATN. Ten PCGs used the typical stop codon 'TAA' and 'TAG', three PCGs terminated with incomplete stop codons (TA). Phylogenetic analyses within the microgastroid complex were performed based on mitochondrial protein-coding genes.

Identification of Neuropeptides and Their Receptors in the Ectoparasitoid, Habrobracon hebetor.

Project description:Neuropeptides are a group of signal molecules that regulate many physiological and behavioral processes by binding to corresponding receptors, most of which are G-protein-coupled receptors (GPCRs). Using bioinformatic methods, we screened genomic and transcriptomic data of the ectoparasitoid wasp, Habrobracon hebetor, and annotated 34 neuropeptide candidate precursor genes and 44 neuropeptide receptor candidate genes. The candidate neuropeptide genes were found to encode all known insect neuropeptides except allatotropin, neuropeptide F, pigment dispersing factor, and CCHamides. When compared with the endoparasitic wasp Pteromalus puparum and the ectoparasitic wasp Nasonia vitripennis, trissin and FMRFamide were found only in H. hebetor. A similar result held for the neuropeptide receptor genes, for the receptors were found in H. hebetor except the receptors of CCHamides and neuroparsin. Furthermore, we compared and analyzed the differences in neuropeptides in eight Braconidae wasps and identified natalisin in H. hebetor, Diachasma alloeum, Fopius arisanus and Microplitis demolitor, but not in the other wasps. We also analyzed the transcriptome data and qRT-PCR data from different developmental stages and tissues to reveal the expression patterns of the neuropeptides and their receptors. In this study, we revealed composition of neuropeptides and neuropeptide receptors in H. hebetor, which may contribute to future neurobiological studies.

Habrobracon hebetor and Pteromalus cerealellae as Tools in Post-Harvest Integrated Pest Management.

Project description:Consumers are increasingly demanding pesticide-free grain/legumes and processed foods. Additionally, there are more restrictions, or complete loss, of insecticides labelled for use in managing stored grain insects in post-harvest ecosystems. Suppression of post-harvest pests using parasitic wasps is a more sustainable alternative than chemical pesticides. Habrobracon hebetor (Say) (Hymenoptera: Braconidae) and Pteromalus cerealellae Ashmead (Hymenoptera: Pteromalidae) are two important parasitoids that limit economically important pests of stored products. Host searching ability and reproductive performances of H. hebetor and P. cerealellae depend on a wide range of factors, such as host species, commodities, and environmental conditions. Further, use of entomopathogens can complement the ability of parasitoids to regulate pest populations. This review provides information on aspects of H. hebetor and P. cerealellae biology and successful regulation of post-harvest pest populations.
